Improved Culture Media for Embryogenic Callus Generation in Sorghum [Sorghum bicolor (L.) Moench]

Many attempts on optimization of sorghum[Sorghum bicolor(L.)Moench]tissue culture induction media have been made,but the culture system remains with some bottlenecks compared to that of other *** study aimed at assessing the suitability of various induction media to produce embryogenic callus(yellow and friable)with high induction rates and reduced phenolic *** six culture medium modifications:3 based on Murashige and Skoog(MS)medium and one each based on Chu N6,Gamborg B5 and 190-2 media respectively were applied in the culture of mature embryos from 10 sorghum *** there was a genotype influence on the attainment of a yellow callus,friability of the callus was determined to be dependent on the culture medium and not the *** strength MS medium with 0.2 mg/l 2,4-D with 2.8 g/l Gelrite®as the gelling agent modified with 1.0 g/l KH_(2)PO_(4),1.0 g/l L-proline,1.0 g/l L-asparagine and 0.16 mg/l CuSO_(4)·5H_(2)O(type E)was found to be the most effective resulting in about 60%yellow coloured callus induction with 25%*** of CuSO_(4)·5H_(2)O,KH_(2)PO_(4),L-proline and L-asparagine significantly reduced the phenolic *** strength MS medium was observed to contribute to quality callus production when compared to full strength MS media modified with the *** half strength MS medium was also observed to suppress phenolic *** 190-2 produced the highest regeneration frequency(40%)among the 3-regeneration media *** results provide information on a suitable sorghum callus induction medium necessary for embryogenesis.

Obesity has become a worldwide epidemic,and its prevalence has been projected to grow by 40% in the next decade. This increasing prevalence has implications for the risk of diabetes,cardiovascular disease and also for Chronic Kidney Disease. A high body mass index is one of the strongest risk factors for new-onset Chronic Kidney Disease. In individuals affected by obesity, a compensatory hyperfiltration occurs to meet the heightened metabolic demands of the increased body weight. The increase in intraglomerular pressure can damage the kidneys and raise the risk of developing Chronic Kidney Disease in the long-term. The incidence of obesity-related glomerulopathy has increased ten-fold in recent years. Obesity has also been shown to be a risk factor for nephrolithiasis,and for a number of malignancies including kidney cancer. This year the World Kidney Day promotes education on the harmful consequences of obesity and its association with kidney disease,advocating healthy lifestyle and health policy measures that makes preventive behaviors an affordable option.

Therapeutic proteasome inhibition in experimental acute pancreatitis

AIM： To establish the therapeutic potential of proteasome inhibition, we examined the therapeutic effects of MG132 （Z-Leu-Leu-Leu-aldehyde） in an experimental model of acute pancreatitis. METHODS： Pancreatitis was induced in rats by two hourly intraperitoneal （ip） injections of cholecystokinin octapeptide （CCK; 2×100μg/kg） and the proteasome inhibitor MG132 （10 mg/kg ip） was administered 30 min after the second CCK injection. Animals were sacrificed 4 h after the first injection of CCK. RESULTS： Administering the proteasome inhibitor MG132 （at a dose of 10 mg/kg, ip） 90 min after the onset of pancreatic inflammation induced the expression of cell-protective 72 kDa heat shock protein （HSP72） and decreased DNA-binding of nuclear factor-κB （NF-κB）. Furthermore MG132 treatment resulted in milder inflammatory response and cellular damage, as revealed by improved laboratory and histological parameters of pancreatitis and associated oxidative stress. CONCLUSION： Our findings suggest that proteasome inhibition might be beneficial not only for the prevention, but also for the therapy of acute pancreatitis.

Immunobiology of congenital cytomegalovirus infection of the central nervous system--the murine cytomegalovirus model

Congenital human cytomegalovirus infection is a leading infectious cause of long-term neurodevelopmental sequelae, including mental retardation and hearing defects. Strict species specificity of cytomegaloviruses has restricted the scope of studies of cytomegalovirus infection in animal models. To investigate the pathogenesis of congenital human cytomegalovirus infection, we developed a mouse cytomegalovirus model that recapitulates the major characteristics of central nervous system infection in human infants, including the route of neuroinvasion and neuropathological findings. Following intraperitoneal inoculation of newborn animals with mouse cytomegalovirus, the virus disseminates to the central nervous system during high-level viremia and replicates in the brain parenchyma, resulting in a focal but widespread, non-necrotizing encephalitis. Central nervous system infection is coupled with the recruitment of resident and peripheral immune cells as well as the expression of a large number of pro-inflammatory cytokines. Although infiltration of cellular constituents of the innate immune response characterizes the early immune response in the central nervous system, resolution of productive infection requires virus-specific CD8＋ T cells. Perinatal mouse cytomegalovirus infection results in profoundly altered postnatal development of the mouse central nervous system and long-term motor and sensory disabilities. Based on an enhanced understanding of the pathogenesis of this infection, prospects for novel intervention strategies aimed to improve the outcome of congenital human cytomegalovirus infection are proposed.

Clinical significance of granuloma in Crohn's disease

AIM: Granuloma is considered the hallmark of microscopic diagnosis in Crohn's disease (CD), but granulomas can be detected in only 21-60% of CD patients. The aim of this study was to evaluate the frequency of granulomas by multiple endoscopic biopsies in patients with CD and to examine whether group of patients with or without granuloma exhibit a different clinical course. METHODS: Fifty-six patients with newly diagnosed CD were included in the study. Jejunoscopy, enteroclysis and ileo-colonoscopy were performed in all patients. At least two biopsy specimens from each examined gastrointestinal segment were examined microscopically searching granuloma. The clinical course was followed in all patients, and extraintestinal manifestations as well as details of any immunosuppressive therapy and surgical intervention were noted. RESULTS: Granuloma was found in 44.6% of the cases (25 patients). Patients with granuloma had higher activity parameters at the time of the biopsies. Extraintestinal manifestations were observed and surgical interventions were performed more often in the granuloma group. The need of immunosuppressive therapy was significantly more frequent in the patients with granuloma. Granuloma formation is mote often seen in younger patients, and mainly in the severe, active penetrating disease. CONCLUSION: The significantly higher frequency of surgical interventions and immunosuppressive therapy suggests that granuloma formation is associated with a more severe disease course during the first years of CD.

Sorghum[Sorghum bicolor(L.)Moench]can benefit from accelerated breeding and release of improved varieties through doubled haploid *** technology has been used in speeding up the breeding of other major cereals such as wheat,maize and rice,for which generally widely applied optimised protocols exist.A reproducible protocol for the crop,that can overcome genotype dependency and other species-specific challenges such as phenolic exudation is however *** study aimed at sorghum doubled haploids production thereby contributing to the development of an improved *** the 28 hybrid genotypes,both F1 registered-and experimental hybrids involved,this study successfully produced haploids from five genotypes and subsequently,four confirmed doubled-haploid lines on W14mf medium or its modification with 1.0 gl^(-1)L-proline,1.0 gl^(-1)L-asparagine and 1.0 gl^(-1)KH_(2)PO_(4).Medium 190-2Cu was used for regeneration and rooting,which occurred successfully,if the calli were transferred on to it less than 7 days after induction,and temperature was maintained at 25°C under light *** dependency was not wholly overcome;however,sorghum’s high tillering ability and abiotic stress tolerance were observed to contribute to attainment of haploid *** diploids producing seeds at rates of upto 80.5%were obtained,therefore eliminating the need for colchicine duplication.

Brain is one of the most temperature sensitive *** the fundamental role of temperature in cellular metabolism,thermal response of neuronal populations is also significant during the evolution of various neurodegenerative *** such critical environmental factor,thorough mapping of cellular response to variations in temperature is desired in the living *** far,limited efforts have been made to create complex devices that are able to modulate temperature,and concurrently record multiple features of the stimulated *** our work,the in vivo application of a multimodal photonic neural probe is ***,thermal,and electrophysiological functions are monolithically integrated in a single *** system facilitates spatial and temporal control of temperature distribution at high precision in the deep brain tissue through an embedded infrared waveguide,while it provides recording of the artefact-free electrical response of individual cells at multiple locations along the probe *** distribution of the optically induced temperature changes is evaluated through in vitro measurements and a validated multi-physical *** operation of the multimodal microdevice is demonstrated in the rat neocortex and in the hippocampus to increase or suppress firing rate of stimulated neurons in a reversible manner using continuous wave infrared light(λ=1550 nm).Our approach is envisioned to be a promising candidate as an advanced experimental toolset to reveal thermally evoked responses in the deep neural tissue.

Dear Editor,The Arabidopsis CONSTITUTIVELY PHOTOMORPHOGENIC1/ SUPPRESSOR OF PHYA-105 （COP1/SPA） complex is a key repressor of light signaling that inhibits light responses in dark- ness. It acts as an E3 ubiquitin ligase, which ubiquitinates posi- tively acting light-signaling intermediates, mainly transcription factors, thereby targeting them for proteolytic degradation by the 26S proteasome. In the light, photoreceptors directly interact with the COP1/SPA complex, leading to its inactivation, which subsequently allows the target transcription factors to accumu- late and initiate vast reprogramming of gene expression （Huang et al., 2014）.

A nuclear import inhibitory peptide ameliorates the severity of cholecystokinin-induced acute pancreatitis

AIM: To assess the effect of our novel cell-permeable nuclear factor-kappaB (NF-κB) inhibitor peptide PN50 in an experimental model of acute pancreatitis. PN50 was produced by conjugating the cell-penetrating penetratin peptide with the nuclear localization signal of the NF-κB p50 ***: Pancreatitis was induced in male Wistar rats by administering 2×100 μg/kg body weight of cholecystokininoctapeptide (CCK) intraperitoneally (IP) at an interval of 1 h. PN50-treated animals received 1 mg/kg of PN50 IP 30 min before or after the CCK injections. The animals were sacrificed 4 h after the first injection of ***: All the examined laboratory (the pancreatic weight/body weight ratio, serum amylase activity,pancreatic levels of TNF-α and IL-6, degree of lipid peroxidation, reduced glutathione levels, NF-κB binding activity, pancreatic and lung myeloperoxidase activity) and morphological parameters of the disease were improved before and after treatment with the PN50 *** to the histological findings, PN50 protected the animals against acute pancreatitis by favoring the induction of apoptotic, as opposed to necrotic acinar cell death associated with severe acute ***: Our study implies that reversible inhibitors of stress-responsive transcription factors like NF-κB might be clinically useful for the suppression of the severity of acute pancreatitis.
