Acute bacterial infection negatively impacts cancer specific survival of colorectal cancer patients

AIM:To assess the impact of bacterial infections on cancer-specific survival in patients with colorectal cancer. METHODS:This was a retrospective cohort study of colorectal cancer patients treated at the A.C. Camargo Cancer Center between January 2006 and April 2010. The presence of bacterial infection during cancer treatment,or up to one year after,was confirmed by laboratory tests or by the physician. Infections of the urinary,respiratory or digestive tracts,bloodstream,skin or surgical site were defined by testing within a single laboratory. Criteria for exclusion from the study were:chronically immunosuppressed patients;transplant patients(due to chronic immunosuppression);human immunodeficiency virus carriers;chronic use of corticosteroids or other immunosuppressive drugs;patients with autoimmune disease or primary immunodeficiency;known viral or parasitic infections. Patients with infections that did not require hospitalization were not included in the study because of the difficulty of collecting and tracking data related to infectious processes. In addition,patients hospitalized for pulmonary thromboembolism,stroke,acute myocardial infarction,uncontrolled diabetes,malignant hypercalcemia or other serious non-infectious complications not directly related to infection were also excluded. Survival curves were plotted using the Kaplan-Meier method,and logrank tests(univariate analysis) and a Cox test assuming a proportional hazards model(multivariate analysis) were performed to examine associations between clinical history and characteristics of infection with cancer-specific survival. RESULTS:One-hundred and six patients with colorectal cancer were divided into two groups based on the presence or absence of bacterial infection. Patient ages ranged from 23 to 91 years,with a median of 55 years. The majority of patients were male(57/106,53.77%) with stage Ⅲ colorectal cancer(45/106,44.11%). The effect of rippling on the mechanical properties of graphene

Graphene is the stiffest material known so far but, due to its one-atom thickness, it is also very ***, free-standing graphene exhibit ripples that has major effects on its elastic properties. Here we will summarize three experiments where the influence of rippling is essential to address the results. firstly, we observed that atomic vacancies lessen the negative thermal expansion coefficient(TEC) of free-standing *** also observed an increase of the Young's modulus with global applied strain and with the introduction of small density defects that we attributed to the decrease of rippling. Here, we will focus on a surprising feature observed in the data: the experiments consistently indicate that only the rippling with wavelengths between 5 and 10 nm influences the mechanics of graphene. The rippling responsible of the negative TEC and anomalous elasticity is thought to be dynamic, i.e. flexural phonons. However, flexural phonons with these wavelengths should have minor effects on the mechanics of graphene, therefore other mechanisms must be considered to address our observations. We propose static ripples as one of the key elements to correctly understand the thermomechanics of graphene and suggest that rippling arises naturally due to a competition of symmetry breaking and anharmonic fluctuations.

Decreased Gait and function in Duchenne Muscular Dystrophy

Duchenne muscular dystrophy (DMD) is a genetic disorder linked to chromosome Xp21, due to absence of dystrophin production. It is clinically characterized by progressive muscle weakness, fatigue, and development of joint contractures that compromise general motor functionality, mainly the gait. Objective: To characterize the motor function and decrease gait in children with DMD using the Portuguese version of the Motor function Measure scale (MfM-P). Methods: A review of medical records including chronological age and scores from MfM-P of children with a DMD who attended at the Neuromuscular Diseases Clinic at Campinas State University (UNICAMP), Brazil was performed in this study. A total of 36 medical records of male patients with confirmed clinical diagnosis of DMD, ambulatory or not, regardless of age;excluding those with other associated diseases or other types of muscular dystrophies were selected. Data were analyzed using Kolmogorov-Smirnov and Spearman correlation statistical tests. Results: Analysis of all data collected showed that 75% of our sample had D1 scores lower than 41.02%. There was a linear relationship between the scores of D2 and D3, but no association between D2 and D1 scores was noted. D1 score was between 40% and 80% in those patients presenting D2 scores between 80% and 100%. In all cases patients with low total score presented a greater risk for loss of gait and their functionality. Conclusion: The standing posture and the postural transfers were the worst activities observed in children with DMD, with positive correlation between proximal and distal motor function. Even with high scores according MfM-P in proximal function, the children showed strong predictors for loss of gait.

Benthic diatom community response to metal contamination from an abandoned Cu mine:Case study of the Gromolo Torrent(Italy)

Environmental contamination has become a global problem of increasing intensity due to the exponential growth of industrialization. One main debated issue is the metal contamination of rivers receiving Acid Mine Drainage(AMD) from active/abandoned mines. In order to assess the quality of lotic systems, diatoms are commonly used, as their assemblage modifies on the basis of changes in environmental parameters. Benthic diatom changes were analyzed along the metal-impacted Gromolo Torrent(Liguria, NW Italy) with the aim of understanding the effects of input from the abandoned Libiola Cu mine. The results support the hypothesis that metals from AMD lead to massive changes in diatoms, resulting in low biological diversity and in a shift of dominance, passing from the genera Cymbella and Cocconeis to more tolerant and opportunistic species, such as Achnanthidium minutissimum and fragilaria rumpens. The high concentrations of labile metals, measured through Diffusion Gradients in Thin-films(DGT) immediately downstream of the two AMD inputs in the torrent, corresponded to a sudden decrease in the presence of diatoms, indicating the possible reaching of acute toxic levels. In particular,A. minutissimum dominated the mining area and was positively correlated with Cu and Zn;whereas f. rumpens bloomed downstream of this area, where the metal content was diluted,and was positively correlated with As and Pb. finally, an important abundance of Nitzschia palea and teratological forms of A. minutissimum and f. rumpens were observed downstream from the mine, indicating that metals may have an important impact on diatoms up to the torrent mouth.

Effect of a Combination of Berberine and Monacolin on CYP Enzymes： An Oral Cocktail Interaction Study in Healthy Volunteers

Objectives： This study was aimed at investigating the potential of the herbal exctracts, berberine and monacolin, administered in combination at the recommended dose, to generate clinically relevant inhibition of the CYP （cytochromes P450） enzyme system after a single oral administration in human healthy volunteers. Methods： Twelve healthy male volunteers received a five-probe drug cocktail alone （reference） or with the combination of berberine and monacolin （test）, in a randomized, open label, crossover fashion. Plasma concentrations of cocktail components, caffeine （CYP1A2）, metoprolol （CYP2D6）, omeprazole （CYP2C 19）, midazolam （CYP3A4）, warfarin （CYP2C9）, and plasma concentrations of berberine, monacolin and its metabolite were measured by LC-MS/MS. Pharmacokinetic parameters were determined by non-compartmental analysis. Lack of inhibition was assumed if the 90% CI （confidence interval） for estimated ratio test/reference was included in the acceptance limits 0.70-1.43 for phenotyping metrics AUClast and Cmax. Key findings： All test/reference ratios were close to unity and CIs （confidence intervals） were within the acceptance limits, except for the upper value of omeprazole Cmax. Given the high intraindividual variability, this finding was considered clinically irrelevant. Conclusions： Clinically relevant inhibition of the activities of the CYP isoenzymes investigated can be excluded when berberine and monacolin are administered in combination at the recommended dose.

Objectives： This study aimed to evaluate the in vitro antioxidant capacity, to determine the anti-inflammatory effect due to lipoxygenase inhibition and to test the antimicrobial activity of ethanolic extracts from leaves of seven climbing species belonging to the Bignoniaceae family. Moore） L. G. Lohmann, fridericia chica （Bonpl.） L. G. Lohmann and Tanaecium selloi （Spreng.） L. G. Lohmann. Methods： The antioxidant activity was evaluated using three methods, 2,2＇-azino-bis（3-ethylbenzothiazo line-6-sulfonic acid）（ABTS）, 2,2-diphenyl-l-picrylhydrazyl （DPPH）, and ferric reducing antioxidant power. Lipoxygenase-inhibiting activity was assayed spectrophotometrically; the result was expressed as percent inhibition. The antimicrobial activity was assessed using the agar disk diffusion method. Minimal inhibitory concentration （MIC） and minimal bactericidal]fungicidal concentration were also determined for each extract against 12 pathogenic bacterial strains of Staphylococcus aureus and seven fungal strains of the Candida genus. The identification of the major compounds present in the most promising extract was established by high-performance liquid chromatography-tandem mass spectrometry. Results： C convoluta, f. caudigera, and f. chica exhibited the best antioxidant activity by scavenging DPPH and ABTS~ radicals and reducing fe3＋ ion. These extracts showed a notable inhibition of lipnxygenase. f. caudigera was found to have the lower MIC value against S. aureus strains and six Candida species. The extracts of f. caudigera and C. convoluta were active even against methicillin-resistant S. aureus. C convoluta had higher total phenol content, better antioxidant activity and superior anti- inflammatory and antimicrobial activity. The main phenolic compounds found in this extract

Effect of food on the Oral Bioavailability of Berberine and Monacolin Administered in Combination in Healthy Male Volunteers

Objectives： To investigate the effect of food on the bioavailability of a combination of monacolin and berberine in healthy volunteers. Methods： Eighteen male volunteers received a single recommended oral dose of the combination under fasted conditions （reference） and fed conditions （high fat meal; test）, in a randomized, open label, crossover fashion. Plasma concentrations of berberine, monacolin and its metabolite were measured by LC-MS/MS. Pharmacokinetic parameters were determined by non-compartmental analysis. No effect of food was assumed if the 90% CIs （confidence intervals） for estimated ratio test/reference was included in the acceptance limits 0.80-1.25 for phenotyping metrics AUCt and Cmax- Key findings： for berberine, the Cmax and AUCt test/reference ratios were 2.97 and 2.69, respectively, and relevant 90% CIs （2.25-3.91 and 2.15-3.36, respectively） were above the acceptance limit. for lovastatin hydroxy acid, the active metabolite of monacolin, the test/reference ratios were 1.18 （Cmax） and 0.98 （ALJCt）. The 90% CIs fell entirely within the acceptance limit for AUCt, （0.85-1.13）, whereas the upper bound of the 90% CIs for Cmax （1.01-1.37） was just above the predefined interval. Conclusions： food intake significantly increases berberine bioavailability and does not significantly affect monacolin bioavailability when these two extracts are administered in combination.

TLR2 and TLR4 polymorphisms influence m RNA and protein expression in colorectal cancer

AIM: To evaluate the effect of promoter region polymorphisms of toll-like receptor(TLR)2-196 to-174 del and TLR4-1607T/C(rs10759932) on m RNA and protein expression in tumor tissue and of TLR4+896A/G(rs4986790) on colorectal cancer(CRC) ***: The TLR2-196 to-174 del polymorphism was investigated using allele-specific polymerase chain reaction(PCR) and the TLR4-1607T/C and TLR4+896A/G by PCR-restriction fragment length p o l y m o r p h i s m( R f L P). W e g e n o t y p e d 4 3 4 D N A samples from 194 CRC patients and 240 healthy individuals. The m RNA relative quantification(RQ) was performed in 40 tumor tissue samples by quantitative PCR Taq Man assay, using specific probes for TLR2 and TLR4 genes, and ACTB and GAPDH reference geneswere used as endogenous controls. Protein expression was analyzed by immunohistochemistry with specific primary ***: No association was found for TLR4-1607T/C and TLR4+896A/G by three statistical models(logadditive, dominant and recessive). However, based on dominant and log-additive models, the polymorphic variant TLR2-196 to-174 del was associated with increased CRC risk [dominant: odds ratio(OR) = 1.72, 95%CI: 1.03-2.89; P = 0.038 and log-additive: OR =1.59, 95%CI: 1.02-2.48; P = 0.039]. TLR2 m RNA expression was increased in tumor tissue(RQ = 2.36) when compared to adjacent normal tissue(RQ = 1; P f TLR2 and TLR4 protein expression was concordant with the findings of m RNA expression. In addition, the TLR2-196 to-174 del variant carriers showed m RNA relative expression 2.19 times higher than wild-genotype carriers. The TLR2 protein expression was also higher for the TLR2-196 to-174 del variant carriers [117 ± 10 arbitrary unit(a.u.) vs 95 ± 4 a.u., P = 0.03]. However, for the TLR4-1607T/C polymorphism no significant difference was found for both m RNA(P = 0.56) and protein expression(P
