Background:Animal models are widely used in scientific research in order to obtain information from a whole organism under a specific set of experimental *** lineages of mice have been used to investigate diseases and new therapeutic strategies,and,consequently,hematological and biochemical tests in these laboratory animals are essential to validate scientific *** study seeks to establish reference values for hematological and biochemical parameters of four lineages of ***:We evaluated the hematological and biochemical profiles of 20 males and 20 females from the lineages Swiss(heterogeneous),BALB/c and C57BL/6(isogenic),and B6D2F1(hybrid),totaling 160 *** were standardized using the systems pocH-100iV Diff™for 19 hematological parameters and VITROS®350 for 12 biochemical ***:Results are shown as means and standard deviation,grouped by lineage and *** the values obtained in this study with the values from previous studies,some variations were detected,which could be explained by differences in methodologies or individual ***:Thus our study shows that knowledge and disclosure of the values of physiological parameters of laboratory animals is necessary,and emphasises the importance of considering variations influenced by gender,lineage and genotype in the choice of the best experimental model.

Dental caries, the disease that causes tooth decay, is infectious, and the mutans streptococci bacteria have long been identified as the primary disease-causing agents. Caries vaccines showed promising results in experimental studies;however, it remains far the effective use in humans due to political-economic and ethical issues. Progress towards practical vaccine development requires evaluation of candidate vaccines in clinical trials. Promising strategies of passive immunization also require further clinical evaluation. The purpose of this chapter is to review the literature on the main research projects aimed at developing caries vaccines.

In Silico Approach for the Identification of Potential Targets and Specific Antimicrobials for Streptococcus mutans

Tooth decay affects most of the population in developed countries. The multifactorial etiology of the disease includes multiple bacterial species, S. mutans is the main pathogen associated with the disease. This bacterium adheres to the tooth surface and allows the colonization of other microorganisms resulting in dental biofilm. Several therapeutic agents are available to treat or prevent tooth decay, but none, with the exception of fluoride, has significantly influenced the disease’s global burden. Moreover, the probable development of resistance of microorganisms to existing antibacterial agents and the scarcity of good antimicrobial agents motivates this effort for innovation. The detailed knowledge obtained in recent years on the S. mutans allowed the identification of potential targets in this microorganism, enabling the development of specific drugs to combat tooth decay. Thus, the identification of potential targets in these pathogens is the first step in the discovery process of new therapeutic agents. Currently, the experimental assays used for this purpose are expensive and time consuming. In contrast, bioinformatics methods to predict drug targets are cheap, quick and workaday in the biotechnology. This article will review the potential drug targets in S. mutans, as well as the bioinformatics methods used to identify these targets and effective drugs for specific pharmacological treatment of dental caries.

Chlorhexidine (CHX) is regarded as one of the most successful antiplaque agents in controlling the formation of dental biofilm. Nevertheless, molecular mechanisms of their effects in Streptococcus mutans are largely unknown. In this work, the effects of sub-lethal and lethal concentrations of chlorhexidine (CHX) on planktonic or biofilm-organized Streptococcus mutans cells were investigated in dose- and time-dependent manner. The Minimum Inhibitory Concentration (MIC) and the Minimum Bactericidal Concentration (MBC) for planktonic cells and biofilm conditions were determined by standard methods. Quantitative PCR (qPCR) was used to quantify the relative levels of glucosyltransferase B (gtfB), gtfC and gtfD transcription of S. mutans in the presence of CHX. The CHX activity in the initial biofilm structure and morphological alterations in planktonic cells were examined by Scanning Electron Microscopy (SEM). The results indicate that CHX increased expression of gtfC and gtfD in planktonic S. mutans cells and CHX reduced the expression of gtfB, gtfC, and gtfD in biofilms. High concentrations of CHX resulted in several wilted S. mutans planktonic cells with spilled intracellular material, while decreased cells’ chain length and matrix was found when the initial biofilm was exposed to increasing concentrations of CHX. CHX’s effects against bacteria depend on the type of growth organization and the concentration and time of exposure to the drug. At sub-lethal concentrations, CHX affects the expression of glucosyltransferases, which may have anticariogenic effect.

Occasional Tillage in a Field Established under Conservation Agriculture for Tomato Cropping

The Conservation Agriculture (CA) is a current concept drives to save natural resources for agricultural production based on the minimum soil disturbance or no-tillage, crop rotation and permanent maintenance of straw on soil surface. The increasing in soil density is a problem to achieve great copping yield under CA, so occasional one-time tillage is considered as an alternative to continuous no-tillage. In this way, this experiment was carried out to compare occasional tillage and no-tillage interacting with cover crops in a field established under conservation agriculture. Thus, the experimental treatments were set up by two tillage methods, conventional tillage and no-tillage and two cover crops, white lupin and millet setting in a randomized blocks with split plot design with four replications. The traits evaluated in the research were soil fertility, soil resistance to penetration, soil moisture and tomato agronomic performance. No-tillage was more efficient to preserve soil moisture;however soil fertility, soil resistance to penetration and tomato yield were favored by conventional tillage. Regarding to cover crops white lupin increased the soil K concentration and enhanced the tomato growth. Although occasional tillage had better performance to the soil fertility and tomato yield, we highlighted that CA is the better way to increase soil health and soil and water conservation along the time leading to so desired regenerative agriculture.

Atypical Presentation of Crohn’s Disease: Diagnostic Challenges in a Clinical Case

Introduction: Crohn’s Disease (CD) is a chronic inflammatory disorder with a heterogeneous presentation. While diarrhea, abdominal pain, and weight loss are hallmarks, atypical manifestations can obscure the diagnosis. This report highlights an unusual presentation of CD to emphasize the need for comprehensive diagnostic strategies. Case Report: A 25-year-old male presented with peripheral edema, anorexia, and abdominal distension but lacked classic gastrointestinal (GI) symptoms. Laboratory findings included microcytic anemia and hypoalbuminemia, while imaging revealed ascites and bowel wall thickening. Elevated fecal calprotectin and positive Anti-Saccharomyces cerevisiae antibodies (ASCA) supported the diagnosis. Endoscopy confirmed ileocolic Crohn’s Disease (L3 + L4). Infliximab therapy resulted in marked clinical improvement. Discussion: This case underscores the complexity of atypical CD presentations. Early use of serological markers, imaging, and endoscopy guided the diagnosis. Recognition of CD’s diverse manifestations is critical for timely intervention. Conclusion: Atypical CD presentations require heightened clinical suspicion and a multidisciplinary approach to reduce diagnostic delays and improve patient outcomes.

Oregano and Rosemary Essential Oils in Collagen Fiber and Polyvinyl Alcohol Films: Application as Biodegradable Active Packaging for Ready-to-Eat Meat Products

Developing packaging using biomaterials offers a solution to the problem of solid plastic waste in urban areas. However, selecting suitable materials remains challenging, particularly when the packaging is intended for food products with high moisture content, such as meat. In response to this challenge, a novel film comprising a blend of collagen fiber and polyvinyl alcohol, supplemented with varying concentrations of oregano and rosemary essential oils, was developed. The mechanical properties, in vitro antioxidant activity, and chemical composition of these collagen fiber and polyvinyl alcohol films, enhanced with oregano and rosemary essential oils, were thoroughly characterized. The compounds γ-terpinene and ρ-cymene played a significant role in the antioxidant activity of the packaging. The impact of this active packaging on the quality of sliced mortadella was evaluated over 0, 3, 6, and 9 days of refrigeration. Time significantly influenced most of the variables examined. The concentration of essential oils (0%, 2%, 4%, and 6%) significantly affected the moisture level, water activity, pH, and Escherichia coli count in the packaged product. For lipid oxidation analysis, the type of essential oil proved significant, with oregano essential oil demonstrating superior activity. A pro-oxidant effect was noted in products with 6% rosemary essential oil. Packages containing essential oils from oregano and rosemary exhibited lower E. coli counts. Films enriched with essential oils showed a higher rate of biodegradation.

BACKGROUND Pediatric living-donor liver transplantation is considered a safe alternative for the treatment of children with end-stage liver *** tertiary centers and specialized medical staff are necessary to ensure compatible long-term survival rates and quality-of-life for these *** To report the results and the 10-year learning curve of a pediatric living-donor liver transplantation *** We conducted a retrospective cohort study of pediatric recipients from 2013 to ***-transplant outcomes and patient survival rates were compared between two 5-year periods of the *** A total of 25 and 48 patients underwent transplantation in the first(2013-2017)and second period(2018-2023),*** vein and hepatic artery thrombosis occurred in 11(15.1%)and seven(9.6%)patients,*** complications were observed in 39 of 73 patients(53.4%).A lower warm ischemia time was observed in the second period compared to the first(32.6±8.6 minutes vs 38.4±9.8 minutes,P=0.018,respectively).Patient survival rates at 1 and 5 years were 84%in the first period and 91.7%in the second period,with no significant difference(P=0.32).CONCLUSION The reported indications and outcomes align with the current *** findings provide crucial evidence regarding the feasibility of establishing a living donor program with consistent results over time.

Serum levels of angiotensin converting enzyme as a biomarker of liver fibrosis

The renin angiotensin system(RAS) is classically conceived as a circulating hormonal system involved in blood pressure control and hydroelectrolyte balance. The discovery that RAS components are locally expressed in a wide range of organs and tissues,including the liver,pointed to a role for this system in the pathogenesis of several conditions including hepatic fibrosis and cirrhosis. It has been widely reported that the classical RAS axis composed by the angiotensin converting enzyme(ACE)-angiotensin(Ang) Ⅱ-Ang type 1(AT1) receptor mediates pro-inflammatory,pro-thrombotic,and pro-fibrotic processes. On the other hand,the alternative axis comprising ACE2-Ang-(1-7)-Mas receptor seems to play a protective role by frequently opposing Ang Ⅱ action. Chronic hepatitis B(CHB) is one of the leading causes of liver fibrosis,accounting for the death of nearly one million people worldwide. Liver fibrosis is a key factor to determine therapeutic interventions for patients with CHB. However,the establishment of non-invasive and accurate methods to detect reversible stages of liver fibrosis is still a challenge. In an elegant study published in the 36 th issue of the World Journal of Gastroenterology,Noguchi et al showed the predictive value of serum ACE levels in detecting not only advanced stages of liver fibrosis but also initial and intermediate fibrotic stages. The serum levels of ACE might represent an accurate,non-invasive,widely available,and easy method to evaluate fibrosis related to CHB. Moreover,therapies involving the inhibition of the classical RAS axis components might be promising in the control of CHB-related liver fibrosis.
