AIM To establish a permanent piwi like RNA-mediated genesilencing 1(PIWIL1) gene knockout in AGP01 gastric cancer cell line using CRISPR-Cas9 system and analyze phenotypic modifications as well as gene expression *** CRISPR-Cas9 system used was purchased from Dharmacon GE Life Sciences(Lafayette, CO, United States) and permanent knockout was performed according to manufacturer's recommendations. Woundhealing assay was performed to investigate the effect of PIWIL1 knockout on migration capability of cells and Boyden chamber invasion assay was performed to investigate the effect on invasion capability. For the gene expression analysis, a one-color microarray-based gene expression analysis kit(Agilent Technologies, Santa Clara, CA, United States) was used according to the protocol provided by the manufacturer. RESULTS PIWIL1 gene knockout caused a significant decrease in AGP01 migration capacity as well as a significant decrease in cell invasiveness. Moreover, functional analysis based on grouping of all differentially expressed m RNAs identified a total of 35 genes(5 up-regulated and 30 down-regulated) encoding proteins involved in cellular invasion and migration. According to current literature, 9 of these 35 genes(DOCK2, ZNF503, PDE4 D, ABL1, ABL2, LPAR1, SMAD2, WASF3 and DACH1) are possibly related to the mechanisms used by PIWIL1 to promote carcinogenic effects related to migration and invasion, since their functions are consistent with the changes observed(being up-or down-regulated after knockout). CONCLUSION Taken together, these data reinforce the idea that PIWIL1 plays a crucial role in the signaling pathway of gastric cancer, regulating several genes involved in migration and invasion processes; therefore, its use as a therapeutic target may generate promising results in the treatment of gastric cancer.

The intertidal zone is a complex coastal area exposed to a wide variety of environmental factors. During low tide, rocky pools are exposed to air for several hours throughout the day, and environmental conditions such as temperature and salinity are highly variable. Fish communities that inhabit these tide pools are also exposed to environmental variability;therefore it is important to look at fish community structure in relation to important environmental factors such as temperature, salinity and dissolved oxygen on tide pools during spring tides, time of greater exposure to air. Therefore, the aim of the present study was to analyze the variation of the taxonomic distinctness of intertidal fish communities considering temperature, salinity and dissolved oxygen of the water that remains trapped in the tide pools during low tide. Visual census surveys were carried out monthly, from January to December 2015, in the intertidal zone of El Faro during spring tides on full moon days, when fluctuations of environmental variables are significant. Total extension of the census surveys was 156 × 5 m (780 m2). A total of 145 tide pools were sampled and divided in three groups (small, medium and large) according to size and depths that ranged from 20 to 45 cm. Temperature, salinity and dissolved oxygen fluctuate throughout the year of study. Thermal difference between open ocean and tide pools ranged from 3.5°C to 4.5°C. A total of 3757 organisms from 22 species, 12 families, four orders and one class were recorded. When comparing taxonomic distinctness and average taxonomic distinctness between months and tide pools of different sizes, values fell within the confidence intervals in the tunnel and were located close to the mean.

The silver oxide nanoparticles （AgONps） used in this study was obtained by laser ablation technique. On the other hand, graphene bilayers were obtained through micro mechanic exfoliation. Both techniques are considered clean, simple and they have economic viability. The analyses have showed a positive charge in nanoparticles and alteration in Raman spectra caused by doping in the bilayer observing the appearing of D band. The alterations in the Raman spectrums are related with the introduced defects in the crystalline chain after the nanoparticles incorporation on bilayers. However, we have been doing a systematic study about the Raman laser output in only graphene bilayer and in graphene bilayer doped with nanoparticles. These measurements have shown a clear difference to both cases. The distance between defects has been showing to be inversely proportional to the ratio ID/I6 in the analyzed cases, increasing （up power） or lowering （down power） the incident laser output, with this it was possible to handle the defect distances in the case of AgO nanoparticles/graphene bilayers. Finally, we analyzed the ratio of the integrated area of D and G band, from these parameters are determined the defect density （ND） and the defects density （LD）. Thus, we clearly find a tuning between the ND and LD as a function of laser power density Raman using to λ = 532 nm.

There has been an increase of community studies that incorporate the use of functional diversity indices. The incorporation of these indices in the analysis of marine communities is recent, however, could contribute with relevant information about the health of those communities. The lagoon of La Paz is a body of water located in La Paz, B.C.S., México. Despite its regional importance, there are not recent studies about its fish communities. Therefore, the aim of the present study was to analyze the temporal and spatial structure of the functional diversity of fish communities associated to soft bottoms, from August 2016 to July 2017, as well as important environmental variables. Functional diversity indices showed significant differences between months. Spatial analysis did not show significant differences perhaps due to low variability of the type of substrate. Temperature showed temporal differences as well. Therefore, temperature may be a limiting environmental variable for many fish species inhabiting coastal lagoons.

Introduction: Breast cancer is a common disease diagnosed in Mexican women and the first leading cause of death [1]. Heterogeneity in patients’ response to treatment is consistently observed across populations. Glutathione S-transferases (GSTs) are involved in the metabolism of environmental carcinogens, reactive oxygen species and chemotherapeutic agents by catalyzing the glutathione with electrophilic compounds. The deletion of GSTT1 and GSTM1 genes result in loss of enzyme activity. A few studies evaluated the response to treatment and the polymorphisms of GSTT1 and GSTM1. The aim of this work is to make the association of the null polymorphisms of GSTT1 and GSTM1 with the response to chemotherapy basically doxorubicin and cyclophosphamide. Methods: The genotyping of thirty patients with breast cancer was made with the Polymerase chain reaction, to identify the polymorphisms of GSTT1 and GSTM1. We determine the status of Her-2 neu, estrogen and progesterone receptors, then the response to treatment was made with an ultrasound and pathological data. We made the association with the χi2 statistics using a p≤0.05. Results: Using the Sigma Stat 3.5 program and the chi-squared analysis, we do not observe a significant association with the GSTT1+/GSTM1+, GSTT1-/GSTM1+ and GSTT1-/GSTM1-polymorphisms and the better or worse response to cyclophosphamide and doxorubicin. With the Her-2 neu, estrogen and progesterone receptors status, we neither found an association with the response to the therapy. Conclusion: This study suggests that GSTT1 and GSTM1 polymorphisms have no statistical significance between the genotype of women with advanced breast cancer and the response to neoadjuvant chemotherapy, but we can see a clear tendency toward better response with the null genotype.
