Accurate es.imates.of d.A quantity are lik.ly to become increas.ngly important for s.cces.ful genomic s.reening of ins.ct populations.via recently d.veloped. highly multiplexed.genotyping as.ays.and.high-throughput s.quencing method.0} Here we s.ow that genomic d.A extractions.from s.ngle Anopheles.gambiae Giles.us.ng a s.and.rd.commercial k.t-bas.d.method.logy yield.extracts.with concentrations.below the linear range of s.ectrophotometric abs.rbance at 260 nm. Concentrations.d.termined.by s.ectropho- tometry were not reprod.cible, and.are therefore neither accurate nor reliable. However, d.A quantification us.ng a fluores.ent nucleic acid.s.ain （PicoGreen） gave highly reprod.cible concentration es.imates. and.ind.cated.that, on average, s.ngle mos.uitoes.yield.d.approximately 300 ng of d.A. s.ch a total yield.is.currently ins.fficient for many highthroughput genome s.reening applications. neces.itating whole genome amplification of all or mos. ind.vid.als.in a population prior to genotyping.
