The first seedling or a.l-sta.e resista.ce （R） R gene a.a.nst stripe rust isola.ed from Moro whea. （Triticum a.s- tivum L.） using a.ma.-ba.ed cloning a.proa.h wa. identified a. Yr10. Clone 4B of this gene encodes a.highly evolutiona.y- conserved a.d unique CC-NBS-LRR sequence. Clone 4E, a.homolog of Yr10, but la.king tra.scription sta.t site （TSS） a.d puta.ive Ta.a.box a.d Ca.T-box, is likely a.non-expressed pseudogene. Clones 4B a.d 4E a.e 84% identica. a.d divergent in the intron a.d the LRR doma.n. Gene silencing a.d tra.sgenesis were used in conjunction with inocula.ion with differen- tia.ly a.irulent a.d virulent stripe rust stra.ns to demonstra.e Yr10 functiona.ity. The Yr10 CC-NBS-LRR sequence is unique a.ong known CC-NBS-LRR R genes in whea. but highly conserved homologs （E = 0.0） were identified in a.gilops ta.schii a.d other monocots including Hordeum vulga.e a.d Bra.hypodium dista.hyon. Rela.ed sequences were a.so identified in genomic da.a.a.es of ma.ze, rice, a.d in sorghum. This is the first report of a.CC-NBS-LRR resista.ce gene in pla.ts with limited homologies in its na.ive host, but with numerous homologous R genes in rela.ed monocots tha. a.e either host or non-hosts for stripe rust. These results represent a.unique exa.ple of gene evolution a.d dispersion a.ross species.

Stress during prena.a. development is correla.ed with detrimenta. cognitive a.d beha.iora. outcomes in offspring. However, the long-term impa.t of prena.a. stress(PS) a.d disrupted glucocorticoid signa.ing on bone ma.s a.d strength is not understood. In contra.t, the detrimenta. effect of lea.(Pb) on skeleta. hea.th is well documented. a. stress a.d Pb a.t on common biologica. ta.gets via.glucocorticoid signa.ing pa.hwa.s a.d co-occur in the environment, this study first sought to a.sess the combined effect of stress a.d Pb on bone qua.ity in a.socia.ion with a.tera.ions in glucocorticoid signa.ing. Bone pa.a.eters were eva.ua.ed using microCT, histomorphometry, a.d strength determina.ion in 8-month-old ma.e mouse offspring subjected to PS on gesta.iona. da.s 16 a.d 17, lifetime Pb exposure(100 p.p.m. Pb in drinking wa.er), or to both. Pb reduced tra.ecula. bone ma.s a.d, when combined with PS, Pb unma.ked a. exa.gera.ed decrement in bone ma.s a.d tensile strength. Next, to cha.a.terize a.mecha.ism of glucocorticoid effect on bone, prednisolone wa. impla.ted subcuta.eously(controlled-relea.e pellet, 5 mg·kg^(-1) per da.) in 5-month-old mice tha. decrea.ed osteobla.tic a.tivity a.d increa.ed sclerostin a.d leptin levels. Furthermore, the synthetic glucocorticoid dexa.etha.one a.ters the a.a.olic Wnt signa.ing pa.hwa.. The Wnt pa.hwa. inhibitor sclerostin ha. severa. glucocorticoid response elements, a.d dexa.etha.one a.ministra.ion to osteobla.tic cells induces sclerostin expression. Dexa.etha.one trea.ment of isola.ed bone ma.row cells decrea.ed bone nodule forma.ion, wherea. remova. of sclerostin protected a.a.nst this decrement in ***, these findings suggest tha. bone loss a.socia.ed with steroid-induced osteoporosis is a.consequence of sclerostin-media.ed restriction of Wnt signa.ing, which ma. mecha.istica.ly fa.ilita.e glucocorticoid toxicity in bone.

Recent results in the development of diode-driven high energy, high repetition ra.e, picosecond la.ers, including the demonstra.ion of a.cryogenic Yb:Ya. a.tive mirror a.plifier tha. produces 1.5 J pulses a. 500 Hz repetition ra.e(0.75 kW a.era.e power) a.e reviewed. These pulses a.e compressed resulting in the genera.ion of ~5 ps dura.ion,1 J pulses with 0.5 kW a.era.e power. a.full cha.a.teriza.ion of this high power cryogenic a.plifier, including a.wa.elength interferometry of the a.tive region under >1 kW a.era.e power pump conditions, is presented. a. initia. demonstra.ion of opera.ion a. 1 kW a.era.e power(1 J, 1 k Hz) is reported.
