Repression of the Arabidopsis thaliana Jasmonic Acid/Ethylene-Induced Defense Pathway by TGA-Interacting Glutaredoxins Depends on Their C-Terminal ALWL Motif

Glutaredoxins are small heat-stable oxidoreductases that transfer electrons from glutathione （GSH） to oxi- dized cysteine residues, thereby contributing to protein integrity and regulation. In Arabidopsis thaliana, floral glutare- doxins ROXY1 and ROXY2 and pathogen-induced ROXY19/GRX480 interact with bZIP transcription factors of the TGACG （TGA） motif-binding family. ROXY1, ROXY2, and TGA factors PERIANTHIA, TGA9, and TGA10 play essential roles in floral development. In contrast, ectopically expressed ROXY19/GRX480 negatively regulates expression of jasmonic acid （JA）/ ethylene （ET）-induced defense genes through an unknown mechanism that requires clade II transcription factors TGA2, TGA5, and/or TGA6. Here, we report that at least 17 of the 21 land plant-specific glutaredoxins encoded in the Arabidopsis genome interact with TGA2 in a yeast-two-hybrid system. To investigate their capacity to interfere with the expression of JA/ET-induced genes, we developed a transient expression system. Activation of the ORA59 （OCTADECANOID-RESPONSIVE ARABIDOPSIS AP2/ERF-domain protein 59） promoter by transcription factor EIN3 （ETHYLENE INSENSITVE 3） was sup- pressed by co-expressed ROXY19/GRX480. Suppression depended on the L＊＊LL motif in the C-terminus of ROXY19/ GRX480. This putative protein interaction domain was recently described as being essential for the TGA/ROXY interaction. Ten of the 17 tested ROXY proteins suppressed ORA59 promoter activity, which correlated with the presence of the C-terminal ALWL motif, which is essential for ROXY1 function in flower development. ROXY19/GRX480-mediated repres- sion depended on the GSH binding site, suggesting that redox modification of either TGA factors or as yet unknown target proteins is important for the suppression of ORA59 promoter activity.

Little is known on the effects of biochar on N uptake and amino acid variability in crops such as winter rye and narrow-leafed lupine despite the fact that amino acids are important indicators,for food quality and plant stress.N uptake of both crops showed contrasting results when treated with different biochar *** amino acid contents referred to total nitro-gen generally tend to decrease in rye grains in the presence of biochar;whereas lupine seeds were more or less unaffected by biochar combined with mineral fertilizer or *** lupine seeds,total amino acid contents significantly increased when biochar was mixed with digestate but decreased when mixed with fermented ***,one of the most limiting amino acids in cereals,reached the recommended value of 4 g kg^(−1) in rye grain for most biochar *** lupine seeds,lysine decreased when biochar had been applied but were still in the recommended range when used as animal ***,an indicator for plant stress,significantly decreased(−49%)in rye when 2 Mg biochar ha^(−1) was added in combination with mineral *** contrast,proline increased when biochar was added to organic(digestate and compost)fertilizers(up to 43%).Further biochar research should focus much more on food quality,which is a key challenge for global food production.

While it is known that patatin-related phospholipase A （pPLA） activity is rapidly activated within 3min by auxin, hardly anything is known about how this signal influences downstream responses like transcription of early auxin-induced genes or other physiological responses. We screened mutants with T-DNA insertions in members of the pPLA gene family for molecular and physiological phenotypes related to auxin. Only one in nine Arabidopsis thaliana ppla knockdown mutants displayed an obvious constitutive auxin-related phenotype. Compared to wild-type, ppla-IIlδ mutant seedlings had decreased main root lengths and increased lateral root numbers. We tested auxin-induced gene expression as a molecular readout for primary molecular auxin responses in nine ppla mutants and found delayed up- regulation of auxin-responsive gene exRression in all of themL Thirty minutes after auxin treatment, up-regulation of up to 40% of auxin-induced genes was delayed in mutant seedlings. We observed only a few cases with hypersensitive auxin-induced gene expression in ppla mutants. While, in three ppla mutants, which were investigated in detail, rapid up- regulation （as early as 10 min after auxin stimulus） of auxin-regulated genes was impaired, late transcriptional responses were wild-type-like. This regulatory or dynamic phenotype was consistently observed in different ppla mutants with delayed up-regulation that frequently affected the same genes. This defect was not affected by pPLA transcript levels which remained constant. This indicates aposttranslational mechanism as a functional link of pPLAs to auxin signaling. The need for a receptor triggering an auxin response without employing transcription regulation is discussed.

Effects of supplementation of green tea extract on the milk performance of peripartal dairy cows and the expression of stress response genes in the liver

Background: We hypothesised that supplementation of green tea extract(GTE) in dairy cows during the transition period can attenuate proinflammatory conditions and prevent endoplasmic reticulum(ER) stress in the liver of these cows. Thirty Holstein cows with an average parity of 3.06(± 1.31, SD) were divided into a control group and a group that received a daily amount of 10 g of GTE from d 7 before the calving day and a daily amount of 20 g of GTE from the day of calving until d 7 of ***: Cows supplemented with GTE did not show differences in energy intake or milk yield in weeks 2–7 of lactation. However, these cows had a lower milk fat concentration and a lower energy corrected milk yield than the control cows and showed a trend of improved energy balance. The relative m RNA concentrations of proinflammatory genes, genes involved in the acute phase reaction and antioxidant genes in the liver in weeks 1, 4 and 7 of lactation were not different between the two groups of cows. The concentrations of α-tocopherol and the Trolox equivalent antioxidant capacity in plasma were not different between the two groups. However, the group supplemented with GTE showed significant reductions of some genes of the unfolded protein response(UPR) in week 1 and a trend of lower liver triacylglycerol(TAG) concentrations in the liver compared to the control ***: This study shows that supplementation of GTE in dairy cows lowers the fat concentration in the milk but overall has no effect on the expression of inflammatory genes and the antioxidative status in dairy cows during early lactation. The finding of reduced m RNA levels of genes involved in the UPR at week 1, however, supports other results showing that supplementation of polyphenols could prevent the development of ER stress in the liver of cows during early lactation. Ultrasound screening of the kidneys and urinary tract in 11.887 newborn infants: A 10-year experience

Objective: To determine the usefulness of sonographic screening of the newborn kidneys and urinary tract over a 10-year time period. Methods: Ultrasound screening of the kidneys and the urinary tract was performed between the third and tenth day after birth on 11.887 newborn infants. The classification of renal pyelectasis (RPE) according to the Society for Fetal Urology (SFU) was used. We evaluated infant renal pathologies and correlated them with the gestational age and weight at birth. Results: Renal pyelectasis (grades 1-4) was detected in 179 cases (1.5%);it was significantly more common in male (vs female) infants (p 4.000 g were the most significant risk factors (p < 0.01). Conclusions: Renal pyelectasis grades 1-4 was detected in only 1.5% of 11.887 consecutive infants subjected to sonographic screening of the kidneys and the urinary tract within the first 10 days after birth. Sonographic screening of the kidneys and the urinary tract is a non-invasive and effective screening method after birth. It allows planning for appropriate diagnostic tests and therapeutic procedures in a timely fashion.
