Sequential carbohydrate synthesis is important for plant survival because it guarantees energy supplies for growth and development during plant ontogeny and reproduction. Starch and fructan are two important carbohydrates in many flowering plants and in human diets. Understanding this coordinated starch and fructan synthesis and unraveling how plants allocate photosynthates and prioritize different carbohydrate synthesis for survival could lead to improvements to cereals in agriculture for the purposes of greater food security and production quality. Here, we report a system from a single gene in barley employing two alternative promoters, one intronic/exonic, to generate two sequence-overlapping but functionally opposing transcription factors, in sensing sucrose, potentially via sucrose/gluco lose 6-phosphate signaling. The system employs an autoregulatory mechanism in perceiving a sucrose- controlled trans activity on one promoter and orchestrating the coordinated starch and fructan synthesis by competitive transcription factor binding on the other promoter. As a case in point for the physiological roles of the system, we have demonstrated that this multitasking system can be exploited in breeding barley with tailored amounts of fructan to produce healthy food ingredients. The identification of an intron/exon-spanning promoter in a hosting gene, resulting in proteins with distinct functions, adds to the complexity of plant genomes.

In a study published in Cell,Pozniak et *** the spectrum of the phenotypic cell states of human metastatic melanomas in their tumor microenvironment before and early during immune checkpoint inhibitor(ICI)therapy and identified an enrichment of mesenchymal-like(MES)state melanoma cells in non-responders to ICI.1 These findings offer valuable insights into the factors contributing to the refractoriness of certain melanoma patients to ICI,while also unveiling novel molecular targets that hold promise for improving their treatment outcomes.

Construction of Social Cohesion and Living Together in the Cameroonian Public Space: Differences of Opinion versus the Emergence of a Common Sense in Discourse

Social cohesion and living together increasingly appear to be a response by the public authorities to the emergence of identity claims in Cameroon, in a context marked by the Anglophone crisis. Indeed, the socio-historical evolution reveals that living together in Cameroonian society changes considerably according to the social crises that animate it. It is therefore necessary to question the construction of these two concepts, in a dynamic marked by the reconfiguration of the public space characterised by occurrences in journalistic discourse. The aim of this proposal is to analyse the different conceptions that emerge from debates on cohesion and living together in the Cameroonian public space, in order to detect the true meaning that emerges from them. Our general objective is to explore the possibilities of constructing a problematic in the public space, namely living together, based on what is said by the actors in televised debates. The observation of the public space in particular that circumscribed by televised debates, reveals a frequency of their discursive use. The questions we asked ourselves were the following: How are the notions of living together and social cohesion constructed in televised debates in Cameroon? How do citizens position themselves in the face of the multitude of identity allegiances with which they identify in the public space? In order to provide some answers to this question, we observed and analysed the statements made by actors on stage in televised debates on four television channels. We were particularly interested in the debates on living together and social cohesion, and by means of discourse analysis we were able to detect the content of these notions. The study of pickling of the EL21T6 substrate in a nitric acid bath firstly provided knowledge of the influence of the different experimental parameters(concentration of acid,temperature and stirring of the solution)in relation to the etching *** experimental part led to the choice of standard pickling(T_(bath)=25℃,moderate stirring(250 rpm),[HNO_(3)]=1.20 mol/L,duration of 2 min)so as to obtain constant removal of material,even after several uses of the pickling *** observations also confirmed that in these operating conditions,pickling concerns both the grains of the matrix and the precipitates of type Mg3(Nd,Gd),leading to uniform removal of material from the *** from a number of assumptions,thermodynamic and chemical kinetic studies then allowed a pickling mechanism to be proposed and led to obtaining values for the corresponding kinetic parameters(order of reaction,constant of mean rate,activation energy)to be obtained.

Communication on Road Safety in Cameroon: Representation in the Press and Government Strategy

Road safety is a major concern in Cameroon, due to the multiplicity of road accidents resulting in the loss of human lives and the deterioration of road infrastructures. In 2019, the year marking the end of the United Nations decade of actions for road safety, the number of deaths on Cameroonian roads was 937. In this article, we have selected to examine the theme of road safety in the Cameroonian press and to look at awareness-raising strategies on the same subject. Our objective is to highlight the importance of communication in solving road safety problems. We therefore asked ourselves the following questions: How is the topic of road safety dealt with in the media? In combination with awareness campaigns, are the representations conveyed through the press sufficient to contribute to a change in road users behaviour? The option of studying both the representations of road safety that we have chosen has enabled us to show how the co-production of this public problem can be explained through the interrelations between the media and institutional fields. We initially postulated that communication achieves its objective in road safety campaigns when it integrates mixed strategies including campaigns aimed at road users, information for the general public through the press and social marketing campaigns aimed at changing behaviour. After a content analysis of articles published on road safety in three newspapers and an analysis of the messages disseminated in the posters and flyers produced during awareness campaigns, we found that awareness-raising through the media is not enough on its own. We therefore conclude that it is important to combine marketing techniques, citizen education and awareness raising aimed at changing user behaviour in road safety communication strategies. Effects of Feeding Programs Based on One or Two Milk Replacer Daily Meals on Growth, Solid Feed Intake and Rumen Fermentation and Development of Dairy Calves

Sixteen Holstein calves were used to study the effects of two feeding programs (FP) on growth, intake, rumen development and ruminal metabolism from birth to weaning. Two feeding programs based on milk replacer (MR) were tested: a once a day (OAD) MR (200 g/L) distribution vs. a standard twice a day (TAD) MR (125 g/L) distribution. All calves received water, wheat straw and a starter concentrate ad libitum. Four calves per group were slaughtered at weaning and rumen epithelium from the ventral sac was sampled for papillae (RP) density. Results showed that the FP had no effect on body weight of calves and total feed intake. From day 42 to day 56, ruminal pH was lower (P = 0.036) and ruminal oxydo-reducing potential was higher (P = 0.001) in OAD than TAD calves. Ruminal total volatile fatty acid (VFA) concentrations did not significantly differ between FP. From day 21 to day 63, butyrate ruminal concentration was significantly higher in OAD than TAD calves (5.17 vs 3.95 mmol/L). This probably explained the higher development of RP in calves fed once daily. Finally, the tested feeding system based on a once daily MR distribution affects the concentrate feeding pattern of calves.

A Bioenergetic-Redox Approach to the Effect of Live Yeast on Ruminal pH during Induced Acidosis in Dairy Cow

The objective of this study was to evaluate the capacity of the live yeast (LY) Saccharomyces cerevisiae in optimizing ruminal pH and in understanding its mode of action during induced acidosis in dairy cow. Two non-lactating cannulated cows were used and offered twice daily a control diet (CD) consisting of 51% corn silage and 49% concentrates or a LY diet (LYD) composed of CD supplemented with 4 g of LY per cow and per day. Measurements of pH and redox potential (Eh) were continuously made at 1 h interval during an experimental period of 9 h per day. Samples of ruminal fluid were also taken at 2 h intervals for analyses of volatile fatty acids (VFA) and lactic acid. Oxygen partial pressure (logPO2) in the ruminal milieu was calculated from the Nernst equation, using either O2-H2O or lactate-propionate redox couples. The results showed an increase of 0.2 unit in ruminal pH when LYD was fed, which was accompanied by a mean difference in Eh of -20 mV with respect to CD. The logPO2 decreased significantly by 0.8 log unit for LYD when compared to CD. Concentrations of VFA and proportion of propionate were higher with LYD (114.4 mM and 17.1% total VFA) compared to CD (102.4 mM and 15.4% total VFA). Proportion of butyrate decreased (from 15.8% to 14.2% total VFA). Lactate concentration decreased by 55% on average. It is proposed that the stabilization of ruminal pH (>6) is the outcome of the LY ability to scavenge oxygen. In doing so, it increased the reducing capacity of the milieu favoring the production of total VFA and depressing the level of lactate. The LY oxygen-scavenging ability was put forward to account for the transformation of lactate (pKa = 3.86) into propionate (pKa = 4.87) under reduced ruminal conditions.

Thermal Properties of the Product of Palm Stearin and Linseed Oil Transesterification

Triacylglycerols are the main constituents of natural fats and oils and contribute directly to their texture and flavor. The DSC （differential scanning calorimetry） of fats and oils is usually performed in order to characterize their melting and crystallization characteristics. The effect of the chemically catalyzed interesterification of two vegetable oils, i.e. palm stearin and linseed oil, on the thermal behavior of the product is studied. The objective of this study is to analyze the impact of the transesterification reaction conditions and of the initial amounts of oils on the thermal behavior of the product. This work shows that both cooling and heating DSC thermograms undergo significant changes as the result of the mixture of linseed oil and palm stearin and of their transesterification.
