Coherent Raman scattering(CRs)microscopy is widely recognized as a powerful tool for tackling biomedical problems based on its chemically specific label-free contrast,high spatial and spectral resolution,and high ***,the clinical translation of CRs imaging technologies has long been hindered by traditional solid-state lasers with environmentally sensitive operations and large *** fibre lasers can potentially overcome these shortcomings but have not yet been fully exploited for CRs imaging,as previous implementations have suffered from high intensity noise,a narrow tuning range and low power,resulting in low image qualities and slow imaging ***,we present a novel high-power self-synchronized two-colour pulsed fibre laser that achieves excellent performance in terms of intensity stability(improved by 50 dB),timing jitter(24.3 fs),average power fluctuation(20 dB)and pulse width variation(wavenumber range(2700-3550 cm^(−1)).The versatility of the laser source enables,for the first time,high-contrast,fast CRs imaging without complicated noise reduction via balanced detection *** capabilities are demonstrated in this work by imaging a wide range of species such as living human cells and mouse arterial tissues and performing multimodal nonlinear imaging of mouse tail,kidney and brain tissue sections by utilizing second-harmonic generation and twophoton excited fluorescence,which provides multiple optical contrast mechanisms simultaneously and maximizes the gathered information content for biological visualization and medical *** work also establishes a general scenario for remodelling existing lasers into synchronized two-colour lasers and thus promotes a wider popularization and application of CRs imaging technologies.

A magnesium alloy containing essential,non-toxic,biodegradable elements such as Ca and Zn has been fabricated using a novel twin-roll casting process(TRC).Microstructure,mechanical properties,in vivo corrosion and biocompatibility have been assessed and compared to the properties of the rare earth(RE)element containing wE43 *** Mg-0.5 wt% Zn-0.5 wt% Ca exhibited fine grains with an average grain size ranging from 70 to 150μ*** properties of a TRC Mg-0.5Zn-0.5Ca alloy showed an ultimate tensile strength of 220 MPa and ductility of 9.3%.The TRC Mg-0.5Zn-0.5Ca alloy showed a degradation rate of 0.51±0.07 mm/y similar to that of the wE43 alloy(0.47±0.09 mm/y)in the rat model after 1 week of *** week 4 the biodegradation rates of both alloys studied were lowered and stabilized with fewer gas pockets around the *** histological analysis shows that both wE43 and TRC Mg-0.5Zn-0.5Ca alloy triggered comparable tissue healing responses at respective times of *** presence of more organized scarring tissue around the TRC Mg-0.5Zn-0.5Ca alloys suggests that the biodegradation of the RE-free alloy may be more conducive to the tissue proliferation and remodelling process.

In contrast to the cries of human infants, sounds made by non-human infants in different stressful behavioral contexts （hunger or physical discomfort, isolation, capture by humans or predators） are usually treated as distinct types of vocalizations. However, if distress vocalizations produced by different species and in different contexts share a common motivational state and associated neurochemical pathways, we can expect them to share a common acoustic structure and adaptive function, showing only limited variation that corresponds to the infant＇s level of arousal. Based on this premise, we review the acoustic structure and adaptive function of two types of distress calls, those given when infants were isolated from their mothers （isolation caUs） or captured by humans （capture calls）. we conducted a within-context comparison examining the two call types across a diverse se- lection of mammalian species and other vertebrate groups, followed by a comparison of how acoustic structure and function dif- fers between these contexts. In addition, we assessed acoustic traits that are critical to the response of caregivers. Across verte- brate species, distress vocalizations produced in these two behavioral contexts tend to be tonal with a simple chevron, flat or de- scending pattern of frequency modulation. Reports that both isolation and capture calls of vertebrate infants serve to attract care- givers are universal, and the fundamental frequency of infant vocalizations is often critical to this response. The results of our re- view are consistent with the hypothesis that differences in the acoustic structure of isolation and capture distress vocalizations re- flect differences in arousal, and not discrete functions. The similarity in acoustic structure and caregiver response observed across vertebrates adds support to the hypothesis that the production and processing of distress vocalizations are part of a highly-conserved system of social vocal behaviour in vertebrate

Performance of alpha-defensin lateral flow test after synovial fluid centrifugation for diagnosis of periprosthetic knee infection

BACKGROUND The quantitative alpha-defensin enzyme-linked immunosorbent assay(ELIsA)demands a prior synovial fluid centrifugation,whereas this processing is not routinely required prior to the alpha-defensin lateral flow *** To evaluate whether a prior synovial fluid centrifugation could lead the lateral flow performance to achieve comparable results to ELIsA during periprosthetic joint infection(PJI)*** Fifty-three cases were included in this study:22 classified as PJI and 31 classified as aseptic cases,according to Musculoskeletal Infection society 2013 *** fluid samples were submitted to centrifugation,and the supernatant was evaluated by ELIsA and lateral flow *** sensitivity(sE),specificity(sP)and accuracy of each method were calculated as well as the agreement between those two *** In all of the 31 samples from aseptic patients,alpha-defensin ELIsA and lateral flow tests showed negative results for *** the 22 infected patients,the lateral flow test was positive in 19 cases(86.4%)and the ELIsA was positive in 21(95.5%).sensibility,sP and accuracy were,respectively,86.4%(95%CI:65.1%-97.1%),100%(95%CI:88.8%-100%)and 93.2%(95%CI:82.8%-98.3%)for the lateral flow test and 95.5%(95%CI:77.2%-99.9%),100%(95%CI:88.8%-100%)and 98.1%(95%CI:89.9%-100%)for *** agreement of 96.2%between those methods were *** statistical difference was found between them(P=0.48).CONCLUsION Alpha-defensin lateral flow test showed high sE,sP and accuracy after a prior synovial fluid centrifugation,achieving comparable results to *** the lower complexity of the lateral flow and its equivalent performance obtained in this condition,a prior centrifugation might be added as a valuable step to enhance the PJI diagnosis.
