Chromatin condensation but not DNA integrity of pig sperm is greater in the sperm-rich fraction

Background Protamination and condensation of sperm chromatin as well as DNA integrity play an essential role during fertilization and embryo development.In some mammals,like pigs,ejaculates are emitted in three separate fractions:pre-sperm,sperm-rich(SRF)and post sperm-rich(PSRF).These fractions are known to vary in volume,sperm concentration and quality,as well as in the origin and composition of seminal plasma(SP),with differences being also observed within the SRF one.Yet,whether disparities in the DNA integrity and chromatin condensation and pro-tamination of their sperm exist has not been interrogated.Results This study determined chromatin protamination(Chromomycin A3 test,CmA_(3)),condensation(Dibromobi-mane test,DBB),and DNA integrity(Comet assay)in the pig sperm contained in the first 10 m L of the SRF(SRF-P1),the remaining portion of the sperm-rich fraction(SRF-P2),and the post sperm-rich fraction(PSRF).While chromatin protamination was found to be similar between the different ejaculate fractions(P>0.05),chromatin condensation was seen to be greater in SRF-P1 and SRF-P2 than in the PSRF(P=0.018 and P=0.004,respectively).Regarding DNA integrity,no differences between fractions were observed(P>0.05).As the SRF-P1 has the highest sperm concentra-tion and ejaculate fractions are known to differ in antioxidant composition,the oxidative stress index(OSi)in SP,calcu-lated as total oxidant activity divided by total antioxidant capacity,was tested and confirmed to be higher in the SRF-P1 than in SRF-P2 and PSRF(0.42±0.06 vs.0.23±0.09 and 0.08±0.00,respectively;Pm concentration of each fraction(Rs=0.973;Pm DNA integrity was not found to differ between ejaculate fractions,SRF-P1 and SRF-P2 were observed to exhibit greater chromatin condensation than the PSRF.This could be related to the OSi of each fraction.

The upper digestive tract is routinely scoped for several causes of malabsorption, and the number of duodenal biopsy specimens has increased notably in the last 10 years. Gluten-sensitive enteropathy(GSE) is an autoimmune disease, which shows an increasing prevalence worldwide and requires a joint clinicopathological approach. The classical histopathology of GSE with partial or total villous blunting is well recognized, but the classification of GSE is not straightforward. moreover, several mimickers of GSE with intraepithelial lymphocytosis have been identified in the last 20 years, with drug interactions and medical comorbidities adding to the conundrum. In this review, we report on the normal duodenal mucosa, the clinical presentation and laboratory diagnosis of GSE, the duodenal intraepithelial lymphocytes and immunophenotype of GSE-associated lymphocytes, the GSE mimickers, the differences "across oceans" among guidelines in diagnosing GSE, and the use of a synoptic report for reporting duodenal biopsies in both children and adults in the 21^(st) century.

Background Gianotti-Crosti syndrome is characterized by an acute onset of a papular or papulovesicular eruption with a symmetrical distribution.Data sources A Pubmed search was conducted using Clinical Queries with the key terms"Gianotti-Crosti syndrome"OR"papular acrodermatitis".The search strategy included meta-analyses,randomized controlled trials,clinical trials,observa-tional studies,and reviews.This paper is based on,but not limited to,the search results.Results The eruption of Gianotti-Crosti syndrome is found predominantly on the cheeks,extensor surfaces of the extremities,and buttocks.There is a sparing of antecubital and popliteal fossae as well as palms,soles,and mucosal surfaces.Although often asymptomatic,the lesions may be mildly to moderately pruritic.Gianotti-Crosti syndrome is most common in children between 1 and 6 years of age.The Epstein-Barr virus and the hepatitis B virus are the most common pathogens associated with Gianotti-Crosti syndrome.No treatment for Gianotti-Crosti syndrome is necessary because it is self-limited.In an era of vaccine hesitancy and refusal,Gianotti-Crosti syndrome may be important to mention to parents,because it can occur and trigger alarmism.Conclusions Gianotti-Crosti syndrome is mainly a disease of early childhood,characterized by an acute onset of a papular or papulovesicular eruption with a symmetrical distribution.With the advent of more universal vaccination against hepatitis B virus,Epstein-Barr virus has become the most common etiologic agent of Gianotti-Crosti syndrome.Few cases of post-vaccination Gianotti-Crosti syndrome have been reported.Currently,the emphasis should be placed on its self-limiting attribution.

In the 21st century,the determination of alert thresholds remains the most challenging and controversial issue in clinical pediatrics.Pre-analytical,analytical,and postanalytical matters will consolidate or undermine the fate of any laboratory process.Pre-analytical issues need to be cleared off before the laboratory physician can dispatch the result to the pediatrician in charge.Once it is cleared off,the classification of essential laboratory results is paramount.It is more than an academic exercise and may be subdivided in the order of priority we handle it to inform promptly and safely the primary physicians.Currently,we are applying new modes of making sure relevant information is transmitted without interrupting the standard workflow of the primary physicians in charge for the child,who eventually need a fast line of action for results that may be lifethreatening.

Fibrolamellar carcinoma (FL-HCC) is overall a quite rare variant of hepatocellular carcinoma (HCC)?that is intriguing and poorly understood at same time. In humans, it affects essentially the young population (children and young adults) in the peculiar absence of underlying liver diseases. FL-HCC has distinctive?clinico-pathologic features with metastases frequently found in the regional lymph nodes, but also peritoneum and lungs. Although initially considered harboring an indolent course, survival outcomes have been recently updated pointing to a neoplasm concealing many aspects of biogenesis. In this review, morphology, molecular pathways, and prognosis of this rare tumor are presented.

microRNAs(miRNAs)are small single-stranded,non-coding RNA molecules involved in the pathogenesis and progression of cancer,including osteosarcoma.We aimed to clarify the pathways involving miRNAs using new bioinformatics tools.We applied WikiPathways and PathVisio,two open-source platforms,to analyze miRNAs in osteosarcoma using miRTar and ONCO.IO as integration tools.We found 1298 records of osteosarcoma papers associated with the word“miRNA”.In osteosarcoma patients with good response to chemotherapy,miR-92a,miR-99b,miR-193a-5p,and miR-422a expression is increased,while miR-132 is decreased.All identified miRNAs seem to be centered on the TP53 network.This is the first application of PathVisio to determine miRNA pathways in osteosarcoma.miRNAs have the potential to become a useful diagnostic and prognostic tool in the management of osteosarcoma.PathVisio is a full pathway editor with the potentiality to illustrate the biological events,augment graphical elements,and elucidate all the physical structures and interactions with standard external database identifiers.

Leaf senescence is a complex phenomenon occurring in all plant species, but it is still poorly understood in plants grown in mediterranean field conditions and well-adapted to harsh climatic conditions. To better understand the physiological processes underlying leaf senescence in mastic trees （Pistacia lentiscus L.）, we evaluated leaf growth, water and N content, photosystem II （PSII） photochemistry, lipid peroxidation and levels of photosynthetic pigments, antioxidants, abscisic acid, and salicylic acid and jasmonic acid during the complete leaf lifespan, from early expansion to late senescence in relation to natural climatic conditions in the field. While mature leaves suffered from water and N deficit during late spring and summer, both young （emerging） and old （senescing） leaves were most sensitive to photo- oxidative stress, as indicated by reductions in the Fv/Fm ratio and enhanced lipid peroxidation during late autumn and winter. Reductions in the FvlFm ratio were associated with low ^-tocopherol （vitamin E） levels, while very old, senescing leaves additionally showed severe anthocyanin losses. We have concluded that both young （emerging） and old （senescing） leaves suffer oxidative stress in mastic trees, which may be linked in part to suboptimal temperatures during late autumn and winter as well as to low vitamin E levels.

Hirschsprung's disease(HSCR) or congenital megacolon is one of the differential diagnoses of chronic constipation mostly in infancy and may indeed represent a challenge for pediatricians, pediatric surgeons, and pediatric pathologists. The diagnosis relies clearly on the identification of the absence of ganglion cells at the plexuses(submucosus and myentericus) of the bowel wall. HSCR is usually located at the terminal(distal) rectum with potential pre-terminal or proximal extension to the less distal large bowel(sigmoid colon). Astonishingly, there is some evidence that Hindu surgeons of prehistoric India may have been exposed and had considerable knowledge about HSCR, but this disease is notoriously and eponymously named to Dr. Harald Hirschsprung(1830-1916), who brilliantly presented two infants with fatal constipation at the Berlin conference of the German Society of Pediatrics more than one century ago. Historical milestones and diagnosis of HSCR(originally called "Die Hirschsprungsche Krankheit") are reviewed. more than 100 years following his meticulous and broad description, HSCR is still a puzzling disease for both diagnosis and treatment. HSCR remains a critical area of clinical pediatrics and pediatric surgery and an intense area of investigation for both molecular and developmental biologists.
