congenital bilateral absence of the vas deferens(CBAVD)is observed in 1%–2%of males presenting with infertility and is clearly associated with cystic fibrosis transmembrane conductance regulator(CFTR)*** is one of the most well-known genes related to male *** frequency of CFTR mutations or impaired CFTR expression is increased in men with nonobstructive azoospermia(NOA).CFTR mutations are highly polymorphic and have established ethnic *** with F508Del in Caucasians,the p.G970D mutation is reported to be the most frequent CFTR mutation in Chinese patients with cystic ***,whether p.G970D participates in male infertility remains ***,a loss-of-function CFTR p.G970D missense mutation was identified in a patient with CBAVD and *** retrospective analysis of 122 Chinese patients with CBAVD showed that the mutation is a common pathogenic mutation(4.1%,5/122),excluding polymorphic ***,we generated model cell lines derived from mouse testes harboring the homozygous Cftr p.G965D mutation equivalent to the CFTR variant in *** Cftr p.G965D mutation may be lethal in spermatogonial stem cells and spermatogonia and affect the proliferation of spermatocytes and Sertoli *** spermatocyte GC-2(spd)ts(GC2)Cftr p.G965D cells,RNA splicing variants were detected and CFTR expression decreased,which may contribute to the phenotypes associated with impaired ***,this study indicated that the CFTR p.G970D missense mutation might be a pathogenic mutation for CBAVD in Chinese males and associated with impaired spermatogenesis by affecting the proliferation of germ cells.

A monolithic 3D IGZO-RRAM-SRAM-integrated architecture for robust and efficient compute-in-memory enabling equivalent-ideal device metrics

Compute-in-memory(CIM) based on various devices such as static random access memory(SRAM) and resistive random access memory(RRAM) and other emerging devices such as indium-gallium-zinc-oxide(IGZO) transistor, magnetoresistive RAM(MRAM), and ferroelectric RAM(FeRAM) has been explored for better performance and energy efficiency on neural networks applications. However, CIM based on a single-type device suffers a variety of non-ideal metrics to reach the simultaneous optimal accuracy, density, and energy efficiency. This work presents equivalent-ideal CIM(Eq-CIM), a monolithic 3D(M3D) IGZO-RRAM-SRAM integrated architecture for robust and efficient compute-in-memory enabling equivalent-ideal device metrics. To overcome the non-ideality(variation, endurance, temperature, etc.) of the single-type devices, system-technology co-optimization(STCO) is performed. This work highlights the non-ideality-aware functionality breakdown for robust high accuracy and simultaneous high density/efficiency, by utilizing 2T0C IGZO for temporal activation storage, RRAM for high-density weight storage, and SRAM for accurate CIM. Device-to-algorithm variation transfer is applied to analyze the system-level accuracy. We benchmark Eq-CIM architecture on CIFAR-10/ImageNet, with 5.06× storage density and 5.05×/2.45× area/energy efficiency compared with single-type-device-based CIM, and high robustness(<0.27% accuracy loss) from-40℃ to 120℃.

BACKGROUND Hepatocellular carcinoma(HCC)is a malignant disease with high incidence and mortality *** study focuses on the TP53 target protein to investigate the potential therapeutic effect of tetrahydrocurcumin(THC)on HCC and its mechanism of *** research hypothesis is that THC can inhibit the proliferation,migration,and invasion of HCC cells,and promote their apoptosis by regulating the TP53 target *** To explore the mechanism by which THC inhibits HCC cell proliferation via the TP53 signaling *** Potential targets of THC and HCC were identified from multiple *** core targets were subjected to analyses using Gene Ontology and Kyoto Encyclopedia of Genes and Genomes databases,and visualization processing,using the online platform Metascape to identify the key molecules and signaling pathways involved in the action of THC against *** molecular mechanisms of action of THC against TP53 in the inhibition of HCC cells were verified using cell counting kit-8,Transwell,apoptosis,and western blotting *** Molecular docking results showed that THC had a high score for the TP53 target *** vitro experiments indicated that THC effectively inhibited the proliferation and migration of HCC cells,and affected the expression levels of TP53,MDM2,cyclin B,Bax,Bcl-2,caspase-9,and *** THC induces the apoptosis of HCC cells through the TP53 signaling pathway,thereby inhibiting their proliferation and migration.

Conducting predictability studies is essential for tracing the source of forecast errors,which not only leads to the improvement of observation and forecasting systems,but also enhances the understanding of weather and climate *** the past few decades,dynamical numerical models have been the primary tools for predictability studies,achieving significant ***,with the advances in artificial intelligence(AI)techniques and accumulations of vast meteorological data,modeling weather and climate events using modern data-driven approaches is becoming trendy,where FourCastNet,Pangu-Weather,and GraphCast are successful *** this perspective article,we suggest AI models should not be limited to forecasting but be expanded to predictability studies,leveraging AI's advantages of high efficiency and self-contained optimization *** this end,we first remark that AI models should possess high simulation capability with fine spatiotemporal resolution for two kinds of predictability *** models with high simulation capabilities comparable to numerical models can be considered to provide solutions to partial differential equations in a data-driven ***,we highlight several specific predictability issues with well-determined nonlinear optimization formulizations,which can be well-studied using AI models,holding significant scientific *** addition,we advocate for the incorporation of AI models into the synergistic cycle of the cognition–observation–model *** predictability studies have the potential to transform“big data”to“big and better data”and shift the focus from“AI for forecasts”to“AI for science”,ultimately advancing the development of the atmospheric and oceanic sciences.

Solar-driven interfacial evaporation is one of the most attractive approaches to addressing the global freshwater ***,achieving an integrated high evaporation rate,salt harvesting,and multifunctionality in evaporator is still a crucial ***,a novel composite membrane with biomimetic micronanostructured superhydrophobic surface is designed via ultrafast laser etching ***,the double-transition-metal(V_(1/2)Mo_(1/2))_(2)CT_(x)MXene nanomaterials as a photothermal layer,exhibiting the enhanced photothermal conversion performance due to elevated joint densities of states,which enables high populations of photoexcited carrier relaxation and heat release,provides a new insight into the photothermal conversion mechanism for multiple principal element ***,the(V_(1/2)Mo_(1/2))_(2)CT_(x)MXene-200 composite membrane can achieve a high evaporation rate of 2.23 kg m^(−2)h^(−1)under one sun,owing to the enhanced“light trap”effect,photothermal conversion,and high-throughput water ***,the membrane can induce the directed precipitation of salt at the membrane edge,thus enabling salt harvesting for recycling and zero-emission of brine ***,the composite membrane is endowed with excellent multifunctionality of anti-/de-icing,anti-fouling,and antibacterial,overcoming the disadvantage that versatility is difficult to be ***,the evaporator and the promising strategy hold great potential for the practical application of solar evaporation.
