Purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (RPE) cells, thereby potentially enabling subretinal implantation in patients. Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from Polycaprolactone (PcL), and different concentrations of gelatin (5%, 15% and 30%). ARPE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARPE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARPE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARPE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARPE-19 (RPE65 and Zo-1) qRT-PcR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of P 0.05 was considered to be statistically significant. Results ARPE-19 cells grew on PcL/gelatin membranes mainly in PcL/30% gelatin combination, which had not cytotoxic effect. RPE65/cytokeratin-18/ and actin-positive ARPE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARPE-19 cells fused among, suggesting the presence of tight junctions. The expression of RPE65 and Zo-1 was unchanged. conclusions: PcL/30% gelatine membranes may imitate the natural BM to such extent that they support RPE-cells and exhibited RPE-like morphology. cost-effectiveness of enhanced liver fibrosis test to assess liver fibrosis in chronic hepatitis c virus and alcoholic liver disease patients

AIM To assess liver fibrosis(LF) in hepatitis c virus(HcV) and alcoholic liver disease(ALD), estimate health outcomes and costs of new noninvasive testing strategies METHoDS A Markov model was developed to simulate LF progression in HcV and ALD for a cohort of 40-yearold men with abnormal levels of transaminases. Nurse care and comfort in the Puerperium of girls/Women: Protocol for a Scoping Review

Purpose: carrying out a scoping review to fill gaps in current knowledge regarding comfort needs in the care provided by nurses to girls/women who experience puerperium. Methods: The procedures guided by the Joanna Briggs Institute will be applied. The searches will be carried out in the Medical Literature Analysis and Retrieval System online, Excerpta Medica Database, cumulative Index to Nursing and Allied Health Literature, cochrane Library, Scopus, Web of Science, Latin American and caribbean Health Sciences Literature, Nursing Database, Scientific Electronic Library databases. Studies which are available in full and published in English, Spanish or Portuguese will be selected. There will be no restrictions to the study design or time frame. Two reviewers will independently screen all citations with the aid of software. The degree of agreement between the researchers will be verified by statistics that measure reliability. Through narrative descriptions, charts, and tables, we will present the results obtained. Data analysis will involve descriptive statistics, and qualitative evaluation. We will use the Preferred Reporting Items for Systematic Reviews and Meta-Analyses extension for Scoping Reviews checklist to review and write this review. conclusion: We will summarize the main information available in the literature on the subject, aiming to obtain an overview of the practices employed, and the gaps present in knowledge that require greater attention from the scientific community.

EgFr （Epidermal growth factor receptor） overexpression has been detected in many tumors of epithelial origin, specifically in breast cancer and it is often associated with tumor growth advantages and poor prognosis. The nimotuzumab is a genetically engineered humanized MAb （monoclonal antibody） that recognizes an epitope located in the extracellular domain of human EgFr. The aim of this study was to assess the pharmacokinetics of nimotuzumab in patients with locally advanced breast cancer who are receiving neoadyuvant therapy combined with the Ac chemotherapy regimen （i.e., 60 mg/m2 of Doxorubicin and 600 mg/m2 of cyclophosphamide in 4 cycles every 21 days）. A single center, non-controlled, open Phase I clinical trial, with histopathological diagnosis of locally advanced stage III breast cancer, was conducted in 12 female patients. Three patients were enrolled at each of the following fixed dose levels： 50, 100, 200 and 400 mg/week. Multiple intermittent short-term intravenous infusions of nimotuzumab were administered weekly, except on weeks 1 and 10, when blood samples were drawn for pharmacokinetic assessments. Nimotuzumab showed dose-dependent kinetics. No anti-idiotypic response against nimotuzumab was detected in blood samples of participants. There was not interaction between the administration of nimotuzumab and chemotherapy at the dose levels studied. The optimal biological doses ranging were estimated to be 200 mg/weekly to 400 mg/weekly.

本研究的主要目的是基于该区域现有的实时宽频带台站,研究地震预警系统(earthquake early warning system,EEWS)对潜在破坏性地震发出警告的可行性。研究区位于伊比利亚半岛(Iberian Peninsula)西南部的圣维森特角(cape San Vicente,SV)和加的斯湾(gulf of cádiz,gc)。该区域历史地震有1755年里斯本(Lisbon)M_w8.5地震和1969年圣维森特角Mw7.8地震。本研究阐明了基于美国地质调查局(U.S. geological Survey,USgS)的Earthworm工具,及由西班牙加泰罗尼亚地质制图研究所(Institut cartogràfici geològicde catalunya,Icgc)进一步研究出的地震预警系统的设计、配置和产出结果。系统的主要功能是实时数据采集、处理(利用预先估算的峰值位移Pd和P波信号的卓越周期τc进行P波拾取、地震事件监测、地震震源定位和震级估算)、数据归档和地震预警发布。系统在完成一些模拟实验并获得最佳配置后进行运行。在运行的第一年内,该区域发生了一次有感地震。与国家地理研究所(Instituto geográfico Nacional,IgN)地震目录相比较,定位和震级结果都相当好。对于大部分区域的预警时间都能达到几十秒,对于葡萄牙和西班牙南部沿海大部分区域来说,足以减轻由gc和SV地震所带来的损害。此系统的初步结果表明,该系统能可靠地、有效地用于伊比利亚西南部地区的可能性。
