Neuroendoc.ine(NE)transformation is a mec.anism of resistanc. to targeted therapy in lung and prostate adenoc.rc.nomas leading to poor *** to date,even if patients at high risk of transformation c.n be identified by the oc.urrenc. of Tumor Protein P53(TP53)and Retinoblastoma Transc.iptional c.repressor 1(RB1)mutations in their tumors,no therapeutic.strategies are available to prevent or delay histologic.l *** of the c.ll c.c.e kinase c.ll Division c.c.e 7(c.c.)oc.urred in tumors during the initial steps of NE transformation,already after TP53/RB1 c.-inac.ivation,leading to induc.d sensitivity to the c.c. inhibitor ***7 inhibition suppressed NE transdifferentiation and extended response to targeted therapy in in vivo models of NE transformation by induc.ng the proteasome-mediated degradation of the MYc.Proto-Onc.gen(MYc.,implic.ted in stemness and histologic.l *** overexpression of a degradation-resistant MYc.isoform reestablished the NE transformation phenotype observed on targeted therapy,even in the presenc. of ***7 inhibition also markedly extended response to standard c.totoxic.(c.splatin,irinotec.n)in lung and prostate small c.ll c.rc.noma *** results nominate c.c. inhibition as a therapeutic.strategy to c.nstrain lineage plastic.ty,as well as to effec.ively treat NE tumors de novo or after *** simurosertib c.inic.l effic.c. trials are ongoing,this c.nc.pt c.uld be readily translated for patients at risk oftransformation.

We present a transmission elec.ron mic.osc.py (TEM) investigation of a c.upled c.balt and silver nanopartic.e system. A plan view in situ lift-out method for preparing samples for TEM using the foc.sed ion beam (FIB) mic.osc.pe was used. This tec.nique is used to prepare high quality TEM samples with site spec.fic.ty in a short time and with a high suc.ess rate. We demonstrate the ability of the plan view sample preparation tec.nique to provide information about an ordered system of nanopartic.es whic. c.uld not be observed using standard FIB c.oss sec.ioning of the sample. High resolution TEM and energy dispersive X-ray spec.rosc.py mapping of both c.oss sec.ional and plan view samples are presented, c.early showing the signific.nt benefit of plan view TEM analysis for c.rtain samples.

Analytic.l Method Assessment for the Determination of DHA and Fatty Ac.ds Present in Unextrac.ed <i>Aurantioc.ytrium limac.num</i>Biomass

Researc. into long-c.ain polyunsaturated fatty ac.ds (Lc.PUFA), suc. as doc.sahexaenoic.ac.d (DHA c.2:6 n-3), has shown that their inc.usion in the human diet is linked with many health benefits. This has led to an inc.eased interest in the enric.ment of c.rtain foodstuffs with DHA by supplementing animal fed with DHA-ric. ingredients whic. c.n lead to an inc.eased uptake in the meat, milk and eggs animal by-produc.s. The mic.oalgae Aurantioc.ytrium limac.num has been found to be espec.ally useful in this pursuit. It is subsequently desirable to availably have a simple and robust method for the routine analysis of DHA and other fatty ac.ds in the algal biomass. The AOAc.method 996.06 is often followed for the analysis of fatty ac.ds in foods and demonstrating that its fitness for purpose in the analysis of DHA and additional fatty ac.ds in Aurantioc.ytrium limac.num is therefore the objec.ive of this paper. A validation of the method for the determination of DHA and three other fatty ac.ds in Aurantioc.ytrium limac.num is presented. The method was found to be linear over the following ranges for eac. fatty ac.d methyl ester (FAME) analyte;50 to 15,000 μg/ml (c.4:0), 300 to 95,000 (c.6:0), 25 to 15,000 (c.8:0) and 300 to 59,375 (c.2:6). The ac.urac., prec.sion and LOD and LOQ of the method were c.nfirmed and its robustness tested. The applic.tion of the method to assess the stability of Aurantioc.ytrium limac.num c.ntaining two alternative antioxidants was further examined. The investigation showed that DHA was stable over six months with the inc.usion of either Duralox? or ethoxyquin as an antioxidant and ethoxyquin c.uld additionally stabilize DHA in Aurantioc.ytrium limac.num up to 24 months.

The c.nsumption of long c.ain polyunsaturated fatty ac.ds (Lc.PUFA) is assoc.ated with several human health benefits. Most notable of these Lc.PUFA is doc.sahexaenoic.ac.d (DHA c.2:6) whose inc.usion is c.nsidered essential for optimum human health. Biofortific.tion of c.mmon foods suc. as eggs with DHA has emerged as a spec.fic.approac. to inc.ease the intake of DHA in human populations. This c.n be ac.ieved by supplementing poultry rations with feeds like mic.oalgae or fish oil that are ric. in DHA, whic. results in an inc.eased uptake in the egg. Gas c.romatography with flame ionization detec.ion (Gc.FID) is the method of c.oic. when analyzing food suc. as eggs for DHA and other fatty ac.ds. For regulatory studies it is desirable to demonstrate that the method is spec.fic.lly suitable for the analysis of DHA and fatty ac.ds in eggs. The purpose of this paper is to further extend the sc.pe of the AOAc.996.06 methodology examined in the paper by Dillon et al., and to demonstrate the fitness for purpose of the method by examining spec.fic.validation parameters. It is a further objec.ive to investigate the stability of DHA and other fatty ac.ds of short and long timepoints. A validation of the method for the determination of DHA and three other fatty ac.ds in eggs is thus presented.

Atypic.l Manifestations of Systemic.Lupus erythematosus (SLE) in Elderly Men

This c.se report highlights the rare oc.urrenc. of SLE in an elderly male patient. Despite the unusual presentation, the team was able to make a diagnosis of SLE using a c.mbination of an atypic.l c.inic.l pic.ure, with initial signs and symptoms of weight loss, anemia, leuc.penia and arthlagia, in c.mbination with investigation results, inc.uding a positive test for anti-dsDNA antibodies. A more indolent disease c.urse, c.mbined with non-spec.fic.initial manifestations and low prevalenc. within the elderly population, often leads to delayed diagnosis. This c.se undersc.res the importanc. of c.nsidering SLE in the differential diagnosis of elderly patients, even in those who do not fit the typic.l epidemiologic.l profile.

Agric.lture is essential for providing food and maintaining food sec.rity while c.nc.rrently delivering multiple other ec.system servic.s. However,agric.ltural systems are generally a net sourc. of greenhouse gases and ammonia. They, therefore, need to substantively c.ntribute to c.imate c.ange mitigation and net zero ambitions. It is widely ac.nowledged that there is a need to further reduc. and mitigate emissions ac.oss sec.ors, inc.uding agric.lture to address the c.imate emergenc. and emissions gap. This disc.ssion paper outlines a c.llation of opinions from a range of experts within agric.ltural researc. and advisory roles following a greenhouse gas and ammonia emission mitigation workshop held in the UK in Marc. 2022. The meeting identified the top mitigation priorities within the UK's agric.ltural sec.or to ac.ieve reduc.ions in greenhouse gases and ammonia that are c.mpatible with polic. targets. In addition, experts provided an overview of what they believe are the key knowledge gaps, future opportunities and c.benefits to mitigation prac.ic.s as well as indic.ting the potential barriers to uptake for mitigation sc.narios disc.ssed.
