BACKGROUND: Since single nucleotide polymorphisms (SNPs) can serve as gene markers, polymorphism profiles may help scientists to identify the full collection of genes that contribute to the development of complex diseases such as cancer. The distribution of interleukin-10 (IL-10) promoter polymorphisms in Chinese Han ethnic patients with hepatitis B virus (HBV) infection and hepatocellular carcinoma (HCC) was investigated in this study. METHODS: The polymorphisms of IL-10 promoter region were detected by pulymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and sequencing. Sixty-six health controls, 42 patients with HBV infection, 30 HCC patients, and cell line SMMC-7721 were examined this way. RESULTS: Polyrnorphisms of T/C or T/N on-872 site occurred frequently in Han ethnic population. Pulyrnorphisms were detected in HBV and HCC patients and cell line SMMC-7721. The hotspot among the pulymorphisms was inserting base A between-1058 and-1057. CONCLUSION: Polymorphisms of IL-10 promoter in HBV and HCC patients may be associated with HBV infection and HCC development.

The understanding of cSNPs of cancer-related genes harboring in high frequency loss regions of tumor chromosomes can advance the disclosure of genetic and variant mechanisms of tumorigenesis,and the investigation of cancer susceptibility. In preparing a gene chip for detecting polymorphisms on coding region of genes in hepatocellular carcinoma tissues, some cSNPs are of interest for their potential links with phenotype. METHODS: The genes harboring in loss regions with high frequency of hepatocellular carcinoma (HCC) were selected, the related information of cSNP sequences was obtained from the SNP database (dbSNP) of the National Center for Biotechnology Information (NCBI). Then appropriate primers and oligonucleotide probes were designed according to the SNP sites, and a gene chip for the detection of SNPs was constructed. The chip included 48 cSNPs of 25 hepatocellular carcinoma-related genes. The PCR products labeled by Dig-dUTP were hybridized with the cSNP chip. RESULTS:The sensitivity, influence by probe concentration, and reiteration of the chip were detected, with a high sensitivity of 6 × 10-3 ng/μl. The signal of hybridization was reduced with a lower concentration of probe. Seven polymorphisms of caspase 9 (rs2308941) C →T and DOK2 (rs2242241) T→G, 6 of polymorphisms of EGFL3 (rs947345) A→G, caspase 9 (rs2308938) C→G and PHGDH (rs1801955)T→A, 5 of polymorphisms of E2F2(rs3218170) G→A,4 of polymorphisms of MUTYH( rs1140507) T→C and BNIP3L(rs1055806)G→T, and 1 of polymorphism of TNFRSF1B (rs1061622)T→G were detected by the chip in the tissues of 10 HCC. Samples of caspase 9 (rs2308941G) and (rs2308941A) were verified by PCR-SSCP and sequencing. CONCLUSION:The cSNP chip of hepatocellular carcinoma-related genes can accelerate the discovery of polymorphic markers on hepatocellular carcinoma.

The community composition,abundance and culturablity of heterotrophic bacterioplankton in the Dapeng Bay of the South china Sea were investigated by morphological method,bacterial cultivation and biochemical methods over an annual *** and Pseudomonas were the predominant genera among the Gram-negative *** bacterioplankton abundance fluctuated from 2.07×10 8 to 2.44×10 9 cells/dm 3 ,and culturable bacteria abundance ranged between 0.1×10 6 and 16.07×10 6 CFU/dm 3 .The Culturablity of heterotrophic bacteria ranged between 0.01% and 1.31%.The relationship between bacterial culturablity and the environmental factors was also studied in different sites of the Dapeng *** the bay,a significantly positive relationship was evidenced between bacterial culturability and temperature,meanswhile bacterial culturability was significantly inversely related with *** positive relationship was shown at the central bay while a negative relationship was observed at the bay mouth and there was no significant correlation at top bay between bacterial culturablity and chlorophyll a of sea water.A positive relationship was found between culturability and DIN all the bay,but DIP showed a significantly negative relationship with bacterial culturability only at the mouth of the bay.

选用中华硬蜱叮咬中肠抗原免疫接种组及初次叮咬组和佐剂对照组宿主并分别在不同时间(叮咬24、48、72h以及5、8d)对蜱中肠蛋白质、糖原及多糖进行了动态观察,并对消化细胞核DNA含量进行测定。结果显示:蜱叮咬中肠抗原免疫接种宿主后可使中肠上皮发生破坏,消化细胞破损,基膜可出现松散和断裂,消化细胞蛋白质含量较初次叮咬组和佐剂对照组宿主明显降低,而糖原及多糖含量基本不变。消化细胞DNA在叮咬无免疫力宿主(初次叮咬组和佐剂对照组)早期24h主要为2倍体,随着吸血量增加、细胞核倍体数加大,第5天达7.06倍体,而叮咬中肠抗原免疫接种组宿主后,消化细胞细胞核倍体数增加不明显,与叮咬无免疫力宿主有显著性差异(P>0.01),提示中华硬蜱叮咬中肠抗原免疫接种宿主后,可干扰中肠消化细胞蛋白质和核酸(DNA)的合成代谢,为进一步揭示抗蜱免疫机理提供了理论依据。

利用pSIREN-RetroQ载体构建了3个沉默多药耐药相关蛋白(MRP1)基因表达质粒pSIREN-siRNAs.并通过限制性内切酶酶切鉴定和DNA测序鉴定,将截断的MRP1和全长MRP1cDNA分别克隆到真核表达载体pEGFP-N2和pcDNA3.1中,产生了pEGFP-MRP1T和pcDNA-MRP1表达质粒.质粒pEGFP-MRP1T分别与3个pSIREN-siRNAs共转染HEK293细胞沉默MRP1T-GFP靶基因,pSIREN-siRNA1作为阴性对照.荧光显微镜下显示结果表明,与pSIREN-siRNA1相比,pSIREN-siRNA2和pSIREN-siRNA3产生的siRNA能够有效沉默MRP1T-GFP融合蛋白的表达.为了沉默全长MRP1基因的表达,pcDNA-MRP1分别与3个pSIREN-siRNAs共转染HEK293细胞.Western印迹和MTT分析表明,pSIREN-siRNA2和pSIREN-siRNA3能有效抑制190kDMRP1在HEK293细胞中的表达,而pSIREN-siRNA1则不能.pSIREN-siRNA2和pSIREN-siRNA3能逆转MRP1转染HEK293细胞产生的多药耐药性.RNA二级结构预测结果分析表明,siRNA1靶序列mRNA局部自由能热动力参数ΔG低于siRNA2和siRNA3靶序列mRNA局部自由能热动力参数,siRNA1的GC含量和Tm值高于siRNA2和siRNA3.这些数据提示,siRNA和局部靶结构可能影响siRNA对MRP1 mRNA表达的沉默作用.

本文探讨了电泳系统上槽泄露引起双向电泳凝胶蛋白点纵向拖尾问题.上槽泄露导致上槽SDS损耗,电流增大以及局部温度升高从而导致双向电泳凝胶高分子量区域蛋白点纵向拖尾.上槽泄露与上槽设计存在不足,使用者不适当操作以及上槽维护不良有关.对Ettan DALT six电泳系统而言,一旦出现上槽泄漏问题,建议更换改良后的新版上槽.电泳过程中应避免上槽泄露,SDS的不足或损耗,电流过大和电泳液温度过高.

A new kind of matrix-valued rational interpolants is recursively established by means of generalized Samelson inverse for matrices, with scalar numerator and matrix-valued denominator. In this respect, it is essentially different from that of the previous works [7, 9], where the matrix-valued rational interpolants is in Thiele-type continued fraction form with matrix-valued numerator and scalar denominator. For both univariate and bivariate cases, sufficient conditions for existence, characterisation and uniqueness in some sense are proved respectively, and an error formula for the univariate interpolating function is also given. The results obtained in this paper are illustrated with some numerical examples.
