Elevated fibroblast growth factor 23(FGF23)in X-linked hypophosphatemia(XLH)results in rickets and phosphate wasting,manifesting by severe bone and dental ***,a FGF23-neutralizing antibody,an alternative to conventional treatment(phosphorus and active vitamin D analogs),showed significant improvement in the long bone ***,we examined whether FGF23 antibody(FGF23-mAb)also improved the dentoalveolar features associated with ***-week-old male Hyp mice were injected weekly with 4 or 16 mg·kg−1 of FGF23-mAb for 2 months and compared to wild-type(WT)and vehicle(PBS)treated Hyp mice(n=3–7 mice).Micro-CT analyses showed that both doses of FGF23-mAb restored dentin/cementum volume and corrected the enlarged pulp volume in Hyp mice,the higher concentration resulting in a rescue similar to WT ***23-mAb treatment also improved alveolar bone volume fraction and mineral density compared to vehicle-treated *** revealed improved mineralization of the dentoalveolar tissues,with a decreased amount of osteoid,predentin and *** periodontal ligament attachment was also observed,evidenced by restoration of the acellular *** preclinical data were consistent with the retrospective analysis of two patients with XLH showing that burosumab treatment improved oral *** together,our data show that the dentoalveolar tissues are greatly improved by FGF23-mAb treatment,heralding its benefit in clinics for dental abnormalities.

Comparison of carbon-stock changes, eddycovariance carbon fluxes and model estimates in coastal Douglas-fir stands in British Columbia

Background： The global network of eddy-covariance （EC） flux-towers has improved the understanding of the terrestrial carbon （C） cycle, however, the network has a relatively limited spatial extent compared to forest inventory data and plots. Developing methods to use inventory-based and EC flux measurements together with modeling approaches is necessary evaluate forest C dynamics across broad spatial extents. Methods： Changes in C stock change （AC） were computed based on repeated measurements of forest inventory plots and compared with separate measurements of cumulative net ecosystem productivity （~NEP） over four years （2003 - 2006） for Douglas-fir （Pseudotsuga menzies# var menziesil} dominated regeneration （HDF00）, juvenile （HDF88 and HDF90） and near-rotation （DF49） aged stands （6, 18, 20, 57 years old in 2006, respectively） in coastal British Columbia. AC was determined from forest inventory plot data alone, and in a hybrid approach using inventory data along with litter fall data and published decay equations to determine the change in detrital pools. These AC-based estimates were then compared with Y_NEP measured at an eddy-covariance flux-tower （EC-flux） and modelled by the Carbon Budget Model - Canadian Forest Sector （CBM-CFS3） using historic forest inventory and forest disturbance data. Footprint analysis was used with remote sensing, soils and topography data to evaluate how well the inventory plots represented the range of stand conditions within the area of the flux-tower footprint and to spatially scale the plot data to the area of the EC-flux and model based estimates, Results： The closest convergence among methods was for the juvenile stands while the largest divergences were for the regenerating clearcut, followed by the near-rotation stand. Transjugular intrahepatic portosystemic shunt vs conservative treatment for recurrent ascites:A propensity score matched comparison

BACKGROUND Transjugular intrahepatic portosystemic shunt(TIPS) placement is an effective intervention for recurrent tense ascites. Some studies show an increased risk of acute on chronic liver failure(ACLF) associated with TIPS placement. It is not clear whether ACLF in this context is a consequence of TIPS or of the pre-existing liver *** To better understand the risks of TIPS in this challenging setting and to compare them with those of conservative *** Two hundred and fourteen patients undergoing their first TIPS placement for recurrent tense ascites at our tertiary-care center between 2007 and 2017 were identified(TIPS group). Three hundred and ninety-eight patients of the same time interval with liver cirrhosis and recurrent tense ascites not undergoing TIPS placement(No TIPS group) were analyzed as a control group. TIPS indication,diagnosis of recurrent ascites, further diagnoses and clinical findings were obtained from a database search and patient records. The in-hospital mortality and ACLF incidence of both groups were compared using 1:1 propensity score matching and multivariate logistic *** After propensity score matching, the TIPS and No TIPS groups were comparable in terms of laboratory values and ACLF incidence at hospital admission. There was no detectable difference in mortality(TIPS: 11/214, No TIPS 13/214). During the hospital stay, ACLF occurred more frequently in the TIPS group than in the No TIPS group(TIPS: 70/214, No TIPS: 57/214, P = 0.04). This effect was confined to patients with severely impaired liver function at hospital admission as indicated by a significant interaction term of Child score and TIPS placement in multivariate logistic regression. The TIPS group had a lower ACLF incidence at Child scores < 8 points and a higher ACLF incidence at ≥ 11 points. Recent Advances in Understanding Proteins Involved in Lipid Droplet Formation, Growth and Fusion

Lipid droplets （LDs） were once viewed as simple, inert lipid micelles. However, they are now known to be organelles with a rich proteome involved in a myriad of cellular processes. LDs are heterogeneous in nature with different sizes and compositions of phospholipids, neutral lipids and proteins. This review takes a focused look at the roles of proteins involved in the regulation of LD formation, expansion, and morphology. The related proteins are summarized such as the fat-specific protein （Fsp27）, fat storage-inducing trans- membrane （FIT） proteins, seipin and ADP-ribosylation factor 1-coat protein complex I （Arf-COPI）. Finally, we present important challenges in LD biology for a deeper understanding of this dynamic organelle to be achieved.

Development of a novel myconanomining approach for the recovery of agriculturally important elements from jarosite waste

In this study, an ecofriendly and economically viable waste management approach have been attempted towards the biosynthesis of agriculturally important nanoparticles from jarosite waste. Aspergillus terreus strain j4 isolated from jarosite（waste from Debari Zinc Smelter,Udaipur, India）, showed good leaching efficiency along with nanoparticles（NPs） formation under ambient conditions. Fourier-transform infrared spectroscopy（FT-IR） and transmission electron microscopy（TEM） confirmed the formation of NPs. Energy dispersive X-ray spectroscopy（EDX analysis） showed strong signals for zinc, iron, calcium and magnesium,with these materials being leached out. TEM analysis and high resolution transmission electron microscopy（HRTEM） showed semi-quasi spherical particles having average size of 10‐50 nm. Thus, a novel biomethodology was developed using fungal cell-free extract for bioleaching and subsequently nanoconversion of the waste materials into nanostructured form. These biosynthesized nanoparticles were tested for their efficacy on seed emergence activity of wheat（Triticum aestivum） seeds and showed enhanced growth at concentration of 20 ppm. These nanomaterials are expected to enhance plant growth properties and being targeted as additives in soil fertility and crop productivity enhancement.

Evaluation of a Wireless Solar Powered Personal Weather Station

We are evaluating dryland cotton production in martin County, Texas, measuring cotton lint yield per unit of rainfall. Our goal is to collect rainfall data per 250 - 400 ha. Upon selection of a rainfall gauge, we realized that the cost of using, for example, a tipping bucket-type rain gauge would be too expensive and thus searched for an alternative method. We selected an all-in-one commercially available weather station;hereafter, referred to as a Personal Weather Station (PWS) that is both wireless and solar powered. Our objective was to evaluate average measurements of rainfall obtained with the PWS and to compare these to measurements obtained with an automatic weather station (AWS). For this purpose, we installed four PWS deployed within 20 m of the Plant Stress and Water Conservation Meteorological Tower that was used as our AWS, located at USDA-ARS Cropping Systems Research Laboratory, Lubbock, TX. In addition, we measured and compared hourly average values of short-wave irradiance (Rg), air temperature (Tair) and relative humidity (RH), and wind speed (WS), and calculated values of dewpoint temperature (Tdew). This comparison was done over a 242-day period (1 October 2022-31 May 2023) and results indicated that there was no statistical difference in measurements of rainfall between the PWS and AWS. Hourly average values of Rg measured with the PWS and AWS agreed on clear days, but PWS measurements were higher on cloudy days. There was no statistical difference between PWS and AWS hourly average measurements of Tair, RH, and calculated Tdew. Hourly average measurements of Rg and WS were more variable. We concluded that the PWS we selected will provide adequate values of rainfall and other weather variables to meet our goal of evaluating dryland cotton lint yield per unit rainfall.

The resistance of wear protective coatings against oxidation is crucial for their use at high ***,three nanocomposite AlCr(Si)N coatings with a fixed Al/Cr atomic ratio of 70/30 and a varying Si-content of 0 at.%,2.5 at.% and 5 at.% were analyzed by differential scanning calorimetry,thermogravimetric analysis and X-ray in order to understand the oxidation behavior depending on their ***,a partially oxidized AlCrSiN coating with 5 at.%Si on a sapphire substrate was studied across the coating thickness by depth-resolved cross-sectional X-ray nanodiffraction and scanning trans-mission electron microscopy to investigate the elemental composition,morphology,phases and residual stress evolution of the oxide scale and the non-oxidized coating *** results reveal enhanced oxidation properties of the AlCr(Si)N coatings with increasing Si-content,as demonstrated by a retarded onset of oxidation to higher temperatures from 1100℃ for AlCrN to 1260℃ for the Si-containing coatings and a simultaneous deceleration of the oxidation *** annealing of the AlCrSiN sample with5 at.%Si at an extraordinary high temperature of 1400℃ for 60 min in ambient air,three zones developed throughout the coating strongly differing in their composition and structure:(i)a dense oxide layer comprising an Al-rich and a Cr-rich zone formed at the very top,followed by(ii)a fine-grained transition zone with incomplete oxidation and(iii)a non-oxidized zone with a porous *** varying elemental composition of these zones is furthermore accompanied by micro-structural variations and a complex residual stress development revealed by cross-sectional X-ray *** results provide a deeper understanding of the oxidation behavior of AlCr(Si)N coatings depending on their Si-content and the associated elemental,microstructural and residual stress evolution during high-temperature oxidation.

Striatins and STRIPAK complex partners in clinical outcomes of patients with breast cancer and responses to drug treatment

Objective: Striatins(STRNs) family, which contains three multi-domain scaffolding proteins, are cornerstones of the striatins interacting phosphatase and kinase(STRIPAK) complex. Although the role of the STRIPAK complex in cancer has become recognized in recent years, its clinical significance in breast cancer has not been fully ***: Using a freshly frozen breast cancer tissue cohort containing both cancerous and adjacent normal mammary tissues, we quantitatively evaluated the transcript-level expression of all members within the STRIPAK complex along with some key interacting and regulatory proteins of STRNs. The expression profile of each molecule and the integrated pattern of the complex members were assessed against the clinical-pathological factors of the patients. The Cancer Genome Atlas(TCGA) dataset was used to evaluate the breast cancer patients’ response to chemotherapies. Four human breast cancer cell lines, MDA-MB-231, MDA-MB-361, MCF-7, and SKBR-3, were subsequently adopted for in vitro ***: Here we found that high-level expressions of STRIP2, calmodulin, CCM3, MINK1 and SLMAP were respectively associated with shorter overall survival(OS) of patients. Although the similar pattern observed for STRN3, STRN4 and a contrary pattern observed for PPP2CA, PPP2CB and PPPR1A were not significant, the integrated expression profile of STRNs group and PPP2 group members constitutes a highly significant prognostic indicator for OS [P<0.001, hazard ratio(HR)=2.04, 95% confidence interval(95% CI), 1.36-3.07] and disease-free survival(DFS)(P=0.003, HR=1.40, 95% CI, 1.12-1.75). Reduced expression of STRN3 has an influence on the biological functions including adhesiveness and migration. In line with our clinical findings, the breast cancer cells responded to STRN3 knockdown with changes in their chemo-sensitivity, of which the response is also breast cancer subtype ***: Our results suggest a possible role of the STRIPAK complex
