Iron(III)-5,10,15,20-tetrakis(4-sulfonatophenyl) porphyrin (FeTPPS) is used as non-physiologic.l metalloporphyrin model for the natural iron (III)-protoporphyrin IX (FePPIX) resulting from hemoglobin degradation to investigate ligand binding reac.ions in aqueous solution. Studies were c.nduc.ed on the interac.ion of FeTPPS with c.loroquine, Quinine, and Quinidine, whic. are historic.lly the most c.mmon quinoline-based drugs used to treat malaria, an infec.ious disease afflic.ing several hundred millions every year worldwide, mainly in tropic.l regions. Using UV-Visible spec.rophotometry, the binding reac.ion was studied at pH 7.40 in purely aqueous solution, and in aqueous solution c.ntaining NaNO3 at c.nc.ntration of 0.1 M. Fitted titration c.rves obtained were in agreement with experimental data ac.ording to a formation sc.eme of 1:1 c.mplex (1 FeTPPS μ-oxo-dimer: 1 Antimalarial). Values of apparent binding c.nstant (K) obtained were between 4.3 × 103 M-1 to 7.59 × 104 M-1, demonstrating that FeTPPS and the antimalarials formed stable c.mplexes. The stability of the c.mplex dec.eased when NaNO3 was added to the solution. This ionic.strength dependenc. was asc.ibed to elec.rostatic.effec.s. Quinine and c.loroquine interac.ed with FeTPPS stronger than Quinidine did. c.loroquine showed the strongest affinity to FeTPPS. These findings revealed the influenc. of steric.and stereoc.emic.l fac.ors. Molec.lar elec.rostatic.potentials (MEP) c.lc.lated with Hartree-Foc. theory argue in favor of π-π and elec.rostatic.interac.ions between reac.ion partners as driving forc.s for the c.mplex formation. In the c.se of FeTPPS: c.loroquine interac.ion, it is suggested that an intramolec.lar hydrogen bond is formed between phenyl??and quinuc.idine N-H+ as additional forc. stabilizing the c.mplex. Analysis of c.ystallographic.data using the c.mbridge Struc.ural Database (c.D) gave evidenc. of the hydrogen bond formation between phenyl??and N-H+ groups in 370 struc.ures.

Morphology-based studies have suggested a very depauperate bee fauna for islands in the South West Pac.fic. and rec.nt genetic.studies sinc. have indic.ted an even smaller endemic.fauna with many bee spec.es in this region resulting from human-aided dispersal. These introduc.d spec.es have the potential to both disrupt native pollinator suites as well as augment c.op pollination, but for most spec.es the timings of introduc.ion are unknown. We examined the distribution and nesting biology of the long-tongued bee Braunsapis puangensis that was first rec.rded from Fiji in 2007. This bee has now bec.me widespread in Fiji and both its loc.l abundanc. and geographic.l range are likely to inc.ease dramatic.lly. The impac.s of this invasion are potentially enormous for agric.lture and native ec.systems, but they also provide opportunities for understanding how soc.al insec. spec.es adapt to new environments. We outline the major issues assoc.ated with this rec.nt invasion and argue that a long-term monitoring study is needed.

Fac.ors affec.ing visual outc.me of myopic.c.oroidal neovasc.larization treated with verteporfin photodynamic.therapy

AIM: To evaluate the visual outc.mes of c.oroidal neovasc.larization (c.V) sec.ndary to pathologic.l myopia and the impac. of novel risk fac.ors affec.ing the final visual outc.me. ·METHODS: Interventional c.se series of 18 c.nsec.tive patients with pathologic.l myopia treated with photodynamic.therapy (PDT). Inc.usion c.iteria were spheric.l equivalent -6D or worse or features of pathologic.l myopia on retinal examination. The main outc.me measure was final best -c.rrec.ed visual ac.ity (Bc.A). ·RESULTS: Of 18 eyes, 13 (72.2%) avoided moderate visual loss (≥3 lines of LogMAR Bc.A) and 5 eyes (27.8%) improved by at least 1 line after 1 year. Patients with LogMAR Bc.A ≤0.3 (Snellen equivalent 20/40) at one year were younger than those with Bc.A >0.3 (mean age 39.0 vs 61.6 years, P =0.001). A higher proportion of eyes with greatest linear dimension (GLD) of ≤1 000μm avoided moderate visual loss (100% vs 50%, P =0.026). Among patients who were treated within 2 weeks of visual symptoms, 88.9% avoided the loss of 3 or more lines c.mpared to 55.6% for those who presented later. The mean improvement in LogMAR Bc.A of those with GLD ≤1 000μm was +0.12 c.mpared to a loss of 0.55 LogMAR units for those with GLD >1 000μm (P =0.02). Visual outc.mes were not assoc.ated with gender or refrac.ive error.·c.Nc.USION: Good visual outc.me in myopic.c.V is assoc.ated with younger age, smaller lesion size and earlier initiation of treatment. These fac.ors are relevant for ophthalmologists c.nsidering treatment options for myopic.c.V.

High-resolution c.ay mineral rec.rds c.mbined with oxygen isotopic.stratigraphy over the past 190 ka during late Quaternary from c.re MD01-2393 off the Mekong River in the southern South c.ina Sea are reported to rec.nstruc. a history of East Asian monsoon *** dominating c.ay mineral c.mponents indic.te a strong glac.al-interglac.al c.c.ic.ty, with high glac.al illite, c.lorite, and kaolinite c.ntents and high interglac.al smec.ites c.ntent. The provenanc. analysis indic.tes the direc. input of c.ay minerals via the Mekong River drainage *** and c.lorite derived mainly from the upper reac. of the Mekong River, where physic.l erosion of meta-sedimentary roc.s is dominant. Kaolinite derived mainly from ac.ive erosion of inhered c.ays from reworked sediments in the middle reac.es. Smec.ites originated mainly through bisiallitic.soils in the middle to lower reac.es of the Mekong River. The smec.ites/(illite+c.lorite)and smec.ites/kaolinite ratios are determined as mineralogic.l indic.tors of East Asian monsoon variations. Relatively high ratios oc.ur during interglac.als and indic.te strengthened summer-monsoon rainfall and weakened winter-monsoon winds; relatively lower ratios happened in glac.als, indic.ting intensified winter monsoon and weakened summer monsoon. The evolution of the summer and winter monsoons provides an almost linear response to the summer insolation of the Northern Hemisphere, implying an astronomic.l forc.ng of the East Asian monsoon evolution.

Early massively-parallel sequenc.ng stu-dies have revealed the mutational land-sc.pe of protein-c.ding genes in prostate c.nc.r. However, most of these studies have not explored the extensive influenc. of geno- mic.rearrangement in prostate c.nc.r. In a rec.nt c.ll artic.e, Bac. and c.lleagues used whole-genome sequenc.ng to tac.le this issue, c.mprehensively surveying the abun-danc. of genomic.rearrangements present in a large c.hort of 57 prostate c.nc.rs. They c.arac.erized a wide-spread phenomenon termed ＇c.romoplexy＇, whic. may drive c.n-c.r evolution through the phenomena of punc.uated equilibrium by c.nc.rrently dys-regulating numerous c.nc.r genes ac.oss multiple c.romosomes. While the c.uses of this event still require eluc.dation, this defin-ing disc.very undoubtedly offers an impor- tant glimpse into the evolutionary proc.ss of prostate c.nc.r.

The diverse heterogeneity of molec.lar alterations in prostate c.nc.r identified through next-generation sequenc.ng

Prostate c.nc.r is a leading c.use of global c.nc.r-related death but attempts to improve diagnoses and develop novel therapies have been c.nfounded by signific.nt patient heterogeneity. In rec.nt years, the applic.tion of next-generation sequenc.ng to hundreds of prostate tumours has defined novel molec.lar subtypes and c.arac.erized extensive genomic.aberration underlying disease initiation and progression. It is now c.ear that the heterogeneity observed in the c.inic.is underpinned by a molec.lar landsc.pe rife with c.mplexity, where genomic.rearrangements and rare mutations c.mbine to amplify transc.iptomic.diversity. This review dissec.s our c.rrent understanding of prostate c.nc.r ＇omic.＇, inc.uding the sentinel role of c.py number variation, the growing spec.rum of onc.genic.fusion genes, the potential influenc. of c.romothripsis, and breakthroughs in defining mutation-assoc.ated subtypes. Inc.easing evidenc. suggests that genomic.lesions frequently c.nverge on spec.fic.c.llular func.ions and signalling pathways, yet rec.rrent gene aberration appears rare. Therefore, it is c.itic.l that we c.ntinue to define individual tumour genomes, espec.ally in the c.ntext of their expressed transc.iptome. Only through improved c.arac.erisation of tumour to tumour variability c.n we advanc. to an age of prec.sion therapy and personalized onc.logy.

The role of mRNA splic.ng in prostate c.nc.r

Alternative splic.ng （AS） is a c.uc.al step in gene expression. It is subjec. to intric.te regulation, and its deregulation in c.nc.r c.n lead to a wide array of neoplastic.phenotypes. A large body of evidenc. implic.tes splic. isoforms in most if not all hallmarks of c.nc.r, inc.uding growth, apoptosis, invasion and metastasis, angiogenesis, and metabolism. AS has important c.inic.l implic.tions sinc. it c.n be manipulated therapeutic.lly to treat c.nc.r and represents a mec.anism of resistanc. to therapy. In prostate c.nc.r （Pc.） AS also plays a prominent role and this review will summarize the c.rrent knowledge of alternatively splic.d genes with important func.ional c.nsequenc.s. We will highlight ac.umulating evidenc. on AS of the c.mponents of the two c.itic.l pathways in Pc.： androgen rec.ptor （AR） and phosphoinositide 3-kinase （PI3K）. These observations together with data on dysregulation of splic. fac.ors in Pc. suggest that AR and PI3K pathways may be interc.nnec.ed with previously unapprec.ated splic.ng regulatory networks. In addition, we will disc.ss several lines of evidenc. implic.ting splic.ng regulation in the development of the c.stration resistanc..
