A fast,robust pipeline for strain mapping of crystalline materials is important for many technological *** electron nanodiffraction allows us to calculate strain maps with high accuracy and spatial resolutions,but this technique is limited when the electron beam undergoes multiple ***-learning methods have the potential to invert these complex signals,but require a large number of training *** implement a Fourier space,complex-valued deep-neural network,FcU-Net,to invert highly nonlinear electron diffraction patterns into the corresponding quantitative structure factor ***-Net was trained using over 200,000 unique simulated dynamical diffraction patterns from different combinations of crystal structures,orientations,thicknesses,and microscope parameters,which are augmented with experimental *** evaluated FcU-Net against simulated and experimental datasets,where it substantially outperforms conventional analysis *** code,models,and training library are open-source and may be adapted to different diffraction measurement problems.

雷州黑鸭是一种蛋用型鸭种,具有较好的产蛋性能,其广泛分布于中国雷州半岛。前期对雷州黑鸭卵巢转录组学分析结果发现,ESR2基因显著富集在与繁殖性能相关的生殖激素调控通路及雌激素信号通路中,提示ESR2可能对雷州黑鸭的繁殖性能具有重要调控作用。ESR2基因属于核受体家族的一员,其主要在卵巢和颗粒细胞中表达,具有调节卵巢卵泡发育和排卵的作用。卵巢和输卵管是产蛋的重要器官,其发育状况直接影响家禽的产蛋性能。因此,本研究旨在评估ESR2基因的多态性及其与产蛋性状的相关关系,通过检测ESR2基因在HPG轴、输卵管和非生殖器官中的m RNA的表达水平及探究利用免疫组织化学分析方法验证ESR2在雷州黑鸭卵巢组织中特异性细胞定位,以此来探讨ESR2在雷州黑鸭卵泡发育中的作用及功能。本研究提取101只雷州黑鸭血液样本的基因组DNA来分析ESR2基因的单核苷酸多态性（SNPs）与雷州黑鸭产蛋性能的关联。10对ESR2基因引物的PcR产物测序结果发现了引物2、5、6和7共有23个SNPs位点,其与产蛋性能的相关性分析结果表明g.56805646 T＞c位点和exon 3-20 G＞A位点与开产蛋重显著相关（P＜0.05）,其中cT基因型和AG基因型的开产蛋重显著高于其它基因型。g.56805646 T＞c和exon 3-20 G＞A两个位点间进行单倍型分析,结果表明两位点间单倍型与开产蛋重显著相关（P＜0.05）,其中H3H4单倍型个体的开产蛋重显著高于其他单倍型个体（P＜0.05）。在引物6上发现的SNPs位点的单倍型与产蛋性能关联分析结果表明,其相关单倍型与蛋重显著相关（P＜0.05）,其中在H1H2单倍型的个体的开产蛋重显著高于其他个体（P＜0.05）。同样,在引物7上发现的SNPs位点的单倍型与开产日龄和开产蛋重显著相关（P＜0.05）,其中在H2H3单倍型的个体的蛋重显著高于其它单倍型个体（P＜0.05）。为了比较和探讨ESR2基因在雷州黑鸭产蛋期和非产蛋期间的m RNA表达水平,收集雷州黑鸭的下丘脑-垂体-卵巢轴组织、输卵管组织（漏斗部、膨大部、峡部和子宫）和非生殖器官（心、肝、脾、肺、肾、胸肌和腿部肌肉）组织。对于HPG轴,qRT/PcR结果显示在产蛋期和非产蛋期两组鸭的卵巢中ESR2的m RNA相对表达量显著高于在下丘脑和垂体中的表达（P＜0.05）。同时,对于HPG轴上组织,产蛋期鸭的ESR2的表达量明显高于非产蛋期雷州黑鸭（P＜0.05）。在输卵管中,qRT/PcR结果显示产蛋鸭和非产蛋鸭的漏斗部和膨大部的ESR2的m RNA相对表达量均显著升高（P＜0.05）,且产蛋鸭输卵管中ESR2 m RNA的表达量显著高于非产蛋鸭（P＜0.01,0.05）。在各种非生殖器官组织中,qRT/PcR结果显示产蛋鸭和非产蛋鸭脾脏中ESR2的m RNA表达水平显著升高（P＜0.05）,ESR2的m RNA表达水平在产蛋鸭的所有非生殖器官组织中的表达均高于非产蛋鸭（P＜0.01,0.05）。为了阐明卵泡中ESR2的表达模式和细胞特异性定位,本研究收集了雷州黑鸭不同等级大小的卵泡。卵泡分为七个等级:小白卵泡（SWF）、大白卵泡（LWF）、小黄卵泡（SYF）、大黄卵泡（LYF）、卵泡5（F5）、卵泡2（F2）和卵泡1（F1）。qRT/PcR结果显示ESR2的m RNA相对表达量在所有卵泡中均有表达,与其他卵泡相比,F1等级的卵泡中ESR2m RNA相对表达量相对最高（P＜0.05）。使用免疫组织化学分析方法验证ESR2在细胞中特异性定位的结果表明ESR2分在所有检测卵泡的颗粒细胞和膜细胞中均有分布;ESR2在SWF、SYF、LYF、F2和F1的各级卵泡颗粒细胞中的表达水平显著高于其膜细胞的表达水平（P＜0.05）;同时,ESR2在LYF颗粒细胞中的表达水平显著高于在其他等级卵泡颗粒细胞中的表达水平。综上所述,ESR2 SNPs位点与雷州黑鸭产蛋性能的关联分析结果显示g.56805646 T＞c和exon 3-20 G＞A位点与开产蛋重显著相关。单倍型分析结果显示,其单倍型与开产日龄（FEA）和开产蛋重（EW）显著相关。ESR2在雷州黑鸭的卵巢中高表达,主要定位于其颗粒细胞中。这些结果为深入研究ESR2基因的相关生物学功能及其在细胞水平上的应用提供了理论基础。此外,该研究表明ESR2基因的SNPs位点与产蛋性能之间存在较强的关联性,可作为筛选高产优质性能雷州黑鸭的相关分子标记。

OBJEcTIVE Wide spread abuse of synthetic cathinones found in bath salts preparations has resulted in regulation of some cathinones *** skirt these laws by altering the chemical structures of first-generation cathinones(ie,MDPV,methylone,and mephedrone),resulting in second-generation cathinones(eg,α-PVP,α-PPP,MDPPP,and MDPBP).Although MDPV is a more effective reinforcer than cocaine,little is known about the reinforcing effectiveness of secondgeneration *** test the hypothesis that synthetic cathinones with higher selectivity for DAT relative to SERT are more effective *** Monoamine transporter inhibition was determined using synaptosomes prepared from rat *** relative reinforcing effectiveness of intravenously self-administered MDPV,MDPBP,MDPPP,α-PVP,α-PPP,and cocaine were directly compared through evaluations of (1)dose-response curves under a progressive ratio(PR)schedule of reinforcement and (2)demand curves obtained for each drug in male Sprague-Dawley *** Rank order selectivity for DAT/SERT wasα-PVP>MDPV>α-PPP≈MDPBP>MDPPP>*** of the maximum number of infusions obtained under a PR schedule of reinforcement(α-PVP>MDPV>α-PPP>MDPBP≈MDPPP>cocaine)and the essential value obtained for each drug in demand analyses(α-PVP>MDPV>α-PPP≈MDPBP≈MDPPP>cocaine)suggest relative reinforcing effectiveness is related to DAT/SERT *** These data provide evidence that DAT/SERT selectivity accounts for select synthetic cathinones functioning as more effective reinforcers than cocaine and may predict the abuse-related effects of novel synthetic cathinones in humans.

Objective The aim of this study is to characterize long-term morbidities of oesophageal atresia(OA)with or without tracheoesophageal fistula(TOF).Methods Infants born with OA/TOF from 2000 to 2016 in Western Australia were included for *** were categorized into high-risk and low-risk groups based on the presence of one or more perioperative risk factors[low birth weight,vertebraldefects,anal atresia,cardiac defects,TOF,renalanomalies,limb abnormalities(VAcTERL),anastomotic leak,long gap OA,and failure to establish oral feeds within the first month]identified by a previous canadian *** of morbidities in infants with perioperative risk factors was *** Of 102 patients,88(86%)had OA with distal TOF(type c).The most common morbidities in our cohort were anastomotic oesophageal strictures(AS)(n=53,52%),tracheomalacia(n=48,47%),gastroesophageal reflux disease(GORD)(n=42,41%)and recurrent respiratory tract infections(n=40,39%).Presence of GORD(30/59 vs 12/43,p=0.04)and median frequency of AS dilatations(8 vs 3,n=59,p=0.03)were greater in the high-risk *** study further confirmed that inability to be fed orally within the first month was associated with high *** Gastrointestinal and respiratory morbidities remain high in OA/TOF regardless of perioperative risk *** to be fed orally within the first month is a predictor of poor outcomes with high frequency of gastrointestinal and respiratory comorbidities.
