Recombineering is an essential tool for molecular biologists,allowing for the facile and efficient manipulation of bacterial genomes directly in cells without the need for costly and laborious in vitro manipulations involving restriction *** main workhorses behind recombineering are bacteriophage proteins that promote the single-strand annealing(SSA)homologous recombination pathway to repair double-stranded DNA *** there have been several reviews examining recombineering methods and applications,comparatively few have focused on the mechanisms of the proteins that are the key players in the SSA pathway:a 5′→3′exonuclease and a single-strand annealing protein(SSAP or“annealase”).This review dives into the structures and functions of the two SSA recombination systems that were the first to be developed for recombineering in ***:the RecET system from *** Rac prophage and the𝜆Red system from bacteriophageλ.By comparing the structures of the RecT and Red𝛽annealases,and the RecE and𝜆Exo exonucleases,we provide new insights into how the structures of these proteins dictate their *** the sequence conservation of the𝜆λExo and RecE exonucleases gives more profound insights into their critical functional ***,as recombineering accelerates and evolves in the laboratory,a better understanding of the mechanisms of the proteins behind this powerful technique will drive the development of improved and expanded capabilities in the future.

A comparison of transmittance properties between ZnO∶Al films for transparent conductors for solar cells deposited by sputtering of AZO and cosputtering of AZO/ZnO

Aluminum-doped zinc oxide (AZO) thin films were deposited on sapphire (002) and glass substrates by two different sputtering techniques radio frequency magnetron cosputtering of AZO and ZnO targets and sputtering of an AZO target. The dependence of the photoluminescence (PL) and transmittance properties of the AZO films deposited by cosputtering and sputtering on the AZO/ZnO target power ratio, R and the O2/Ar flow ratio, r were investigated, respectively. Only a deep level emission peak appears in the PL spectra of cosputtered AZO films whereas both UV emission and deep level emission peaks are observed in the PL spectra of sputtered AZO films. The absorption edges in the transmittance spectra of the AZO films shift to the lower wavelength region as R and r increase. Effects of crystallinity, surface roughness, PL on the transmittance of the AZO films were also explained using the X-ray diffraction (XRD), atomic force microscopy (AFM), and PL analysis results.

A recent study by Adams and colleagues published in Nature unveiled more than one hundred genes that are involved in micronucleus formation,1 which is a biomarker for genomic instability and associated with aging,cancer,and other disorders.2 As an outstanding achievement,the authors identified DSCC1(DNA Replication And Sister Chromatid Cohesion 1)as a critical gene protecting against early developmental defects,genomic instability,reduced proliferative capacity,cohesinopathy-related phenotypes,and tumor development in vivo.

Necrotic enteritis and antibiotic-free production of broiler chickens:Challenges in testing and using alternative products

The global trend towards raising broiler chickens without the use of in-feed antibiotics(IFAs) means that there is an ongoing need to develop alternative treatments capable of delivering the benefits that IFAs previously provided. IFAs supported the productivity performance of chickens and played a key role in maintaining their health. Necrotic enteritis(NE) is an important disease of broilers that affects health,productivity, and welfare, and was previously well controlled by IFAs. However, with the reduction in IFA use, NE is resurgent in some countries. Vaccines and various feed additives, including pre-, pro-, and postbiotics, phytobiotics, fatty acids, and phage therapies have been introduced as alternative methods of NE control. While some of these feed additives have specific activity against the NE pathogen, Clostridium perfringens, most have the more general goal of reinforcing gut health. Extensive reviews of the effects of many of these feed additives on gut health have been published recently. Hence, rather than cover previously well reviewed areas of research this review focuses on the challenges and pitfalls in undertaking experimental assessment of alternative NE treatments and translating laboratory research to real world commercial production settings. The review is based on the author's particular experience,reading, thoughts, and analysis of the available information and inevitably presents a particular understanding that is likely to be at odds with others thinking on these issues. It is put forward to stimulate thinking and discussion on the issues covered.

Are hydrides under high-pressure-high-temperature superconductors?

Contrary to the current consensus, I argue that the existing evidence for high-temperature superconductivity in hydrides under high pressure is not compelling. I suggest that the focus of the field should urgently shift to establish unequivocally experimentally whether or not superconductivity in pressurized hydrides exists, instead of continuing to search for new materials that might show elusive signals of unproven superconductivity at ever higher temperatures. The implications of a negative finding for the theoretical understanding of superconductivity are discussed.

TRANSIENT FLOWS AND PRESSURE WAVES IN PIPES

Transient laminar flows and pressure-wave propagation in pipes, commonly called as water hammer, have been analyzed. A pressure-wave equation and a linearized velocity equation were derived from the equations of mass and momentum conservation. The waveform distortion due to viscous dissipation and elastic expansion of pipe wall was characterized by a dimensionless transmission number. The damping coefficients of pressure waves were found to be related to the roots of Bessel function. An exact solution of pressure-wave equation was obtained numerically. The relationship between the distortion of a traveling wave and transmission number was studied. The problem was also calculated with a general-purpose computer code, COMMIX, which solves exact mass conservation equation and Navier-Stokes equations. The computational results of the COMMIX code agreed well with the analytical solutions.

Provenance and tectonic setting of sandstones of the Lomas Coloradas Formation, Cabullona group, Sonora, México: Constraints on petrography and geochemistry

The sedimentary rocks of the Cabullona group are well exposed in the Cabullona Basin of northeastern Sonora, Mexico, which represent syntectonic sedimentation associated with Laramide-style deformation during the Campanian-early Maastrichtian. The sandstones of the Lomas Coloradas Formation deposited at the upper part of the Cabullona group. Petrography and geochemical(major, trace, and rare earth elements) studies were carried out on the sandstones to interpret the palaeoweathering, provenance,and tectonic setting of the Lomas Coloradas Formation sandstones. Most of the sandstone modal analysis data plotted in the QFL and Qm FLt ternary diagrams fall into the fields of dissected arcs and recycled orogens,respectively. geochemically the sandstones are classified as litharenites while few samples plot into the arkosic field. The chondrite-normalized REE diagrams for sandstones have LREE-enriched, relatively flat HREE patterns with negative Eu anomalies. CIA values and the A-CN-K diagram indicate a low intensity of chemical weathering in the source area. The tectonic setting diagram suggests both arc and rift settings for the studied sandstones. The bivariate and ternary diagrams and elemental ratios suggest a felsic source for the Lomas Coloradas Formation. The average REE patterns of the studied sandstones are comparable with REE patterns of the granite from the Caborca block, the rhyolite and granite of the Tarahumara Formation, and the Mesozoic felsic volcanic rocks. The modeled mixture of 40% Proterozoic granite, 30% Triassic arc, 10% jurassic arc, and 20% Laramide arc represents the probable source rocks for the Lomas Coloradas Formation.

Analysis of mid-spatial frequency wavefront distortions from a liquid-cooled flash-lamp pumped Nd:phosphate laser amplifier

Mid-spatial frequency wavefront deformation can be deleterious for the operation of high-energy laser systems. When fluid cooled high-repetition-rate amplifiers are used, the coolant flow is likely to induce such detrimental mid-spatial frequency wavefront deformations. Here, we describe the design and performance of a 90 mm × 90 mm aperture, liquid-cooled Nd:phosphate split-slab laser amplifier pumped by flash-lamps. The performance of the system is evaluated in terms of wavefront aberration and gain at repetition rates down to 1 shot per minute. The results show that this single cooled split-slab system exhibits low wavefront distortions in the medium to large period range, compatible with a focus on target, and despite the use of liquid coolant traversed by both pump and amplified wavelengths. This makes it a potential candidate for applications in large high-energy laser facilities.

Laboratory evaluation of toxicity of selected insecticides against egg and larval stages of cotton pink bollworm, Pectinophora gossypiella (Saunders) (Lepidoptera: gelechiidae)

Background The cryptic nature of pink bollworm Pectinophora gossypiella(Saunders)larvae enables its reduced vul-nerability to insecticidal ***,the development of resistance against Bacillus thuringiensis(Bt)toxins posed a serious threat to transgenic cotton *** necessitated determining the critical timing of spray applications on the control *** study assessed the influence of egg age(freshly laid ***-day-old)and the loca-tion of larvae(directly exposed to the insecticide residues on the boll rind *** inside the bolls)on insecticide control *** The results revealed a significant decrease in the ovicidal activity of tested insecticides with an increase in the age of eggs from one day old to three days old(paired t-test,P80%mortality)than the larvae exposed after they had bur-rowed inside the bolls(gy were more pronounced in the experiment on pre-larval release insecticide treatment compared with insecticide treatment given post-larval release and entry inside the *** Egg age influences the insecticide susceptibility,as does the larval location,directly exposed vs bur-rowed inside the *** eggs and the larvae that had burrowed inside the green bolls of cotton were relatively less susceptible to the insecticide *** toxic effects of insecticides on egg and larval stages were primar-ily *** findings are significant for devising a comprehensive strategy for pink bollworm management on a sustainable basis.
