HCAR2 and the activatio. o. thes. recepto.s.pro.o.es.an anti-inflammato.y res.o.s. which may be related with immuno.2}ppres.io. o.s.rved in trans.tio. dairy cattle. The o.jective o. this.s.udy was.to.identify and tes. antago.is.s.fo. HCAR2 in bo.ine immune cells.cultured with *** We o.s.rved expres.io. o. HCAR2 at the pro.ein level within lympho.ytes. mo.o.ytes. and granulo.ytes. The pro.o.tio. o. cells.expres.ing HCAR2 tended to.be greater in mid-lactatio. compared to.early lactatio. cows.the increas. was.a res.lt o. increas.d pro.o.tio. o. T and B cells.expres.ing HCAR2. s.imulatio. o. HCAR2 with niacin o. BHBA pro.o.ed Ca^(2+) mo.ilizatio. in neutro.hils.and mo.o.uclear cells. Mo.o.uclear cells.treated with BHBA had diminis.ed intracellular Ca^(2+) res.o.s.s.when HCAR2 was.k.o.k.d do.n by s. RNA s.lencing, indicating Ca^(2+) mo.ilizatio. was.mediated by HCAR2 s.gnaling. Two.candidate antago.is.s.fo. HCAR2, s.nthes.zed fro. niacin(NA-1 and NA-5), were tes.ed;mo.o.ytes.and neutro.hils.pre-treated with NA-1 and NA-5 had reduced Ca^(2+) mo.ilizatio. after incubatio. with BHBA.
