Irritable bowel syndrome (IBS) is a lifelong condition with a high prevalence among children and adults. As the diet is a frequent factor that triggers the symptoms, it has been assumed that by avoiding the consumption of fermentable oligosaccharides, disaccharides, monosaccharides and polyols (FODMAP), the symptoms might be improved. Therefore, in the past decade, low FODMAP diet has been intensively investigated in the management of IBS. The capacity of FODMAPs to trigger the symptoms in patients with IBS was related to the stimulation of mechanoreceptors in the small and large intestine. This stimulation appears as a response to a combination of increased luminal water (the osmotic effect) and the release of gases (carbon dioxide and hydrogen) due to the fermentation of oligosaccharides and malabsorption of fructose, lactose and polyols. Numerous studies have been published regarding the efficacy of a low FODMAP diet compared to a traditional diet in releasing the IBS symptoms in adults, but there are only a few studies in the juvenile population. The aim of this review is to analyze the current data on both low FODMAP diet in children with IBS and the effects on their nutritional status and physiological development, given the fact that it is a restrictive diet.

High strength threaded fasteners are widely used in the aircraft industry, and service experience shows that for structures where shear loading of the joints is significant, like skin splices, fuselage joints or spar caps-web attachments, more cracks are initiated and grow from the edges of the fastener holes than from features like fillets radii and corners or from large access holes. The main causes of this cracking are the stress concentrations introduced by the fastener holes and by the threaded fasteners themselves, with the most common damage site being at the edge of the fastener holes. Intuitively, it is easy to visualize that after the crack initiation, during the growth stages, some of the load transferred initially by the fastener at the cracked hole will decrease, and it will be shed to the adjacent fasteners that will carry higher loads than in uncracked condition. Using currently available computer software, the method presented in this paper provides a relatively quick and quantitatively defined solution to account for the effects of crack length on the fastener loads transfer, and on the far field and bypass loads at each fastener adjacent to the crack. At each location, these variations are determined from the 3-dimensional distribution of stresses in the joint, and accounting for secondary bending effects and fastener tilt. Two cases of a typical skins lap splice with eight fasteners in a two rows configuration loaded in tension are presented and discussed, one representative for wing or fuselage skins configurations, and the second case representative for cost effective laboratory testing. Each case presents five cracking scenarios, with the cracks growing from approx. 0.03 inch to either the free edge, next hole or both simultaneously.

The design and sizing of new mechanical components which are intended to operate under cyclic loads often require an acceptable level of confidence that the components will meet pre-defined fatigue strength objectives for crack initiation. For loads with multiple amplitudes and mean values, models based on Palmgren-Miner’s linear cumulative damage hypothesis and on multiple S/N curves (stress-no. cycles) are widely used in estimations for the duration of crack initiation. In this paper a procedure for generating S/N curves for multiple stress ratios by interpolation or extrapolation from the data available for two such curves is proposed. At any number of cycles, the stress for crack initiation is calculated from the far field macroscopic stresses of the known curves using Dang Van fatigue criterion and microscopic stresses evaluated at the grain level. An algorithm is presented for uniaxial loading and results verifications against curves established from laboratory tests with elastic and plastic stresses are shown and discussed for notched and un-notched specimens.

Carbonic anhydrases(CA, EC 4.***.1.1) catalyze the interconversion bewteen carbon dioxide and bicarbonate with generation of protons. The carbonic anhydrase isozyme Ⅸ(CA Ⅸ) is highly overexpresed in hypoxic tumors and shows very restricted expression in normal tissues. CA Ⅸ is a dimeric protein possessing very high catalytic activity for the hydration of carbon dioxide to protons and bicarbonate. Its quaternary structure is unique among members of this family of enzymes, allowing for structure-based drug design campaigns of selective inhibitors. Inhibition of CA Ⅸ with sulfonamide and/or coumarin inhibitors was recently shown to lead to a potent retardation for the growth of both primary tumors and metastases. Some fluorescent sulfonamides were shown to accumulate only in hypoxic tumor cells overexpressing CA Ⅸ, and might be used as diagnostic tools for imaging of hypoxic cancers. Sulfonamide inhibitors were also more effective in inhibiting the growth of the primary tumors when associated with irrdiation. CA Ⅸ is thus both a diagnostic and therapeutic validated target for the management of hypoxic tumors normally non-responsive to classical chemio- and radiotherapy.

Intracellular protein routing is mediated by vesicular transport which is tightly regulated in eukaryotes. The protein and lipid homeostasis depends on coordinated delivery of de novo synthesized or recycled cargoes to the plasma membrane by exocytosis and their subsequent removal by rerouting them for recycling or degradation. Here, we report the characterization of protein affected trafficking 3 （pat3） mutant that we identified by an epifluorescence-based for- ward genetic screen for mutants defective in subcellular distribution of Arabidopsis auxin transporter PIN1-GFR While pat3 displays largely normal plant morphology and development in nutrient-rich conditions, it shows strong ectopic intracellular accumulations of different plasma membrane cargoes in structures that resemble prevacuolar compart- ments （PVC） with an aberrant morphology. Genetic mapping revealed that pat3 is defective in vacuolar protein sorting 35A （VPS35A）, a putative subunit of the retromer complex that mediates retrograde trafficking between the PVC and trans-Golgi network. Similarly, a mutant defective in another retromer subunit, vps29, shows comparable subcellular defects in PVC morphology and protein accumulation. Thus, our data provide evidence that the retromer components VPS35A and VPS29 are essential for normal PVC morphology and normal trafficking of plasma membrane proteins in plants. In addition, we show that, out of the three VPS35 retromer subunits present in Arabidopsis thaliana genome, the VPS35 homolog A plays a prevailing role in trafficking to the lyric vacuole, presenting another level of complexity in the retromer-dependent vacuolar sorting.

BACKGROUND Hydatid liver disease remains an important issue in endemic areas,which may require immediate *** laparoscopic surgery is on the rise,the presence of certain complications may require conversion to the open *** To compare the results of laparoscopic treatment and the open approach in the context of a 12-year single institution experience,and to perform a further comparison between results from the current study and those from a previous *** Between January 2009 and December 2020,247 patients underwent surgery for hydatic disease of the liver in our *** the 247 patients,70 underwent laparoscopic treatment.A retrospective analysis between the two groups was performed,as well as a comparison between current and previous laparoscopic experience(1999-2008).RESULTS There were statistically significant differences between the laparoscopic and open approaches regarding the cyst dimension,location,and presence of cystobiliary *** were no intraoperative complications in the laparoscopic *** cutoff value for the cyst size regarding the presence of cystobiliary fistula was 6.85 cm(P=0.001).CONCLUSION Laparoscopic surgery still plays an important role in the treatment of hydatid disease of the liver,with an increase in its usage over the course of years that has shown benefits regarding the postoperative recovery with a decreased rate of intraoperative *** experienced surgeons can perform laparoscopic surgery in the most difficult conditions,there are some selection criteria that need to be maintained for higher quality results.

An Evaluation of Nearshore Sediment Data after the Deepwater Horizon Blowout

The deepwater horizon blowout led to the release of 4.9 million barrels of crude oil into the Gulf of Mexico （GOM）. Here the paper analyzes available sediment datasets to determine whether changes in petrogenic contaminants are evident in GOM sediment following the blowout. Locations sampled by EPA （Environmental Protection Agency）, British Petroleum （BP） and NOAA （National Oceanic and Atmospheric Administration） were reduced and grouped into 46 similar locations and analyzed. Eleven groups suggested an increase in PAIl （polycyclic aromatic hydrocarbon） and nickel, one group showed a statistically significant increase in Phenanthrene. Four locations were analyzed for time trends and differences between initial and peak concentrations of oil range organics （ORO）, diesel range organics （DRO）, vanadium and nickel. One location had significant increases in ORO, DRO and vanadium and a suggestive increase in nickel. Correlations between ORO/DRO, ORO/V, ORO/Ni, V/Ni were computed （RE= 0.85, 0.67, 0.64, 0.92, respectively）. Overall, the analyses suggest that future monitoring should employ a sampling strategy that coordinates response sampling to previously sampled locations, such that baseline datasets can be used in detection of event associated contamination.

The Influence of Gravity on Single Crystallization in Large Volume Drops

The chemical, physical, and biological properties of more than two millions of proteins which follow to be synthesized by Pharmaceutical Industry, can be anticipated (by using their XRD diffrac-tograms) if they will be grown from aqueous drops as high quality, large volume single-crystals. This is not a simple task and usually the growing process is seen as art rather than a science. The growing is expensive, time consuming, and finally an amorphous aggregate may result instead one single-crystal. In this article, we show for the first time how one single crystal can be grown in large volume hanging drops through their fast evaporation. The single nucleation is determined by choosing the proper sense of gravitational force relative to the drop triple line contact. In a special configuration, single-crystals of glycine and threonine were rapidly grown.
