Global prevalence of non-alcoholic fatty liver disease(NAFLD)and of NAFLD-hepatocellular carcinoma(HCC)is estimated to grow in the next *** burden of NAFLD and the evidence that NAFLD-HCC arises also in noncirrhotic patients,explain the urgent need of a better characterization of the molecular mechanisms involved in NAFLD *** and diabetes cause a chronic inflammatory state which favors changes in serum cytokines and adipokines,an increase in oxidative stress,DNA damage,and the activation of multiple signaling pathways involved in cell ***,a role in promoting NAFLD-HCC has been highlighted in the innate and adaptive immune system,dysbiosis,and alterations in bile acids *** dietary,genetic,or combined mouse models have been used to study nonalcoholic steatohepatitis(NASH)development and its progression to HCC,but models that fully recapitulate the biological and prognostic features of human NASH are still *** humans,four single nucleotide polymorphisms(PNPLA3,Tm6SF2,GCKR,and mBOAT7)have been linked to the development of both NASH and HCC in cirrhotic and non-cirrhotic patients,whereas HSD17B13 polymorphism has a protective *** addition,higher rates of somatic ACVR2A mutations and a novel mutational signature have been recently discovered in NASH-HCC *** knowledge of the molecular pathogenesis of NAFLD-HCC will be helpful to personalized screening programs and allow for primary and secondary chemopreventive treatments for NAFLD patients who are more likely to progress to HCC.

Aims Light requirements for cactus seed germination have been considered to be associated with their adult plant height and seed mass,but this has not been thoroughly studied for other succulent *** order to understand seed photosensitivity from desert species belonging to Asparagaceae(subfamily Agavoideae)and Cactaceae,we performed a germination experiment with and without light for 12 species and 2 varieties from 1 species from the Southern Chihuahuan *** also determined if adult growth is totally determined by seedling‘growth form’in *** We performed a germination experiment using light and darkness for 13 species from Southern Chihuahuan Desert:10 rosette species(Asparagaceae),as well as 1 globose,1 columnar and 2 varieties from 1 depressed-globose species(Cactaceae).The response variables were seed germination percentage and relative light germination(RLG).In addition,in order to determine if adult-globose cacti could have cylindrical seedlings,we calculated the shape index(height/width ratio)for Coryphanta clavata and mammillaria *** Findings All species were considered neutral *** species had similar seed germination in both light and dark conditions,and three taxa(*** and the two varieties of Ferocactus latispinus)showed higher germination with light than without *** salmiana,*** and the two varieties of *** had higher RLG than the other *** mass was an important factor because with higher seed mass there was lower dependence to *** findings support the hypothesis that small seed mass and light requirements have coevolved as an adaptation to ensure *** adult-globose cactus species,***,and one adult-columnar species,***,had small seeds and neutral *** from these two species exposed to light were cylindrical and those under darkness conditions were *** seeds from this species are able to germinate in

AIm:To investigate the effectiveness of Clostridium novyi(***)-NT spores for the treatment of established subcutaneous pancreatic tumor in the syngeneic,immunocompetent Panc02/C57Bl/6 model. mETHODS:***-NT spores were applied intravenously to animals carrying established pancreatic tumors of three different *** immune responses in peripheral blood and spleen were examined by flow ***,cytotoxic activity of lymphocytes against syngeneic tumor targets was analyzed. RESULTS:Application of spores identified,that(1) small tumors(mm 3 )were completely unaffected (n=10);(2)very large tumors(>450 mm3)responded with substantial necrosis followed by shrinkage and significant lethality most likely due to tumor lysis syndrome (n=6);and(3)an optimal treatment window exists for tumors of approximately 250 mm 3 (n=21).In this latter group,all tumor-bearing animals had complete tu-mor regression and remained free of tumor recurrence. In subsequent tumor rechallenge experiments a significant delay in tumor growth compared to the initial tumor cell inoculation was observed(tumor volume at day 28:197.8±87.3 mm 3 vs 500.1±50.9 mm3,Pmpanied by systemic activation of immune response mechanisms predominantly mediated by the innate arm of the immune system. CONCLUSION:The observed complete tumor regression is encouraging and shows that immunotherapy with ***-NT is an interesting strategy for the treatment of pancreatic carcinomas of defined sizes.

Exocytosis of mam maUan sperm dense-core secretory granule relies on the same fusion molecules as all other secretory cells; one such molecule is the small GTPase Rab3A. Here, we report an in-depth biochemical characterization of the role of Rab3A in secretion by scrutinizingthe exocytotic response of streptolysin O-permeabiUzed human sperm to the acute application of a number of Rab3A-containing constructs and correlating the findings with those gathered with the endogenous protein. Full length, geranyigeranyiated, and active Rab3A elicited human sperm exocytosis perse. With Rab3A/Rab22A chimeric proteins, we demonstrated that the carboxy-terminal domain of the Rab3A molecule was necessary and sufficient to promote exocytosis, whereas its *** prevented calcium-triggered secretion. Interestingly, full length Rab3A halted secretion when added after the docking of the acrosome to the plasma membrane. This effect depended on the inability of Rab3A to hydrolyze GTP. We combined modified immunofluorescence and acrosomal staining protocols to detect membrane fusion and the activation status of endogenous Rab3 simultaneously in individual cells, and found that GTP hydrolysis on endogenous Rab3 was mandatory for fusion pores to open. Our findings contribute to establishing that Rab3 modulates regulated exocytosis differently depending on the nucleotide bound and the exocytosis stage under study.

Optimization of wavelength range and data interval in chemometric analysis of complex pharmaceutical mixtures

The performance of different chemometric approaches was evaluated in the spectrophotometric determination of pharmaceutical mixtures characterized by having the amount of components with a very high ratio. Principal component regression （PCR）, partial least squares with one dependent variable （PLS1） or multi-dependent variables （PLS2）, and multivariate curve resolution （mCR） were applied to the spectral data of a ternary mixture containing paracetamol, sodium ascorbate and chlorpheniramine （150：140：1, m/m/m）, and a quaternary mixture containing paracetamol, caffeine, phenylephrine and chlorpheniramine （125：6. 25：1.25：1, m/m/m/m）. The UV spectra of the calibration samples in the range of 200-320 nm were pre-treated by removing noise and useless data, and the wavelength regions having the most useful analytical information were selected using the regression coefficients calculated in the multivariate modeling. All the defined chemometric models were validated on external sample sets and then applied to commercial pharmaceutical formulations. Different data intervals, fixed at 0.5, 1.0, and 2.0 point/nm, were tested to optimize the prediction ability of the models. The best results were obtained using the PLSlcalibration models and the quantification of the species of a lower amount was sig- nificantly improved by adopting 0.5 data interval, which showed accuracy between 94.24% and 107.76%.

The primary goal in the analysis of hierarchical distributed monitoring and control architectures is to study the spatiotemporal patterns of the interactions between areas or *** this paper,a novel conceptual framework for distributed monitoring of power system oscillations using multiblock principal component analysis(mB-PCA)and higher-order singular value decomposition(HOSVD)is proposed to understand,characterize,and visualize the global behavior of the power *** proposed framework can be used to evaluate the influence of a given area or utility on the oscillatory behavior,uncover low-dimensional structures from high-dimensional data,and analyze the effects of heterogeneous data on the modal characteristics and interpretation of power *** metrics are then investigated to examine the relationships between the dynamic patterns and participation of individual data blocks in the global behavior of the *** application of these techniques is demonstrated by case studies of two systems:a 14-machine test system and a 5449-bus 635-generator equivalent model of a large power system.
