Automated Chinese medicinal plants classification based on machine learning using leaf morpho-colorimetry,fractal dimension and visible/near infrared spectroscopy

The identification of Chinese medicinal plants was conducted to rely on ampelographic manual assessment by *** recently,machine learning algorithms for pattern recognition have been successfully applied to leaf recognition in other plant *** new tools make the classification of Chinese medicinal plants easier,more efficient and cost *** study showed comparative results between machine learning models obtained from two methods:i)a morpho-colorimetric method and ii)a visible(VIS)/Near Infrared(NIR)spectral analysis from sampled leaves of 20 different Chinese medicinal ***,the automated image analysis and VIS/NIR spectral based parameters obtained from leaves were used separately as inputs to construct customized artificial neural network(ANN)*** showed that the ANN model developed using the morpho-colorimetric parameters as inputs(Model A)had an accuracy of 98.3%in the classification of leaves for the 20 medicinal plants *** the case of the model based on spectral data from leaves(Model B),the ANN model obtained using the averaged VIS/NIR spectra per leaf as inputs showed 92.5%accuracy for the classification of all medicinal plants *** A has the advantage of being cost effective,requiring only a normal document scanner as measuring *** method can be adapted for non-destructive assessment of leaves in-situ by using portable wireless *** B combines the fast,non-destructive advantages of VIS/NIR spectroscopy,which can be used for rapid and non-invasive identification of Chinese medicinal plants and other applications by analyzing specific light spectra overtones from leaves to assess concentration of pigments such as chlorophyll,anthocyanins and others that are related active compounds from the medicinal plants.

How University Athletics Can Impact Mental Health among Student Athletes at the University of Evansville

In recent years, the NCAA student athlete population in the United States has surpassed 500,000, and is continuing to rise each year [1]. These student athletes work their entire lives academically and athletically to reach the ultimate goal: competing in university athletics. However, when these athletes reach university, they are met with non-stop training, homework, exams, and evolving social lives. We have conducted a study at the University of Evansville evaluating how participation in university athletics may impact mental health status among these student athletes, as well as measuring players’ awareness of accessible mental health resources. Over fifty percent of participants reported experiencing at least one mental health condition while competing in their sport;eighty percent reported having knowledge of the mental health resources available to them on campus, however, nearly thirty percent of those knowledgeable reported not knowing how to access these resources. This has indicated a gap in awareness and utilization of mental health resources among student athletes at the University of Evansville.

Extracellular vesicles-coupled miRNAs from oviduct and uterus modulate signaling pathways related to lipid metabolism and bovine early embryo development

Background Extracellular vesicles(EVs) present in oviductal(OF) and uterine fluid(UF) have been shown to enhance bovine embryo quality during in vitro culture by reducing lipid contents and modulating lipid metabolism-related genes(LMGs), while also influencing cell proliferation, suggesting their involvement on the regulation of different biological pathways. The regulation of signaling pathways related to cell differentiation, proliferation, and metabo-lism is crucial for early embryo development and can determine the success or failure of the pregnancy. Bioactive molecules within EVs in maternal reproductive fluids, such as micro RNAs(miRNAs), may contribute to this regulatory process as they modulate gene expression through post-transcriptional *** This study evaluated miRNA cargo in OF-EVs from the early luteal phase and UF-EVs from the mid-luteal phase, coinciding with embryo transit within oviduct and uterus in vivo, and its possible influence on LMGs and sign-aling pathways crucial for early embryo development. A total of 333 miRNAs were detected, with 11 exclusive to OF, 59 to UF, and 263 were common between both groups. From the 20 differentially expressed miRNAs, 19 up-regulated in UF-EVs(bta-miR-134, bta-miR-151-3p, bta-miR-155, bta-miR-188, bta-miR-181b, bta-miR-181d, bta-miR-224, bta-miR-23b-3p, bta-miR-24-3p, bta-miR-27a-3p, bta-miR-29a, bta-miR-324, bta-miR-326, bta-miR-345-3p, bta-miR-410, bta-miR-652, bta-miR-677, bta-miR-873 and bta-miR-708) and one(bta-miR-148b) in OF-EVs. These miRNAs were predicted to modulate several pathways such as Wnt, Hippo, MAPK, and lipid metabolism and degradation. MicroRNAs （miRNAs） control gene expression mostly post-transcriptionally by guiding transcript cleavage and/or translational repression of complementary mRNA targets, thereby regulating developmental processes and stress responses. Despite the remarkable expansion of the field, the mechanisms underlying miRNA activity are not fully understood. In this article, we describe a transient expression system in Arabidopsis mesophyll protoplasts, which is highly amenable for the dissection of miRNA pathways. We show that by transiently overexpressing primary miRNAs and target mimics, we can manipulate miRNA levels and consequently impact on their targets. Furthermore, we developed a set of luciferase-based sensors for quantifying miRNA activity that respond specifically to both endogenous and overexpressed miRNAs and target mimics. We demonstrate that these miRNA sensors can be used to test the impact of putative components of the miRNA pathway on miRNA activity, as well as the impact of specific mutations, by either overexpression or the use of protoplasts from the corresponding mutants. We further show that our miRNA sensors can be used for investigating the effect of chemicals on miRNA activity. Our cell-based transient expression system is fast and easy to set up, and generates quantitative results, being a powerful tool for assaying miRNA activity in vivo.

Modern and paleoclimate changes may have altered species dynamics by shifting species’niche suitability over space and *** analyze whether the current genetic structure and isolation of the two large American felids,jaguar(Panthera onca)and puma(Puma concolor),are mediated by changes in climatic suitability and connection routes over modern and paleoclimatic *** estimate species distribution under 5 climatic landscapes(modern,Holocene,last maximum glaciations[LMG],average suitability,and climatic instability)and correlate them with individuals’genetic isolation through causal modeling on a resemblance *** species exhibit genetic isolation patterns correlated with LMG climatic suitability,suggesting that these areas may have worked as“allele refuges.”However,the jaguar showed higher vulnerability to climate changes,responding to modern climatic suitability and connection routes,whereas the puma showed a continuous and gradual transition of genetic *** differential responsiveness to climate change,both species are subjected to the climatic effects on genetic configuration,which may make them susceptible to future climatic changes,since these are progressing faster and with higher intensity than changes in the ***,the effects of climatic changes should be considered in the design of conservation strategies to ensure evolutionary and demographic processes mediated by gene flow for both species.

The opening of maitotoxin-sensitive calcium channels induces the acrosome reaction in human spermatozoa： differences from the zona pellucida

The acrosome reaction （AR）, an absolute requirement for spermatozoa and egg fusion, requires the influx of Ca2＋ into the spermatozoa through voltage-dependent Ca2＋ channels and store-operated channels. Maitotoxin （MTx）, a Ca2＋-mobilizing agent, has been shown to be a potent inducer of the mouse sperm AR, with a pharmacology similar to that of the zona pellucida （ZP）, possibly suggesting a common pathway for both inducers. Using recombinant human ZP3 （rhZP3）, mouse ZP and two MTx channel blockers （U73122 and U73343）, we investigated and compared the MTx- and ZP-induced ARs in human and mouse spermatozoa. Herein, we report that MTx induced AR and elevated intracellular Ca2＋（[Ca2＋]~） in human spermatozoa, both of which were blocked by U73122 and U73343. These two compounds also inhibited the MTx-induced AR in mouse spermatozoa. In disagreement with our previous proposal, the AR triggered by rhZP3 or mouse ZP was not blocked by U73343, indicating that in human and mouse spermatozoa, the AR induction by the physiological ligands or by MTx occurred through distinct pathways. U73122, but not U73343 （inactive analogue）, can block phospholipase C （PLC）. Another PLC inhibitor, edelfosine, also blocked the rhZP3- and ZP-induced ARs. These findings confirmed the participation of a PLC-dependent signalling pathway in human and mouse zona protein-induced AR. Notably, edelfosine also inhibited the MTx-induced mouse sperm AR but not that of the human, suggesting that toxin-induced AR is PLC-dependent in mice and PLC-independent in humans.
