Histone deacetylase 1(HDAC1)is an important epigenetic controller involvedin transcriptional regulation throughmodification of chromatin *** and epigenetic changes and deregulation of signal transduction pathways have been implicated in the development of breast *** of estrogen receptor a(ERa)expression is one of the mechanisms behind the acquisition of endocrine *** and increased hormone and growth factor receptor signaling in breast cancer cells contribute to resistance to endocrine *** HDACs and the PI3K/mTOR signaling pathway are becoming promising targets in breast cancer,reversing also acquired hormone *** we show how mitogens,activating the PI3K/mTOR pathway,trigger the phosphorylation of HDAC1 in breast cancer cells,which is completely dependent on the activity of the p70 S6 kinase(S6K1).Our findings show that S6K1,overexpressed in many breast cancers,controls HDAC1-dependent transcriptional regulation of ERa levels upon mitogenic stimuli,controlling HDAC1 recruitment to the ERa ***,cell treatment with both mTOR and HDACs inhibitors shows an additive effect in inhibiting breast cancer *** confirms the novel cross-talk between the HDAC1 and PI3K pathways with clinical implications towards the treatment of this malignant disease.

Multimedia content,user mobility and heterogeneous client devices require novel systems that are able to support ubiquitous access to the Web *** this scenario,solutions that combine flexibility,efficiency and scalability in offering edge services for ubiquitous access are *** propose an original intermediary framework,namely Scalable Intermediary Software Infrastructure （SISI）,which is able to dynamically compose edge services on the basis of user preferences and device *** SISI framework exploits a per-user profiling mechanism,where each user can initially set his/her personal preferences through a simple Web interface,and the system is then able to compose at run-time the necessary *** basic framework can be enriched through new edge services that can be easily implemented through a programming model based on APIs and internal *** experiments demonstrate that flexibility and edge service composition do not affect the system *** show that this framework is able to chain multiple edge services and to guarantee stable performance.

Early-onset colorectal cancer(EOCRC)has seen an alarming rise worldwide over the past two *** reason for this global trend is poorly *** appears to have its own unique clinical and molecular features when compared with late-onset colorectal *** patients appear to have more distal or rectal disease,a more advanced stage of disease at presentation,and more unfavorable histological *** risk factors for EOCRC is the first step in mitigating the rising burden of this *** we summarize several noteworthy biological factors and environmental exposures that are postulated to be responsible *** can hopefully translate in clinical practice to the development of better risk stratification tool for identifying highrisk individuals for early colorectal cancer screening,and identifying areas needed for further research to curb this rising trend.

One disadvantage of direct anticoagulant drug is the lack of an antidote, which may become relevant in patients with traumatic brain in- jury. A 77-years old man with atrial fibrillation and syncope received dabigatran despite recurrent falls. Due to a ground-level-fall, he suffered from subarachnoidal and intraparenchymal hemorrhages, subdural hematoma and brain edema with a midline shift. Despite osteoelast/c trepanation and hematoma-evacuation he remained comatose and died seven days later without regaining consciousness. Most probably, decreased dabigatran clearance due to increased age might have contributed to the fatal course. We suggest withholding anticoagulant therapy in patients with unexplained falls. If anticoagulant therapy is deemed necessary, vitamin-K-antagonists with their potential for laboratory monitoring and reversal of anticoagulant activity should be preferred.

Aim： To simultaneously determine the localization of histones and protamines within human sperm nuclei. Methods： Immunofluorescence of the core histones and protamines and fluorescence in situ hybridization of the telomere region of chromosome 16 was assessed in decondensed human sperm nuclei. Results： Immunofluorescent localization of histones, protamine 1 （PRM1） and protamine 2 （PRM2） along with fluorescence in situ hybridization localization of chromosome 16 telomeric sequences revealed a discrete distribution in sperm nuclei. Histones localized to the posterior ring region （i.e. the sperm nuclear annulus）, whereas PRM1 and PRM2 appeared to be dispersed throughout the entire nucleus. Conclusion： The co-localization of the human core sperm histones with the telomeric regions of chromosome 16 is consistent with the reorganization of specific non-protamine regions into a less compacted state.

Orthodontic forces have been reported to significantly increase nicotine-induced periodontal bone loss. At present, however, it is unknown, which further （side） effects can be expected during orthodontic treatment at a nicotine exposure corresponding to that of an average European smoker. 63 male Fischer344 rats were randomized in three consecutive experiments of 21 animals each （A/B/C） to 3 experimental groups （7 rats, 112/3）. （A） cone-beam-computed tomography （CBCT）; （B） histology/serology; （C） reverse- transcription quantitative real-time polymerase chain reaction （RT-qPCR）/cotinine serology--（1） control; （2） orthodontic tooth movement （OTM） of the first and second upper left molar （NiTi closed coil spring, 0.25 N）; （3） OTM with 1.89 mg-kg- 1 per day s.c. of L（- ）-nicotine. After 14 days of OTM, serum cotinine and IL-6 concentration as well as orthodontically induced inflammatory root resorption （OIIRR）, osteoclast activity （histology）, orthodontic tooth movement velocity （CBCT, within 14 and 28 days of OTM） and relative gene expression of known inflammatory and osteoclast markers were quantified in the dental-periodontal tissue （RT-qPCR）. Animals exposed to nicotine showed significantly heightened serum cotinine and IL-6 levels corresponding to those of regular European smokers. Both the extent of root resorption, osteoclast activity, orthodontic tooth movement and gene expression of inflammatory and osteoclast markers were significantly increased compared to controls with and without OTM under the influence of nicotine. We conclude that apart from increased periodontal bone loss, a progression of dental root resorption and accelerated orthodontic tooth movement are to be anticipated during orthodontic therapy, if nicotine consumption is present. Thus patients should be informed about these risks and the necessity of nicotine abstinence during treatment.

Surgical treatment of peripheral nerve injuries is still a major challenge in human *** to now,none of the well-developed microsurgical treatment options is able to guarantee a complete restoration of nerve *** restriction is also effective for novel clinically approved artificial nerve *** this review,we compare surgical repair techniques primarily for digital nerve injuries reported with relatively high prevalence to be valuable attempts in clinical digital nerve repair and point out their advantages and *** furthermore discuss the use of artificial nerve grafts with a focus on chitosan-based nerve guides,for which our own studies contributed to their approval for clinical *** the second part of this review,very recent future perspectives for the enhancement of tubular(commonly hollow)nerve guides are discussed in terms of their clinical translatability and ability to form three-dimensional constructs that biomimick the natural nerve *** includes materials that have already shown their beneficial potential in in vivo studies like fibrous intraluminal guidance structures,hydrogels,growth factors,and approaches of cell ***,we highlight upcoming future perspectives comprising co-application of stem cell *** our overview,we conclude that already simple attempts are highly effective to increase the regeneration supporting properties of nerve guides in experimental *** for bringing nerve repair with bioartificial nerve grafts to the next level,*** of defects>3 cm in human patients,more complex intraluminal guidance structures such as innovatively manufactured hydrogels and likely supplementation of stem cells or their secretome for therapeutic purposes may represent promising future perspectives.

For a decade,sorafenib remained the only drug approved for the treatment of advanced hepatocellular carcinoma(HCC)due to the failure of several phase III trials in which alternative angiogenesis inhibitors were tested(1).In recent years,the field of systemic treatments of HCC has evolved quickly and a recent position paper of the European Association for the Study of the Liver has well described the current advances in the systemic treatment of advanced HCC as well as the unmet needs in the field(1).First,lenvatinib was approved as an alternative to sorafenib in the first-line setting,and,thus,regorafenib,cabozantinib and ramucirumab as second-line treatments for patients who progressed on sorafenib(1).

The catalytic oxidative cou pling of methane(OCM)to C_(2) hydrocarbons(C_(2)H_(6) and C_(2)H_(4))represents one of the most effective ways to convert natural gas to more useful products,which can be performed effectively using La_(0.6)Sr0.4Co_(0.2)Fe_(0.8)O_(3-δ)(LSCF)perovskite hollow fiber membrane *** this work,the effects of adding a thin BaCe_(0.8)Gd_(0.2)O_(3-δ)(BCG)catalyst film onto the inner LSCF fiber surface as the OCM catalyst and a porous Ba0.5Sr_(0.5)Co_(0.8)Fe_(0.2)O_(3-δ)(BSCF)perovskite layer onto the outer LSCF surface to improve the oxygen permeation were *** 700℃and 1000℃,methane conversion increased in the order of uncoated,BCG and BSCF-coated,and BCG-coated LSCF hollow fiber while Coselectivity and C_(2)-yield increased in the order of BCG and BSCF-coated,uncoated,and BCG-coated LSCF hollow *** permeation flux at the same temperature range,on the other hand,was enhanced in the order of uncoated,BCG-coated,and BCG and BSCF-coated LSCF hollow *** finding demonstrates the complex interplay between oxygen permeation and OCM *** BCG and BSCFcoated hollow fiber was also subjected to thermal cycles between 850℃and 900℃for up to 175 hours during which the fiber showed minor degradation in oxygen permeation fluxes and relatively stable OCM performance.
