This work explores the potential of a triple combination of meropenem(MEM),a novel metallo-blactamase(MBL)inhibitor(indole-2-carboxylate 58(InC58)),and a serine-b-lactamase(SBL)inhibitor(avibactam(AVI))for broad-spectrum activity against carbapenemase-producing bacteria.A diverse panel comprising MBL-and SBL-producing strains was used for susceptibility testing of the triple combination using the agar dilution *** frequency of resistance(FoR)to MEM combined with InC58 was *** were sequenced and tested for cross resistance,fitness,and the stability of the resistance *** with the double combinations of MEM plus an SBL or MBL inhibitor,the triple combination extended the spectrum of activity to most of the isolates bearing SBLs(oxacillinase-48(OXA-48)and Klebsiella pneumoniae carbapenemase-2(KPC-2))and MBLs(New Delhi metallo-blactamases(NDMs)),although it was not effective against Verona integron-encoded metallo-blactamase(VIM)-carrying Pseudomonas aeruginosa(***)and OXA-23-carrying Acinetobacter baumannii(***).The FoR to MEM plus InC58 ranged from 2.22×10^(-7)to 1.13×10^(-6).The resistance correlated with mutations to ompC and comR,affecting porin C and copper permeability,*** mutants manifested a fitness cost,a decreased level of resistance during passage without antibiotic pressure,and cross resistance to another carbapenem(imipenem)and a b-lactamase inhibitor(taniborbactam).In conclusion,compared with the dual combinations,the triple combination of MEM with InC58 and AVI showed a much wider spectrum of activity against different carbapenemaseproducing bacteria,revealing a new strategy to combat b-lactamase-mediated antimicrobial resistance.

Deformation at low and high stress-loading rates

In an extensional shear zone in the Talea Ori, Crete, quartz veins occur in high-pressure low-temperature metamorphic sediments at sites of dilation along shear band boundaries, kink band boundaries and boudin necks. Bent elongate grains grown epitactically from the host rock with abundant fluid inclusion trails parallel to the vein wall indicate vein formation by crack-seal increments during dissolutionprecipitation creep of the host rock. The presence of sutured high-angle grain boundaries and subgrains shows that temperatures were sufficiently high for recovery and strain-induced grain boundary migration, i.e. higher than 300 -350℃, close to peak metamorphic conditions. The generally low amount of strain accumulated by dislocation creep in quartz of the host rock and most veins indicates low bulk stress conditions of a few tens of MPa on a long term. The time scale of stress-loading to cause cyclic cracking and sealing is assumed to be lower than the Maxwell relaxation time of the metasediments undergoing dissolution-precipitation creep at high strain rates(10^(-10) s^(-1) to 10^(-9) s^(-1)), which is on the order of hundred years. In contrast, some veins discordant or concordant to the foliation show heterogeneous quartz microstructures with micro-shear zones, sub-basal deformation lamellae, shortwavelength undulatory extinction and recrystallized grains restricted to high strain zones. These microstructures indicate dislocation glide-controlled crystal-plastic deformation(low-temperature plasticity) at transient high stresses of a few hundred MPa with subsequent recovery and strain-induced grain boundary migration at relaxing stresses and temperatures of at least 300 -350℃. High differential stresses in rocks at greenschist-facies conditions that relieve stress by creep on the long term, requires fast stress-loading rates, presumably by seismic activity in the overlying upper crust. The response of cells during spreading and motility is dictated by several multi-physics events,which are triggered by extracellular cues and occur at different *** this sake,it is not completely appropriate to provide a cell with classical notions of the mechanics of materials,as for“rheology”or“mechanical response”.Rather,a cell is an alive system with constituents that show a reproducible response,as for the contractility for single stress fibers or for the mechanical response of a biopolymer actin network,but that reorganize in response to external cues in a non-exactly-predictable and reproducible *** of such complexity,in this note we aim at formulating a multi-physics framework for modeling cells spreading and motility,accounting for the relocation of proteins on advecting lipid membranes.

Probiotics to counteract biofilm-associated infections: promising and conflicting data

Altered bowel flora is currently thought to play a role in a variety of disease conditions, and the use of Bifidobacterium spp. and Lactobacillus spp. as probiotics has been demonstrated to be health-promoting, even if the success of their administration depends on the applied bacterial strain（s） and the targeted disease. In the last few decades, specific probiotics have been shown to be effective in the treatment or the prevention of acute viral gastroenteritis, pediatric post-antibiotic-associated diarrhea, some pediatric allergic disorders, necrotizing enterocolitis in preterm infants, inflammatory bowel diseases and postsurgical pouchitis. The potential application of probiotics is continuously widening, with new evidence accumulating to support their effect on the prevention and treatment of other disease conditions, including several oral diseases, such as dental caries, periodontal diseases and oral malodor, as well as genitourinary and wound infections. Considering the increasingly widespread ability of pathogens to generate persistent biofilm-related infections, an even more attractive proposal is to administer probiotics to prevent or counteract biofilm *** response of biofilm-based oral, intestinal, vaginal and wound infections to probiotics treatment will be reviewed here in light of the most recent results obtained in this field.

Applying design science approach to architectural design development

Unlike research methods for social and positivist sciences,those for architectural design lack a discipline-specific conceptual *** science aims at producing outcomes for future use and therefore needs a robust methodological approach that encompasses different techniques and methods supporting an evidence-based architectural design *** study suggests that design science can be successfully applied to architectural design development and provides architects and designers with a powerful tool bridging the gap between research and *** so doing,this study explores the application of design science to implement a user-centered design approach.A design challenge is reframed within the robust framework of design science by referring to a case study on refugee *** traditional method by which shelter optimization is pursued by designers is questioned by involving the final users through an ethnographic approach within the framework of design *** design outcome produced through this process is a list of specifications allowing designers to create different architectural solutions and matching the requirements expressed by future *** authors argue that a user-centered design outcome can be achieved and validated through design science.
