Association between Polymorphisms of Glutathione S-Transferase and Progression to Cervical Cancer in Women from Burkina Faso and Mali

Although persistence of high-risk human papillomavirus infection is the main risk factor, Glutathione S-Transferase highly polymorphic enzyme involved in the metabolism of xenobiotics, is a good candidate gene. The objective of this study was to compare the polymorphisms of Glutathione S-Transferase M1-null in women with cancerous lesions and without lesions. This study consisted of 322 uterine cervix samples of women from Mali and Burkina Faso with Cervical Intra-epithelial Neoplasia 2 and 3, adenocarcinoma and squamous cell carcinoma and 100 women with no lesions. Human Papillomavirus genotyping was performed by Real-time multiplex Polymerase Chain Reaction. Glutathione S-Transferase gene polymorphisms were determined using conventional Polymerase Chain Reaction followed by migration on agarose gel. A statistically significant association with high relative risks of 10.77 for the development of High grade Superficial or Squamous Intra-epithelial Lesion (95% CI = 5.59 - 20.72;p < 0.001), and 13.20 for cancer development (95% CI = 6.79 - 25.63;p < 0.001) was found in women with the null genotype of Glutathione S-Transferase M1 in the study population. In Burkina Faso and Mali, Glutathione S-Transferase M1-null presented relative risks of 9 and 11.05 for high-grade lesions, 15 and 11.40 for cancer. Similarly, significant results had been observed in women with human papillomavirus positive and human papillomavirus negative. The results of the present study support the idea that the deletion of Glutathione S-Transferase M1 plays a crucial role in the progression of high-grade lesions and cervical cancer.

Anti-hepatitis C virus potency of a new autophagy inhibitor using human liver slices model

AIM: To evaluate the antiviral potency of a new antihepatitis C virus(HCV) antiviral agent targeting the cellular autophagy machinery. METHODS: Non-infected liver slices, obtained from human liver resection and cut in 350 μm-thick slices(2.7 × 106 cells per slice) were infected with cell culture-grown HCV Con1b/C3 supernatant(multiplicity of infection = 0.1) cultivated for up to ten days. HCV infected slices were treated at day 4 post-infection with GNS-396 for 6 d at different concentrations. HCV replication was evaluated by strand-specific real-time quantitative reverse transcription- polymerase chain reaction. The infectivity titers of supernatants were evaluated by foci formation upon inoculation into naive Huh-7.5.1 cells. The cytotoxic effect of the drugs was evaluated by lactate dehydrogenase leakage assays. RESULTS: The antiviral efficacy of a new antiviral drug, GNS-396, an autophagy inhibitor, on HCV infection of adult human liver slices was evidenced in a dosedependent manner. At day 6 post-treatment, GNS-396 EC50 was 158 nmol/L without cytotoxic effect(compared to hydroxychloroquine EC50 = 1.17 μmol/L).CONCLUSION: Our results demonstrated that our ex vivo model is efficient for evaluation the potency of autophagy inhibitors, in particular a new quinoline derivative GNS-396 as antiviral could inhibit HCV infection in a dosedependent manner without cytotoxic effect.

热红外天底探测仪是观测诸如水汽、二氧化碳和臭氧等大气组分总柱含量或垂直剖面的理想仪器。低于5cm-1光谱分辨率的高分辨率探测仪能够分辨痕量气体细微的光谱特征。在第一台高光谱探测仪IRIS出现的40年后,现在几款改进了仪器特性的仪器已经在轨运行。本文评述了应用热红外天底探测仪观测痕量气体,重点在于红外大气探测干涉仪(IASI)的新近观测。文章介绍了14种非常活泼的痕量气体的典型观测。从天底视角观测气体讲,一些组分属首次报道,包括亚硝酸、呋喃、乙炔、丙基烯、乙酸、甲醛和氰化氢;这些组分是在2009年2月澳大利亚丛林大火的火积云中观测到的。由于能够观测这些大量活性的痕量气体,所以我们关于源排放及其对环境和气候方面影响的知识可能会加深。

Income Diversification through a Crop-Livestock-Forest Integration System in the Midwest Brazilian Region

Sustainability on farms is a challenge to competitiveness in the globalized market. In this scenario, due to the strong environmental, economic and social appeal, crop-livestock-forest integration systems have been considered as the future of agriculture. Regarding the economic approach, this system is based on the diversification of income generating activities, with revenue entry at different times, rationalization of resource use and reduction of risk of financial losses. Thus, the purpose of this work was to carry out the economic analysis of a crop-livestock-forest integration system, located at Boa Vereda"s farm, in the municipality of Cachoeira Dourada in the state of Goiás in the central-west region of Brazil. The system was composed of the following traditional crops of the region: soybean, corn, pasture, beef cattle and eucalyptus. The technical coefficients and the prices used for economic evaluation were obtained from the experimental unit and from the local market when crops were harvested in 2016/2017. The economic indicators used to assess economic viability were the net present value (NPV) and the equivalent annual uniform value (EAUV). The results showed a return greater than the opportunity cost of the land, showing the attractiveness of the crop-livestock-forest integration system. Thus, this system represents an economically viable alternative that, among other benefits, allows the diversification of its sources of income, with a reduction of risk.

Impact of Heap Fermentation of Cocoa on Microbial Dynamics and Soil Physicochemical Parameters

The overall objective of the present study is to evaluate the impact of the heap fermentation of cocoa on microbial dynamics and physicochemical parameters of the soil. The methodology was to heap fermentation broad beans 600 cocoa pods moved to a place after the soil was taken for microbiological and physicochemical analyzes considered the control sample. In addition, cocoa lixiviate and soil were subjected to analyze. Chemical analysis of cocoa lixiviate revealed the absence of heavy metals such as cadmium, chromium. It appears from the analysis of soil than clays represent on average 46.67%, 8.03% for fine silt, heavy silt 5.69%, 15.39% fine sands and heavy sands 20.02%. Microbiological analysis revealed the abundance of total coliform up to 4.6 × 103 CFU/g soil. The variations of the abundance of yeasts are 0.01 × 103 CFU/g soil obtained on day 2 at 12 o"clock to 3.5 × 103 CFU/g soil observed on day 3 to 18 pm (0-3 cm deep). However, further study on the assessment of biodiversity after the fermentation would determine its species richness.

Assessment of Trace Metal Levels of Groundwater in Lubumbashi,Kampemba and Kamalondo Communes of Lubumbashi City,Democratic Republic of Congo

Trace metal levels of groundwater in Lubumbashi,Kampemba and Kamalondo communes of Lubumbashi city were assessed from October 2016 to February *** hundred forty water samples collected from twenty-two spade-sunk wells and twelve drilled wells in these three communes of Lubumbashi city were analyzed for their metal contents using ICP-SF-MS(Inductively Coupled Plasma-Sector Field Mass Spectrometry).Twenty trace elements including strontium,molybdenum,cadmium,cesium,barium,tungsten,thallium,lead,bismuth,uranium,aluminum,vanadium,chromium,manganese,iron,cobalt,nickel,copper,zinc and arsenic were recorded at varying concentrations in all the water samples and were compared with the WHO(World Health Organization),US EPA(Environmental Protection Agency)and EU(European Union)drinking water MCLs(Maximum Concentration Limits)for cadmium,barium,thallium,lead,uranium,aluminum,chromium,manganese,iron,nickel,copper,zinc and *** cadmium,lead,aluminum,manganese,iron,nickel,zinc and arsenic levels respectively exceeded the WHO,US EPA and EU drinking water MCLs in 6.66%,3.38%,26.67%,5.02%,30.03%,3.38%,1.64% and 5.02% of the samples with the highest mean levels of 116.89μg/L for cadmium,38.162μg/L for lead,2,712.5μg/L for aluminum,1,242.68μg/L for manganese,17,325.98μg/L for iron,64.647μg/L for nickel,9,900.72μg/L for zinc and 65.458μg/L for *** water pH values ranged from 4.7 to 11.1 with 19.17% of the groundwater samples having mean pH values outside the WHO drinking water pH optimum range values of 6.5-8.5,including 5.02% of the water samples which were acidic(mean pH values ranging from 4.7 to 6.4)and 14.15% which were alkaline(mean pH values ranging from 8.6 to 11.1).With such physicochemical and trace metal contamination status of groundwater in the three communes of Lubumbashi city,there is a high risk to the health of people who use that water to meet their drinking water needs.
