Crohn's disease genotypes of patients in remission vs relapses after infliximab discontinuation

AIM:To investigate genetic differences between Crohn's disease(CD) patients with a sustained remission vs relapsers after discontinuing infliximab while in corticosteroid-free ***:Forty-eight CD patients received infliximab and were in full corticosteroid-free clinical remission but then discontinued infliximab for reasons other than a loss of response,were identified by review of an electronic database and ***-associated remission was defined as corticosteroid-free plus normalization of clinical disease activity [CD activity index(CDAI) 220) and a therapeutic intervention with CD medication(s),or a hospitalization with complications related to active *** analyses were performed on samples from 14 patients(n = 6 who had a sustained long term remission after stopping infliximab,n = 8 who rapidly relapsed after stopping infliximab).Nucleotide-binding oligomerization domain 2(NOD2)/caspase activation recruitment domain 15(CARD15) polymorphisms(R702W,G908R and L1007fs) and the inflammatory bowel disease 5(IBD5) polymorphisms(IGR2060a1 and IGR3081a1) were analyzed in each ***:Five single nucleotide polymorphisms of IBD5 and NOD2/CARD15 genes were successfully analyzed for all 14 *** was no significant increase in frequency of the NOD2/CARD15 polymorphisms(R702W,G908R and L1007fs) and the IBD5 polymorphisms(IGR2060a1 and IGR3081a1) in either group of patients;those whose disease relapsed rapidly or those who remained in sustained long term remission following the discontinuation of *** a third of patients in full clinical remission who stopped infliximab for reasons other than loss of response remained in sustained clinical remission,while two-thirds relapsed *** was a marked difference in the

Peptide photocaging:A brief account of the chemistry and biological applications

Light is arguably the most convement non-invasive stimulant to perturb or control specinc cnemical reactions in the biological systems. Upon light illumination, photosensitive molecules incur conformational changes or formation/breaking of chemical bonds. Consequently, these molecules can be used to transfer signals from one location to another in the cell, from outside of the cell to the inside, or from a light bulb to the interior of animal tissues. The development of the photochemical reactions of organic compounds has paved the road towards their use in peptides and peptide-based biological applications. In this mini-review, we summarized the state-of-the-art development of photo-protecting groups for peptide photocaging including the un-caging mechanism of different PPGs, the synthesis of photo-caged peptides, and the recent applications of peptide photocaging in chemical biology.

A CHALLENGE OF THE EXPERIMENT ON TYPHOON INTENSITY CHANGE IN COASTAL AREA

The Experiment on Typhoon Intensity Change in Coastal Area(EXOTICCA) was proposed by the China Meteorological Administration(CMA) and Hong Kong Observatory(HKO) and endorsed by the ESCAP/WMO Typhoon Committee(TC). The major goals and objectives of the EXOTICCA are: 1) to conduct the field campaigns on the intensity and structural characteristics of the target offshore and landfall tropical cyclones by employing integrated and novel observation techniques, and 2) to conduct demonstration research on the utilization of the synergized field observation data with the aim of deepening the understanding of the mechanism of structure and intensity changes, to improve the relevant capability of operational analysis, numerical weather prediction(NWP) models forecast, reliable storm surge and flooding and associated risk assessment. The Organizational structure and implementation schedule etc. are also introduced in this paper.

Efficacy and tolerability of vardenafil in Asian men with erectile dysfunction

Aim： To evaluate the efficacy and tolerability of vardenafil, a phosphodiesterase type-5 （PDE-5） inhibitor, in men of Asian ethnicity with erectile dysfunction （ED）. Methods： In this prospective, double-blind, multinational study, Asian men were randomized to receive vardenafil （10 mg） or placebo （4：1 ratio） for 12 weeks. The primary efficacy variables were the International Index of Erectile Function erectile function domain （IIEF-EF）, and Sexual Encounter Profile （SEP） questions related to penetration and intercourse completion. Significant mean improvements were required in all three measures to show positive benefits of vardenafil treatment. Secondary efficacy variables included the Global Assessment Question （GAQ） on erection improvement. Results： Least-squares mean baseline IIEF-EF domain scores （vardenafil 14.6, placebo 13.4） were consistent with moderate ED. After 12 weeks, vardenafil treatment was associated with significant increases from the baseline in IIEF-EF domain scores compared with the placebo （22.4 vs. 14.3; P 〈 0.001）. Vardenafil was associated with significant improvements from baseline in least squares （LS） mean success rates for SEP-2 （vardenafil 82.2 vs. placebo 43.6; P 〈 0.001） and SEP-3 （vardenafil 66.1 vs. placebo 24.0; P 〈 0.001）. Positive GAQ responses were reported by 81.8% of vardenafil recipients vs. 24.3% of placebo recipients. Adverse events were reported by 25.4% of the vardenafil group, the majority mild and transient. Conclusion： Vardenafil （10 mg） is a highly effective and well-tolerated treatment for moderate ED in Asian men. These results add to the increasing amount of data demonstrating the safety and efficacy of vardenafil for the treatment of ED in a range of patient populations.
