Increased expression of chondroitin sulphate proteoglycans in rat hepatocellular carcinoma tissues

AIM:To investigate the expression of chondroitin sulphate proteoglycans(CSPGs)in rat liver tissues of hepatocellular carcinoma(HCC).MeTHODS:Thirty male Sprague Dawley rats were randomly divided into two groups:control group(n=10) and HCC model group(n=20).Rats in the HCC model groups were intragastrically administrated with 0.2%(w/v)N-diethylnitrosamine(DeN)every 5 d for 16 wk,whereas 0.9%(w/v)normal saline was administered to rats in the control *** 16 wk from the initiation of experiment,all rats were killed and livers were collected and fixed in 4%(w/v)*** tissues were embedded in paraffin and *** staining(hematoxylin and eosin and Toluidine blue)was performed to demonstrate the onset of HCC and the content of sulphated glycosaminoglycan(sGAG).Immunohistochemical staining was performed to investigate the expression of chondroitin sulphate(CS)/dermatan sulphate(DS)-GAG,heparan sulphate(HS)-GAG,keratan sulphate(KS)-GAG in liver ***,expression and distribution of CSPG family members,including aggrecan,versican,biglycan and decorin in liver tissues,were also immunohistochemically ***:After 16 wk administration of DeN,malignant nodules were observed on the surface of livers from the HCC model group,and their hepatic lobule structures appeared largely disrupted under *** blue staining demonstrated that there was an significant increase in sGAG content in HCC tissues when compared with that in the normal liver tissues from the control group[0.37±0.05 integrated optical density per stained area(IOD/area)and 0.21± 0.01 IOD/area,Pemical studies demonstrated that this increased sGAG in HCC tissues was induced by an elevated expression of CS/DS(0.28±0.02 IOD/area and 0.18±0.02 IOD/area,Pea and 0.17±0.02 IOD/area,Pes thereby were performed to investigate the expression and distribution o

Objective： To investigate the effects of mycotoxin moniliformin （MON） on the metabolism of aggrecan and type 11 collagen in human chondrocytes in vitro and the relationship between MON and Kashin-Beck disease （KBD）. Methods： Human chondrocytes were isolated and cultured on bone matrix gelatin to form an artificial cartilage model in vitro with or without MON toxin. Cell viability was determined by 3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide （MTT） assay. The expression of aggrecan and type II collagen in the cartilage was determined using immunocytochemical staining. Results： MON toxin inhibited chondrocyte viability in dose-dependent and time-dependent manners. MON reduced aggrecan and type Ⅱ collagen syntheses in the tissue-engineered cartilage. MON also increased the expression of matrix metalloproteinase-1 （MMP-1）, MMP-13, BC4 epitopes, and CD44 in cartilages. However, the expression of 3B3（-） epitopes in cartilages was inhibited by MON. Selenium partially alleviated the damage of aggrecan induced by MON toxin. Conclusion： MON toxin promoted the catabolism of aggrecan and type II collagen in human chondrocytes.

Microstructure and properties of Ti-6Al-4V fabricated by low-power pulsed laser directed energy deposition

Thin-wall structures of Ti-6A1-4V were fabricated by low-power pulsed laser directed energy deposition. During deposition, consistent with prior reports, columnar grains were observed which grew from the bottom toward the top of melt pool tail. This resulted in a microstructure mainly composed of long and thin prior epitaxial β columnar grains (average width ^200μm). A periodic pattern in epitaxial growth of grains was observed, which was shown to depend upon laser traverse direction. Utilizing this, a novel means was proposed to determine accurately the fusion boundary of each deposited layer by inspection of the periodic wave patterns. As a result it was applied to investigate the influence of thermal cycling on microstructure evolution. Results showed that acicular martensite,α' phase, and a small amount of Widmanstatten, a laths, gradually converted to elongated acicular a and a large fraction of Widmanstatten a laths under layer-wise thermal cycling. Tensile tests showed that the yield strength, ultimate tensile strength and elongation of Ti-6Al-4V thin wall in the build direction were 9.1 %, 17.3% and 42% higher respectively than those typically observed in forged solids of the same alloy. It also showed the yield strength and ultimate tensile strength of the transverse tensile samples both were 13.3% higher than those from the build direction due to the strengthening effect of a large number of vertical β grain boundaries, but the elongation was 69.7% lower than that of the build direction due to the uneven grain deformation of β grains.

Impact on dietary intake of a self-directed, gender-tailored diabetes prevention program in men

AIMTo investigate changes in dietary intake following a 6-mo randomised controlled trial of the self-directed, gender-tailored type 2 diabetes mellitus (T2DM) Prevention Using LifeStyle education (PULSe) program in men. MeTHODSMen aged 18-65 years, with a body mass index (BMI) 25-40 kg/m2, and at high risk for developing T2DM were recruited from the Hunter Region of New South Wales, Australia. eligible participants were randomised into one of two groups: (1) waitlist control; or (2) PULSe intervention. Dietary intake was assessed at baseline and immediately post-program using the Australian eating Survey food frequency questionnaire and diet quality measured using the Australian Recommended Food Score (ARFS). ReSULTSOne hundred and one participants (n = 48, control; n = 53, intervention, mean age 52.3 ± 9.7 years, BMI of 32.6 ± 3.3 kg/m2) commenced the study. Following the active phase, differences between groups were observed for proportion of total energy consumed from healthful (core) foods (+7.6%eI, P energy-dense, nutrient-poor foods (-7.6%eI, P et quality (ARFS) (+4.3, P = 0.004), including sub-scales for fruit (+1.1, P = 0.03), meat (+0.9, P = 0.004) and non-meat protein (+0.5, P = 0.03). CONCLUSIONThe PULSe prevention program’s nutrition messages led to significant improvements in dietary intake in men at risk of T2DM.

A Modified Crank-Nicolson Numerical Scheme for the Flory-Huggins Cahn-Hilliard Model

In this paper we propose and analyze a second order accurate numericalscheme for the Cahn-Hilliard equation with logarithmic Flory Huggins energy potential. A modified Crank-Nicolson approximation is applied to the logarithmic nonlinear term, while the expansive term is updated by an explicit second order AdamsBashforth extrapolation, and an alternate temporal stencil is used for the surface diffusion term. A nonlinear artificial regularization term is added in the numerical scheme,which ensures the positivity-preserving property, i.e., the numerical value of the phasevariable is always between -1 and 1 at a point-wise level. Furthermore, an unconditional energy stability of the numerical scheme is derived, leveraging the special formof the logarithmic approximation term. In addition, an optimal rate convergence estimate is provided for the proposed numerical scheme, with the help of linearizedstability analysis. A few numerical results, including both the constant-mobility andsolution-dependent mobility flows, are presented to validate the robustness of the proposed numerical scheme.

As a greater proportion of VLBW and eLBW babies survive, the population of babies at risk increases. In various regions of the world, different identification criteria are used to determine which babies are at risk of blindness in order to provide timely diagnostic examinations and treatment as needed. Methods for preventiing ROP include better ante-natal and obstetric care leading to a reduction in the rate of prematurity, the use of ante-natal corticosteroids, and better neonatal care practices. Recent developments have indicated that management of oxygen supplementation is important for the prevention of severe ROP; however,there is not yet known what oxygen saturation target should be adopted. Sepsis increases severe ROP in very preterm infants. Genetic associations and a telemedicine approach may be explored to detect ROP. Cholangiocarcinoma is a rare cancer originating from the neoplastic transformation of the epithelial cells (i.e. cholangiocytes) that line the biliary tract. The prognosis for patients with cholangiocarcinoma is grim due to lack of viable treatment options. The increase in world-wide incidence and mortality from cholangiocarcinoma highlights the importance of understanding the intracellular mechanisms that trigger the neoplastic transformation of cholangiocytes and the growth of biliary cancers. The purpose of the following review is to address what has been learned over the past decade concerning the molecular basis of cholangiocarcinogenesis. The material presented is divided into two sections:(1) mechanisms regulating neoplastic transformation of cholangiocytes; and (2) factors regulating cholangiocarcinoma growth. An understanding of the growth regulatory mechanisms of cholangiocarcinoma will lead to the identification of therapeutic targets for this devastating cancer.

Dual-energy X-ray absorptiometry scans accurately predict differing body fat content in live sheep

Background: There is considerable interest in implementing mobile scanning technology for on-farm body composition analysis on live *** experiments evaluated the use of dual energy X-ray absorptiometry(DXA) as an accurate method of total body fat measurement in live ***: In exp.1,visceral and whole body fat analysis was undertaken in sheep with body condition scores(BCS)in the range 2 to 3.25(scale 1: thin to 5: fat).The relationship of BCS was moderately correlated with visceral fat depot mass(r = 0.59,P e body fat(r = 0.70,P exp.2,sheep with BCS in the range 2.25 to 3.75 were blood sampled to analyse circulating leptin concentrations,and were DXA scanned immediately post mortem for total body *** leptin concentrations had low correlations with BCS(r = 0.50,P elation with chemical body fat(r = 0.17,P > 0.05,n = 9).There was a moderate correlation between DXA body fat and BCS(r = 0.70,P elated with chemical body fat(r = 0.81,P exp.3,a series of five DXA scans,at 8-week intervals,was performed on growing sheep over a 32-week *** average BCS ranged from 2.39 ± 0.07(S.e.M.) to 3.05 ± 0.11 and the DXA body fat(%) ranged from 16.8 ± 0.8 to 24.2 ± *** was a moderate correlation between DXA body fat and BCS over the 32 weeks(r = 0.61,P erall,these experiments indicated that there was good agreement between BCS,DXA and chemical analysis for measuring total body fat in sheep,and that DXA scanning is a valid method for longitudinal measurement of total body fat in live sheep.

Hypoxia promotes growth of stem cells in dental follicle cell populations

Adult stem cells (ASC) have been found in many tis-sues and are of great therapeutic potential due to their capability of differentiation. However, ASC comprise only a small fraction of the tissues. In order to use ASC for therapeutic purposes, it is important to obtain relatively pure stem cells in large quantities. Current methods for stem cell purification are mainly based on marker-dependent cell sorting techniques, which have various technical difficulties. In this study, we have attempted to develop novel conditions to favor the growth of the dental follicle stem cells (DFSC) such that the resultant cell populations are enriched in stem cells. Specifically, a heterogeneous dental follicle cell (H-DFC) population containing stem cells and homogenous non-stem cell dental follicle cell population were cultured at 1% or 5% hypoxic conditions. Only the heterogeneous population could increase proliferation in the hypoxic condition whereas the homogenous DFC did not change their proliferation rate. In addition, when the resultant cells from the heterogonous population were subjected to differentiation, they appeared to have a higher capacity of adipogenesis and osteogenesis as compared to the controls grown in the normal at-mosphere (normoxic condition). These hypoxia- treated cells also express higher levels of some stem cell markers. Together, these data suggest that stem cells are enriched by culturing the heterogeneous cell populations in a reduced O2 condition.
