Identification and characterization of genes that control fat deposition in chickens

Background： Fat deposits in chickens contribute significantly to meat quality attributes such as juiciness, flavor, taste and other organoleptic properties. The quantity of fat deposited increases faster and earlier in the fast- growing chickens than in slow-growing chickens. In this study, Affymetrix Genechip~ Chicken Genome Arrays 32773 transcripts were used to compare gene expression profiles in liver and hypothalamus tissues of fast-growing and slow-growing chicken at 8 wk of age. Real-time RT-PCR was used to validate the differential expression of genes selected from the microarray analysis. The mRNA expression of the genes was further examined in fat tissues. The association of single nucleotide polymorphisms of four lipid-related genes with fat traits was examined in a F2 resource population. Results： Four hundred genes in the liver tissues and 220 genes hypothalamus tissues, respectively, were identified to be differentially expressed in fast-growing chickens and slow-growing chickens. Expression levels of genes for lipid metabolism （SULTIB1, ACSBG2, PNPLA3, LPL, AOAH） carbohydrate metabolism （MGAT4B, XYLB, GBE1, PGM1, HKDCl）cholesttrol biosynthesis （FDPS, LSS, HMGCR, NSDHL, DHCR24, IDI1, MEI） HSD17B7 and other reaction or pro- cesses （CYPIA4, CYP1A1, AKR1BI, CYP4V2, DDO） were higher in the fast-growing White Recessive Rock chickens than in the slow-growing Xinghua chickens. On the other hand, expression levels of genes associated with multicellular organism development, immune response, DNA integration, melanin biosynthetic process, muscle organ develop- ment and oxidation-reduction （FRZB, DMD, FUT8, CYP2C45, DHRSX, and CYP2C18） and with glycol-metabolism （GCNT2, ELOVL d, and FASN）, were higher in the XH chickens than in the fast-growing chickens, fiT-PCR validated high expression levels of nine out of 12 genes in fat tissues. With the aim of studying the effect of water dynamics on the properties of biological systems, in this paper, we present a quasi-elastic neutron scattering study on three different types of living cells, differing both in their morphological and tumor properties. The measured scattering signal, which essentially originates from hydrogen atoms present in the investigated systems, has been analyzed using a global fitting strategy using an optimized theoretical model that considers various classes of hydrogen atoms and allows disentangling diffusive and rotational motions. The approach has been carefully validated by checking the reliability of the calculation of parameters and their 99% confidence intervals. We demonstrate that quasi-elastic neutron scattering is a suitable experimental technique to characterize the dynamics of intracellular water in the angstrom/picosecond space/time scale and to investigate the effect of water dynamics on cellular biodiversity.

本研究旨在编制适合我国儿童青少年的校园欺凌保护行为问卷,从而阐明在中国文化背景下保护行为的多样性。研究分两个子研究,研究1以文献分析以及开放式问卷调查结果为基础建构理论模型并搜集行为样例,初步确定测量校园欺凌中儿童青少年保护行为的条目。在此基础上,通过对824名(Mage=11.25)儿童青少年施测,进行项目分析和因素分析,确定问卷的最终条目。研究2对1086名(Mage=11.18)儿童青少年进行测试,检验问卷的信度和效度。研究1的结果表明,该问卷最终确定24个条目,包括果断型保护(assertive defending)、攻击型保护(aggressive defending)、安慰受欺凌者(comforting victims)、向权威人士报告(reporting to authority)以及计策型保护(tactical defending)五个因子。五因素模型拟合良好,χ(166)=350.76,p<.001,CFI=.973,TLI=.954,SRMR=.020,RMSEA=.037,90%CI=[.031,.042]。研究2验证了该问卷的信度(α=.78-.93)及其结构效度和效标关联效度。五种保护行为与认知和情感共情、攻击性、社交能力、同伴喜爱和受欺负有不同程度的相关。具体来说,果断型保护行为与认知共情、社交能力呈显著正相关;攻击型保护行为与认知共情、同伴喜爱、社交能力呈显著负相关,与攻击性、受欺负呈显著正相关;安慰受欺凌者与认知共情、情感共情、同伴喜爱和社交能力呈显著正相关,与攻击性呈显著负相关;向权威人士报告与认知共情呈显著正相关;计策型保护行为与受欺负呈显著正相关,与认知共情呈显著负相关。本研究结果表明,中国儿童青少年保护行为包含五个因子,研究所编制的问卷具有良好的信度和效度,可用于校园欺凌事件中儿童青少年保护行为的测评。

The use of plastics from petrochemical resources poses environmental impacts, and one of the alternative solutions is the use of starch. The objective of this present work has been to present the literature on starch, and to highlight the debate in the development of composite films. The approach adopted was to present the state of the art on starch and thermoplastic starch matrix composites. The work shows that starch is available worldwide and can be used in the manufacture of biodegradable plastics;the debate remains on the reinforcement of thermoplastic starch to improve its physical and mechanical properties poor;then researchers must diversify the reinforcements to see the impact on the properties of thermoplastic starch.
