Multiple myeloma(MM) is still an incurable hematologic malignancy, which is eagerly to the discovery of novel therapeutic targets and methods. N-acetyltransferase 10(NAT10) is the first reported regulator of mRNA acetylation that is activated in many cancers. However, the function of NAT10 in MM remains unclear. We found significant upregulation of NAT10 in MM patients compared to normal plasma cells, which was also highly correlated with MM poor outcome. Further enforced NAT10 expression promoted MM growth in vitro and in vivo, while knockdown of NAT10 reversed those effects. The correlation analysis of acetylated RNA immunoprecipitation sequencing(ac RIP-seq) and ribosome profiling sequencing(Ribo-seq) combined with RIP-PCR tests identified centrosomal protein 170(CEP170) as an important downstream target of NAT10. Interfering CEP170 expression in NAT10-OE cells attenuated the acceleration of cellular growth caused by elevated NAT10. Moreover,CEP170 overexpression promoted cellular proliferation and chromosomal instability(CIN) in ***, remodelin, a selective NAT10 inhibitor, suppressed MM cellular growth, induced cellular apoptosis in vitro and prolonged the survival of 5TMM3VT mice in vivo. Collectively, our data indicate that NAT10 acetylates CEP170 mRNA to enhance CEP170 translation efficiency, which suggests that NAT10 may serve as a promising therapeutic target in MM.

Xanthomonas spp. cause severe bacterial diseases. However, effective strategies for prevention and management of these diseases are scarce. Thus, it is necessary to improve the efficiency of control of diseases caused by Xanthomonas. In this study, Xanthomonas oryzae pv. oryzae(Xoo), which causes rice bacterial leaf blight, has been studied as a representative. A transposon insertion library of Xoo, comprising approximately 200,000 individual insertion mutants, was generated. Transposon sequencing data indicated that the mariner C9 transposase mapped at 35.7–36.4% of all potential insertion sites, revealing 491 essential genes required for the growth of Xoo in rich media. The results show that, compared to the functions of essential genes of other bacteria, the functions of some essential genes of Xoo are unknown, 25 genes might be dangerous for the Xanthomonas group, and 3 are specific to Xanthomonas. High-priority candidates for developing broad-spectrum, Xanthomonas-specific, and environment-friendly bactericides were identified in this study. In addition, this study revealed the possible targets of dioctyldiethylenetriamine using surface plasmon resonance(SPR) in combination with high performance liquid chromatography–mass spectrometry(HPLC–MS). The study also provided references for the research of some certain bactericides with unknown anti-bacterial mode of action. In conclusion, this study urged a better understanding of Xanthomonas,provided meaningful data for the management of bacterial leaf blight, and disclosed selected targets of a novel bactericide.

Growth and Development of Callosobruchus chinensis L.(Coleoptera:Bruchidae) on Different Beans

[ Objective ] The paper was to study the growth and development characteristics of Callosobruchus chinensis L. on different beans, and to explore the resistance of different beans to C. chiaensis. [ Method] The egg hatchability, growth and development of larva and adult of C. chinensis L. on different beans were observed in the laboratory at 30℃. [ Result] All the eight kinds of beans were spawned, but there were differences in egg hatchability among different beans. The egg hatchahility on Vigna radiate was the highest of （78.47 ±6.70） %. After hatching, larvae only drilled into grains of V. radiate, Vigna umbellate, Vicia faba, Pisum sativum and Cicer arietiaum, but there were significant differences in generation survival rate. The generation survival rate on V. radiate was the highest of （68.75 ±9.08）% and that on 1/. faba was the lowest of （26.93± 10.28 ）%. The development duration of C. chinensis on V. radiate was the shortest of （34.78 ± 2.02）d; the intrinsic rate of increase and finite rate of increase were greater of 0. 041 09 and 1. 041 95, respectively. [ Conclusion] A large number of C. chinertsis could be reproduced within a short period if feeding on V. radiate.

Schistosomiasis is one of the most prevalent parasitic diseases in China, and hepatic fibrosis caused by schistosome infection is the principal cause of death. The aim of this study was to evaluate the efficacy of NPll-4- derived immunotoxin scFv-artesunate on Schistosoma japonicum-induced hepatic fibrosis. A single-chain variable fragment （scFv） was generated from the murine anti-Schistosoma japonicum （S. japanicum） monoclonal antibody NP11-4. The scFv was expressed as a soluble protein and purified by Ni-affinity chromatography. After conjuga- tion with artesunate, the binding ability with soluble egg antigens （SEA） was determined by an enzyme-linked immunosorbent assay （ELISA）. The biological activity of purified scFv, scFv-artesunate （immunotoxin）, and artesunate was detected in vivo. Image-Pro Plus software was used to analyze the size of egg granuloma and the extent of liver fibrosis. The recombinant scFv expession vector was constructed and expressed successfully. After purification by a His-trap Ni-affinity column, the scFv yield was approximately 0.8 mg/L of culture medium. ELISA results showed that chemical conjugation did not affect the binding activity of the immunotoxin. Our animal experiments indicated that the immunotoxin could significantly reduce the size of egg granuloma in the liver and inhibit hepatic fibrosis. The immunotoxin could be used as a promising candidate in the targeted therapy of S. .japonicum-induced hepatic fibrosis.

Screening of Fungicides against Broad Bean Chocolate Spot(Botrytis fabae)

[ Objective] The paper was to select the effective fungicides to control broad bean chocolate spot. [ Method] Seven different fungicides were selected and sprayed in experimental fields in Jiangsu Yanjiang Area Agricultural Science Research Institute of China in 2012, and yield increase rate and control effect were calculated. [ Result] The control effects of mancozeb 80% WP and pyraclostrobin ＋ metiram 60% WDG against broad bean chocolate spot were 80.88% - 89.26%, and the yield increase rates were 10.85% -41.73%. Mancozeb 80% WP and pyraclostrobin ＋ metiram 60% WG should have beautiful prospect against broad bean chocolate spot owing to excellent control effect and increase yield. [ Conclusion] The study provided reference for field control against broad bean choc- olate spot.

Pest damage is a significant factor that cuts agricultural production, and cultivation of pest-resistant varieties is the most economic and effective approach of controlling pest damage. Plant pest resistance includes antixenosis, antibiosis, and tolerance. Current popular methods for the genetic improvement of crop pest resistance are traditional breeding method, transgenic breeding, and molecular mark-assisted breeding. This paper introduced major mechanisms and genetic bases of plant pest resistance, ＂and reviewed research progress of domestic and international genetic improvement of plant pest resistance, analyzed the problems and its de- velopment prospects.

Field Efficacy Trials of 10% Cyantraniliprole OD against Phytomyza horticola Gourean

In order to screen effective insecticides against Phytomyza horticola(Gourean), a comparative trial on the control efficacy of 10%cyantraniliprole OD and 75% cyromazine WP was conducted, and the optimal dose of 10% cyantraniliprole OD was determined. The results showed that 10% cyantraniliprole OD sprayed at the dosages of 375-450 mL/hm^2 had good control effect on P. horticola, which also increased the yield of fresh pea pods. At 15 d post spraying, the control effects of 10% cyantraniliprole OD against P. horticola were 81.98%-86.12% when sprayed at the dosages of 375-450 mL/hm^2. The yield increase rates of 10% cyantraniliprole OD were 8.39%-13.46%.
