Strong orthogonal arrays(SOAs) were recently introduced and studied as a class of spacefilling designs for computer experiments. To surely realize better space-filling properties, SOAs of strength three or higher are desirable. In addition, orthogonality is also an important property for designs of computer experiments, because it guarantees that the estimates of the main effects are uncorrelated. This paper first provides a systematic study on the construction of(nearly) orthogonal strength-three SOAs with better space-filling properties. The newly proposed strength-three SOAs enjoy almost the same space-filling properties of strength-four SOAs, and can accommodate much more columns than the latter. Moreover, they are(nearly) orthogonal and flexible in run sizes. The construction methods are straightforward to implement, and their theoretical supports are well established. In addition to the theoretical results, many designs are tabulated for practical needs.

Mepanipyrim induces cardiotoxicity of zebrafish(Danio rerio) larvae via promoting AhR-regulated COX expression pathway

The wide use of pesticides has seriously threatened human health and the survival of beneficial organisms. The fungicide mepanipyrim is widely used in viticulture practices. Studies of mepanipyrim-induced toxicity in organisms are still scarce, especially studies on cardiotoxicity. In this study, we aimed to investigate mepanipyrim-induced cardiotoxicity in zebrafish(Danio rerio) larvae. We found that mepanipyrim could induce cardiotoxicity by altering the heart rate and cardiomyocyte diameter of larvae. Meanwhile, RNA sequencing and RT-qPCR data indicated that mepanipyrim exposure could dramatically alter the m RNA expression of calcium signaling pathway-, cardiac muscle contraction-, and oxidative respiratory chain-related genes. Interestingly, by the CALUX cell bioassay, we found that most cytochrome c oxidase(COX) family genes exhibited potential AhR-regulated activity, suggesting that mepanipyrim induced cardiotoxicity via a novel AhR-regulated manner in larvae.Additionally, the AhR antagonist CH_(2)23191 could effectively prevent mepanipyrim-induced cardiotoxicity in zebrafish larvae. In conclusion, the AhR agonist mepanipyrim could induce cardiotoxicity in a novel unreported AhR-regulated manner, which could specifically affect the expression of COX family genes involved in the mitochondrial oxidative respiratory chain. Our data will help explain the toxic effects of mepanipyrim on organisms and provide new insight into the AhR agonistic activity pesticide-induced cardiotoxicity.

Neburon is a phenylurea herbicide that is widely used worldwide,but its toxicity is poorly studied.In our previous study,we found that neburon has strong aryl hydrocarbon receptor(AhR)agonist activity,but whether it causes reproductive toxicity is not clear.In the present study,zebrafish were conducted as a model organism to evaluate whether environmental concentrations of neburon(0.1,1 and 10μg/L)induce reproductive disorder in males.After exposure to neburon for 150 days from embryo to adult,that the average spawning egg number in high concentration group was 106.40,which was significantly lower than 193.00in control group.This result was mainly due to the abnormal male reproductive behavior caused by abnormal transcription of genes associated with reproductive behavior in the brain,such as secretogranin-2a.The proportions of spermatozoa in the medium and high concentration groups were 82.40%and 83.84%,respectively,which were significantly lower than 89.45%in control group.This result was mainly caused by hormonal disturbances and an increased proportion of apoptotic cells.The hormonal disruption was due to the significant changes in the transcription levels of key genes in the hypothalamus-pituitary-gonadal axis following neburon treatment.Neburon treatment also significantly activated the AhR signaling pathway,causing oxidative stress damage and eventually leading to a significant increase in apoptosis in the exposed group.Together,these data filled the currently more vacant profile of neburon toxicity and might provide information to assess the ecotoxicity of neburon on male reproduction at environmentally relevant concentrations.

Antibody therapeutics and vaccines for coronavirus disease 2019(COVID-19)have been approved in many countries,with most being developed based on the original strain of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).SARS-CoV-2 has an exceptional ability to mutate under the pressure of host immunity,especially the immune-dominant spike protein of the virus,which is the target of both antibody drugs and vaccines.Given the continuous evolution of the virus and the identification of critical mutation sites,the World Health Organization(WHO)has named 5 variants of concern(VOCs):4 are previously circulating VOCs,and 1 is currently circulating(Omicron).Due to multiple mutations in the spike protein,the recently emerged Omicron and descendent lineages have been shown to have the strongest ability to evade the neutralizing antibody(NAb)effects of current antibody drugs and vaccines.The development and characterization of broadly neutralizing antibodies(bNAbs)will provide broad strategies for the control of the sophisticated virus SARS-CoV-2.In this review,we describe how the virus evolves to escape NAbs and the potential neutralization mechanisms that associated with bNAbs.We also summarize progress in the development of bNAbs against SARS-CoV-2,human coronaviruses(CoVs)and other emerging pathogens and highlight their scientific and clinical significance.

A series of novel atropisomeric diphosphine ligands termed Tan Phos were designed and synthesized,which has a smaller bite angle compared with that of other ligands such as BINAP.Tan Phos showed high reactivity and enantioselectivity in the rhodiumcatalyzed asymmetric hydrogenation ofα-dehydro amino ketones,and up 99%yield and 99%ee were obtained for a wide range of chiralα-amino ketones.

The abnormal regulation of alternative splicing is usually accompanied by the occurrence and development of tumors,which would produce multiple different isoforms and diversify protein expression.The aim of the present study was to conduct a systematic review in order to describe the regulatory mechanisms of alternative splicing,as well as its functions in tumor cells,from proliferation and apoptosis to invasion and metastasis,and from angiogenesis to metabolism.The abnormal splicing events contributed to tumor progression as oncogenic drivers and/or bystander factors.The alterations in splicing factors detected in tumors and other mis-splicing events(i.e.,long non-coding and circular RNAs)in tumorigenesis were also included.The findings of recent therapeutic approaches targeting splicing catalysis and splicing regulatory proteins to modulate pathogenically spliced events(including tumor-specific neo-antigens for cancer immunotherapy)were introduced.The emerging RNA-based strategies for the treatment of cancer with abnormally alternative splicing isoforms were also discussed.However,further studies are still required to address the association between alternative splicing and cancer in more detail.

The main defense response to Soybean mosaic virus(SMV)infection in soybean[Glycine max(L.)Merr.]is thought to be blockage of intercellular virus transport by callose deposition on plasmodesmata.But the specific regulatory mechanism remains largely unknown.In this study,we found that hydrogen peroxide(H_(2)O_(2))signal downstream of NO was associated with the regulation of callose accumulation.Abundant H_(2)O_(2)was produced on the cell membrane and cell wall in the incompatible combination of soybean cultivar Jidou 7 and SMV strain N3,whereas no obvious H_(2)O_(2)was observed in the compatible combination of Jidou 7 and strain SC-8.When H_(2)O_(2)production was inhibited,callose accumulation induced by SMV infection decreased to a level insufficient to restrict virus transport in the incompatible combination.The H_(2)O_(2)-associated transcriptome dynamics of soybean during SMV infection was investigated.Transcriptome and functional analysis using virus-induced gene silencing showed that Gm SEOB and Gm PAP27,two genes regulated by H_(2)O_(2),functioned in resistance by positively regulating the accumulation of callose in response to SMV infection.These results lay a foundation for further research on the signal transduction and molecular regulation of callose deposition during soybean resistance to SMV infection.

Hydrogel patch-based stem cell transplantation and microenvironment-regulating drug delivery strategy is promising for the treatment of myocardial infarction(MI).However,the low retention of cells and drugs limits their therapeutic efficacies.Here,we propose a prefixed sponge carpet strategy,that is,aldehyde-dextran sponge(ODS)loading anti-oxidative/autophagy-regulating molecular capsules of 2-hydroxy-β-cyclodextrin@resveratrol(HP-β-CD@Res)is first bonded to the rat’s heart via capillary removal of interfacial water from the tissue surface,and the subsequent Schiff base reaction between the aldehyde groups on ODS and amino groups on myocardium tissue.Then,an aqueous biocompatible hydrazided hyaluronic acid(HHA)solution encapsulating mesenchymal stem cells(MSCs)is impregnated into the anchored carpet to form HHA@ODS@HP-β-CD@Res hydrogel in situ via click reaction,thus prolonging the in vivo retention time of therapeutic drug and cells.Importantly,the HHA added to outer surface consumes the remaining aldehydes to contribute to nonsticky top surface,avoiding adhesion to other tissues.The embedded HP-β-CD@Res molecular capsules with antioxidant and autophagy regulation bioactivities can considerably improve cardiac microenvironment,reduce cardiomyocyte apoptosis,and enhance the survival of transplanted MSCs,thereby promoting cardiac repair by facilitating angiogenesis and reducing cardiac fibrosis.

Electrolyte design strategies towards long-term Zn metal anode for rechargeable batteries

Rechargeable Zinc(Zn)batteries exhibit great potentials as alternative energy storage devices due to their high safety,low cost,and environmental friendliness.However,the long-standing issues of low Coulombic efficiency(CE)and poor cycle stability of Zn anode,derived from dendrite,H_(2)evolution,and passivation are directly related to their thermodynamic instability in aqueous electrolyte,severely shorten the battery's cycle life.Recently reported electrolyte design strategies,which have made great progress to address Zn metal anode problems,are summarized into two categories,that is,aqueous electrolytes about cation-water interaction controlling and interface adjusting,and novel types of electrolytes towards less water,non-aqueous solvents,even no solvents.The final section shows the brief comparisons,including failure mechanisms of electrolyte exhaustion and short circuit for aqueous and nonaqueous electrolyte based full cells respectively,and possible perspectives for future research.
