Cell walls in plants,particularly forest trees,are the major carbon sink of the terrestrial ecosystem.Chemical and biosynthetic features of plant cell walls were revealed early on,focusing mostly on herbaceous model species.Recent developments in genomics,transcriptomics,epigenomics,transgenesis,and associated analytical techniques are enabling novel insights into formation of woody cell walls.Here,we review multilevel regulation of cell wall biosynthesis in forest tree species.We highlight current approaches to engineering cell walls as potential feedstock for materials and energy and survey reported field tests of such engineered transgenic trees.We outline opportunities and challenges in future research to better understand cell type biogenesis for more efficient wood cell wall modification and utilization for biomaterials or for enhanced carbon capture and storage.

Objective:To scrutinize the definitions of minimal invasive surgical therapy(MIST)and to investigate urologists’knowledge,attitudes,and practices for benign prostatic obstruction surgeries.Methods:A 36-item survey was developed with a Delphi method.Questions on definitions of MIST and attitudes and practices of benign prostatic obstruction surgeries were included.Urologists were invited globally to complete the online survey.Consensus was achieved when more than or equal to 70%responses were“agree or strongly agree”and less than or equal to 15%responses were“disagree or strongly disagree”(consensus agree),or when more than or equal to 70%responses were“disagree or strongly disagree”and less than or equal to 15%responses were“agree or strongly agree”(consensus disagree).Results:The top three qualities for defining MIST were minimal blood loss(n=466,80.3%),fast post-operative recovery(n=431,74.3%),and short hospital stay(n=425,73.3%).The top three surgeries that were regarded as MIST were Urolift®(n=361,62.2%),Rezum®(n=351,60.5%),and endoscopic enucleation of the prostate(EEP)(n=332,57.2%).Consensus in the knowledge section was achieved for the superiority of Urolift®,Rezum®,and iTIND®over transurethral resection of the prostate with regard to blood loss,recovery,day surgery feasibility,and post-operative continence.Consensus in the attitudes section was achieved for the superiority of Urolift®,Rezum®,and iTIND®over transurethral resection of the prostate with regard to blood loss,recovery,and day surgery feasibility.Consensus on both sections was achieved for EEP as the option with the better symptoms and flow improvement,lower retreatment rate,and better suitable for prostate more than 80 mL.Conclusion:Minimal blood loss,fast post-operative recovery,and short hospital stay were the most important qualities for defining MIST.Urolift®,Rezum®,and EEP were regarded as MIST by most urologists.

Diabetes has become a global concern at present,among which type 2 diabetes mellitus(T2DM)accounts for approximately 90%-95%of patients.T2DM is a type of metabolic disorder syndrome that results from a genetic defect,and it is based on insulin resistance and an insulin secretion disorder.The occurrence of T2DM is usually the outcome of both genetic and environmental factors and their interactions.The etiology and pathogenesis of diabetes have not been fully elucidated,and no radical therapeutic cure has been found.Patients with T2DM suffer from complications such as the development of a chronic hyperglycemic condition and even serious metabolic disorders and organ damage in the body and depression and dementia,in addition to other chronic complications.Many studies have suggested that diet is crucial in the development of diabetes and the control of blood glucose.Natural substances have the characteristics of low toxicity and few side effects and may be key to the development of diabetic health products and preventive treatments.This paper reviews the etiology,pathogenesis,risks,treatments and diets related to T2DM to summarize the types of recently available natural products,from both local and foreign sources,for lowering blood glucose at home and their application in supplementary hypoglycemic foods.The key findings and conclusions suggest that there are various known T2DM-inducing factors,including genetic and environmental factors and three types of hybrid factors.

We report in this study the identification of a natural product-like antagonist(1a) of Vps34 as a potent autophagy modulator via structure-based virtual screening. Aurone derivative 1a strongly inhibited Vps34 activity in cell-free and cell-based assays. Significantly, 1a prevents autophagy in human cells induced either by starvation or by an mT OR inhibitor. In silico modeling and kinetic data revealed that 1a could function as an ATP-competitive inhibitor of Vps34. Moreover, it suppressed autophagy in vivo and without inducing heart or liver damage in mice. 1a could be utilized as a new motif for more selective and efficacious antagonists of Vps34 for the potential treatment of autophagy-related human diseases.

Cyclin-dependent kinase 9(CDK9) activity is correlated with worse outcomes of triplenegative breast cancer(TNBC) patients. The heterodimer between CDK9 with cyclin T1 is essential for maintaining the active state of the kinase and targeting this protein-protein interaction(PPI) may offer promising avenues for selective CDK9 inhibition. Herein, we designed and generated a library of metal complexes bearing the 7-chloro-2-phenylquinoline CN ligand and tested their activity against the CDK9cyclin T1 PPI. Complex 1 bound to CDK9 via an enthalpically-driven binding mode, leading to disruption of the CDK9-cyclin T1 interaction in vitro and in cellulo. Importantly, complex 1 showed promising anti-metastatic activity against TNBC allografts in mice and was comparably active compared to cisplatin. To our knowledge, 1 is the first CDK9-cyclin T1 PPI inhibitor with anti-metastatic activity against TNBC. Complex 1 could serve as a new platform for the future design of more efficacious kinase inhibitors against cancer, including TNBC.

【目的】探究壮通饮通过Nrf2-SCL7A11/xCT-Gpx4通路调控铁死亡对大脑中动脉栓塞模型小鼠脑缺血再灌注损伤的保护作用及其机制。【方法】将C57BL/6J小鼠随机分为假手术组（Sham）、模型组（MCAO）、低剂量壮通饮组（ZTY-L+MCAO），高剂量壮通饮组（ZTY-H+MCAO），每组5只。其中MCAO组采用硅胶线栓法进行造模，小鼠大脑中动脉栓塞1h，拔出线栓再灌注72h后进行心脏灌注取脑，Sham组除了不插入线栓其余操作同MCAO组。采用Zea-Longa法对小鼠神经功能进行评分；采用TTC染色评估小鼠脑梗死体积；采用HE染色、尼氏染色评估脑损伤程度；采用普鲁士蓝染色评价三价铁离子蓄积水平；qPCR检测铁离子转运相关载体受体TfR1和DMT1、脂质过氧化相关基因ACSL4以及铁死亡标志物PTGS2 mRNA表达；ELISA试剂盒检测4-HNE的含量评估小鼠大脑脂质过氧化水平；Western-blot、免疫荧光检测小鼠脑组织中Nrf2、SCL7A11/xCT、Gpx4蛋白水平。【结果】(1)壮通饮改善了MCAO/R后小鼠神经功能（P<0.05）和大脑梗死体积（P<0.05）及缓解MCAO/R后大脑皮层细胞病理损伤。(2)壮通饮减弱了MCAO/R后小鼠大脑组织的三价铁离子蓄积状态；减少MCAO/R后小鼠大脑中与铁离子转运入细胞内相关载体TfR1和DMT1 mRNA的表达（P<0.001）；下调MCAO/R后小鼠大脑脂质过氧化物4-HNE含量和组织脂质过氧化物酶ACSL4 mRNA表达（P<0.001）；降低MCAO/R后小鼠大脑铁死亡标志物PTGS2 mRNA表达（P<0.001）。(3)壮通饮增加MCAO/R后Nrf2入核表达（P<0.001），以及增加xCT和Gpx4在神经元中的表达（P<0.001）。【结论】壮通饮能够改善MCAO/R小鼠神经功能和脑梗死体积，缓解大脑皮层细胞病理损伤，影响Nrf2-SCL7A11/xCT-Gpx4通路关键信号分子表达，所以壮通饮改善小鼠MCAO/R损伤的作用机制可能是通过Nrf2-SCL7A11/xCT-Gpx4通路调节铁死亡水平达到的。

山东淄博峨庄镇后紫峪剖面寒武系张夏组顶部发育树形石,根据宏观露头岩性变化所反映的沉积相特征,研究区张夏组层序可以划分为三个四级亚层序。随着微生物影响树形石的形成过程的证据不断丰富,广为接受的“CCM机制引发蓝细菌鞘钙化作用”的观点正不断受到挑战。本文通过野外勘探采样、室内薄片分析相结合的方式,镜下识别刻画树形石的组构,并与现代实例进行类比进而分析其成因机制。树形石的主要组构为暗色泥晶基质、亮晶方解石胶结物和碎屑颗粒,显微镜下显示,树形石内部发育钙化蓝细菌生物化石,包括附枝菌(Epiphyton)、基座菌(Hedstroemia)、肾形菌(Renalcis),还可见孢网菌(Bacinella)、石松藻(Lithocodium)状组构。通过对研究区树形石中多种类型钙化蓝细菌和钙化蓝细菌微生物膜钙化残余物质的研究,为树形石内钙化微生物多样性做出了较为翔实的补充,明确了树形石是附枝菌等蓝细菌主导的微生物膜或微生物席钙化作用的产物,其中胞外聚合物(EPS)在树形石的发育和矿物沉淀过程中起关键作用,为树形石的“微生物影响成因”提供了一个典型实例。

BACKGROUND Chronic liver disease(CLD)was associated with adverse clinical outcomes among people with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection.AIM To determine the effects of SARS-CoV-2 infection on the incidence and treatment strategy of hepatocellular carcinoma(HCC)among patients with CLD.METHODS A retrospective,territory-wide cohort of CLD patients was identified from an electronic health database in Hong Kong.Patients with confirmed SARS-CoV-2 infection[coronavirus disease 2019(COVID-19)+CLD]between January 1,2020 and October 25,2022 were identified and matched 1:1 by propensity-score with those without(COVID-19-CLD).Each patient was followed up until death,outcome event,or November 15,2022.Primary outcome was incidence of HCC.Secondary outcomes included all-cause mortality,adverse hepatic outcomes,and different treatment strategies to HCC(curative,non-curative treatment,and palliative care).Analyses were further stratified by acute(within 20 d)and post-acute(21 d or beyond)phases of SARS-CoV-2 infection.Incidence rate ratios(IRRs)were estimated by Poisson regression models.RESULTS Of 193589 CLD patients(>95%non-cirrhotic)in the cohort,55163 patients with COVID-19+CLD and 55163 patients with COVID-19-CLD were included after 1:1 propensity-score matching.Upon 249-d median follow-up,COVID-19+CLD was not associated with increased risk of incident HCC(IRR:1.19,95%CI:0.99-1.42,P=0.06),but higher risks of receiving palliative care for HCC(IRR:1.60,95%CI:1.46-1.75,P<0.001),compared to COVID-19-CLD.In both acute and post-acute phases of infection,COVID-19+CLD were associated with increased risks of allcause mortality(acute:IRR:7.06,95%CI:5.78-8.63,P<0.001;post-acute:IRR:1.24,95%CI:1.14-1.36,P<0.001)and adverse hepatic outcomes(acute:IRR:1.98,95%CI:1.79-2.18,P<0.001;post-acute:IRR:1.24,95%CI:1.13-1.35,P<0.001),compared to COVID-19-CLD.CONCLUSION Although CLD patients with SARS-CoV-2 infection were not associated with increased risk of HCC,they were more likely to receive palliative treatment

The development of transition metal complex-based luminescent chemosensors has recently aroused increasing interest for protein biomarker labelling and detection,especially for the real-time diagnosis and treatment of disease.This is owing to their unique photophysical properties,particularly their long-lived and environmentally sensitive emission,which can be easily controlled via the structural modification of ligands.In this overview,we highlight recent examples of protein biomarker detection using group 8–9 metal-based luminescent chemosensors,including the frequently employed ruthenium(II)and iridium(III)complexes.Various mechanisms and sensing modes are described and compared,and the outlook and future directions of this field are discussed as well.
