In recent years, studies on evaluation of the therapeutic and toxic activity of herbal medicinal products became available and popular. The advances in modern biotechnology have led to discovery of many new active constituents. However, it is a constant challenge to establish the pharmacological basis for efficacy and safety of herbal medicinal products. A better understanding of the effects and bioavailability of phytopharmaceuticals can help in discovering suitable and rational therapies. In this review, we present the bioavailability studies in immune system that has been conducted for some of the more important or widely used phytopharmaceuticals. Furthermore, various new drug targets worthy of using for drug development in immunomodulating herbal medicine area and their regulatory mechanisms are also discussed. Adverse effects, drug interactions, and contraindications are also discussed which show that caution should be exercised when combining phytopharmaceuticals with chemically derived pharmaceutical components. The structures, energies, stabilities and spectroscopies of doublet C4H2＋ cations were explored at the DFT/B3LYP/6-311G（d,p）, CCSD（T）/6-311＋G（2df2pd）（single-point）, and G3B3 levels. Ten minimum isomers including the chainlike, three-member-ring, and four-member-ring structures are interconverted by means of 15 interconversion transition states. The potential energy surface was investigated. At the CCSD（T）/6-311＋G（2df2pd） and G3B3 levels, the global minimum isomer was found to be a linear HCCCCH. The structures of the stable isomer and its relevant transition state are further optimized at the QCISD/6-311G（d,p） level. The bonding nature and structure of isomer HCCCCH were analyzed.

Beta-nerve growth factor(β-NGF) is known to be a major leading cause of neuronal plasticity. To identify the possible action mechanisms of β-NGF gene therapy for sciatic nerve recovery, experimental dogs were randomly divided into control, pyridoxine, and pyridoxine + β-NGF groups. We observed chronological changes of morphology in the dorsal root ganglia in response to pyridoxine toxicity based on cresyl violet staining. The number of large neurons positive for cresyl violet was dramatically decreased after pyridoxine intoxication for 7 days in the dorsal root ganglia and the neuron number was gradually increased after pyridoxine withdrawal. In addition, we also investigated the effects of β-NGF gene therapy on neuronal plasticity in pyridoxine-induced neuropathic dogs. To accomplish this, tyrosine kinase receptor A(TrkA), βIII-tubulin and doublecortin(DCX) immunohistochemical staining was performed at 3 days after the last pyridoxine treatment. TrkA-immunoreactive neurons were dramatically decreased in the pyridoxine group compared to the control group, but strong TrkA immunoreactivity was observed in the small-sized dorsal root ganglia in this group. TrkA immunoreactivity in the dorsal root ganglia was similar between β-NGF and control groups. The numbers of βIII-tubulin-and DCX-immunoreactive cells decreased significantly in the pyridoxine group compared to the control group. However, the reduction of βIII-tubulin-and DCX-immunoreactive cells in the dorsal root ganglia in the β-NGF group was significantly ameliorated than that in the pyridoxine group. These results indicate that β-NGF gene therapy is a powerful treatment of pyridoxine-induced neuropathic damage by increasing the TrkA and DCX levels in the dorsal root ganglia. The experimental protocol was approved by the Institutional Animal Care and Use Committee(IACUC) of Seoul National University, South Korea(approval No. SNU-060623-1, SNU-091009-1) on June 23, 2006 and October 9, 2009, respectively.

A peer-to-peer (P2P) multimedia conferencing service is operating that users share their resources to each other on the Internet. It can solve the problem in the centralized conferencing architecture, such as the centralized loading, single point error, and expensive infrastructure. However, P2P networks have the problem that a peer has a difference between the physical location and logical location in the overlay network. In the viewpoint of P2P networks, the nearest conference resource may be far away geographically. The P2P-session initiation protocol (P2P-SIP) multimedia conference is to construct an application-based logical multicast network efficiently according to physical network information. Thus, this paper proposes a real-time streaming relay mechanism for P2P conferences on hierarchical overlay networks. The real-time streaming relay mechanism can improve the transportation efficiency of conferencing stream exchange well based on the application-layer multicast (ALM) structure and the hierarchical overlay networks.

Double layered self-expanding metal stents for malignant esophageal obstruction, especially across the gastroesophageal junction

AIM: To evaluate the clinical outcomes of double-layered self-expanding metal stents (SEMS) for treatment of malignant esophageal obstruction according to whether SEMS crosses the gastroesophageal junction (GEJ). METHODS: Forty eight patients who underwent the SEMS insertion for malignant esophageal obstruction were enrolled. Patients were classified as GEJ group (SEMS across GEJ, 18 patients) and non-GEJ group (SEMS above GEJ, 30 patients) according to SEMS position. Double layered (outer uncovered and inner covered stent) esophageal stents were placed. REsuLTS: The SEMS insertion and the clinical improvement were achieved in all patients in both groups. Stent malfunction occurred in seven patients in the GEJ group and nine patients in the non-GEJ group. Tumor overgrowth occurred in five and eight patients, respectively, food impaction occurred in one patient in each group, and stent migration occurred in one and no patient, respectively. There were no significant differences between the two groups. Reflux esophagitis occurred more frequently in the GEJ group (eight vs five patients, P = 0.036) and was controlled by proton pump inhibitor. Aspiration pneumonia occurred in zero and five patients, respectively, and tracheoesophageal fistula occurred in zero and two patients, respectively. CONCLUSION: Double-layered SEMS are a feasible and effective treatment when placed across the GEJ for malignant esophageal obstruction. Double-layered SEMS provide acceptable complications, especially migration, although reflux esophagitis is more common in the GEJ group.

High-and medium-entropy alloys(HEAs and MEAs)possess high solid-solution *** investigations have been conducted on its impact on yield strength,however,there are limited reports regarding the relation between solid-solution strengthening and strain-hardening *** addition,no attempt has been made to account for the dislocation-mediated plasticity;most works focused on twinning-or transformation-induced plasticity(TWIP or TRIP).In this work we reveal the role of solidsolution strengthening on the strain-hardening rate via systematically investigating evolutions of deformation structures by controlling the Cr/V ratio in prototypical V_(1-x)Cr_(x)CoNi *** the TWIP of CrCoNi with the dislocation slip of V_(0.4)Cr_(0.6)CoNi,the hardening rate of CrCoNi was superior to slip-band refinements of V_(0.4)Cr_(0.6)CoNi due to the dynamic Hall-Petch ***,as V content increased further to V_(0.7)Cr_(0.3)CoNi and VCoNi,their rate of slip-band refinement in V_(0.7)Cr_(0.3)CoNi and VCoNi with high solid-solution strength surpassed that of *** it is generally accepted in conventional alloys that deformation twinning results in a higher strain-hardening rate than dislocation-mediated plasticity,we observed that the latter can be predominant in the former under an activated huge solid-solution strengthening *** high solid-solution strength lowered the cross-slip activation and consequently retarded the dislocation rearrangement rate,i.e.,the dynamic *** delay in the hardening rate decrease,therefore,increased the strain-hardening rate,results in an overall higher strain-hardening rate of V-rich alloys.
