Epac2-deficiency leads to more severe retinal swelling, glial reactivity and oxidative stress in transient middle cerebral artery occlusion induced ischemic retinopathy

Ischemia occurs in diabetic retinopathy with neuronal loss, edema, glial cell reactivity and oxidative stress. Epacs, consisting of Epac 1 and Epac2, are cAmP mediators playing important roles in maintenance of endothelial barrier and neuronal functions To investigate the roles of Epacs in the pathogenesis of ischemic retinopathy, transient middle cerebral artery occlusion （tmCAO） was performed on Epacl-deficient （Epacl-/- ） mice, Epac2-deficient （Epac2-/-） mice, and their wild type counter-parts （Epacl＋/＋ and Epac2＋/＋）. Two-hour occlusion and 22-hour reperfusion were conducted to induce ischemia/reperfusion injury to the retina. After tmCAO, the contralateral retinae displayed similar morphology between different genotypes. Neu-ronal loss, retinal edema and increase in im.unoreactivity for aquaporin 4 （AQP4）, glial fibrillary acidic protein （GFAP）, peroxiredoxin 6 （Prx6） were observed in ipsilateral retinae. Epac2 / ipsilateral retinae showed more neuronal loss in retinal ganglion cell layer, increased retinal thickness and stronger im.unostaining of AQP4, GFAP, and Prx6 than those of Epac2＋/＋. However, Epacl-/- ipsilateral retinae displayed similar pathology as those in Epacl＋/＋ mice. Our observations suggest that Epac2-deficiency led to more severe ischemic retinopathy after retinal ischemia/reperfusion injury.

Provenance of Cored Sediments from Active margin off Southwestern Taiwan Deduced from Geochemical Constraints

The cored sediments sampled by R/V marion Dufrense in the active margin off southwestern Taiwan on the east side of the manila Trench are mostly muds consisting mainly of quartz, feldspar, illite, chlorite ＋ kaolinite with a trace am.unt of montmorillonite. The cored sediments have higher Fe, Ti, Cr, Cs, Li, Ni, and V but lower Ca, Na, mn, Hf, Nb, Sr, Ta, u and Zr contents when compared with the upper continental crust（uCC, Taylor and mcLennan, 1985）. The Eu/Eu＊ ratios of the sediments averaging 0.69 indicate that the recycling effect on the Eu anomaly may be minor and the contribution of mafic source rock was unimportant, which can also be depicted in the La-Th-Sc plot（Cullers, 1994） showing a ＂mixed origin＂ close to the felsic end member. The chemistry of the cored sediment can be explained by a mixing model designed by Ho and Chen（1996） with four end members i.e., greywacke, shale, quartzite and limestone, which are widely distributed in the Cenozoic formations of southern Taiwan with greywacke as the dominant contributor followed by shale.

Chemical and Sr-Nd compositions and ^(40)Ar/^(39)Ar ages of NW-trending dolerite dikes of Burkina Faso: Evidence for a mesoproterozoic magmatism in the West African Craton

The Paleoproterozoic basement of the northeastern part of the Leo-man craton is intruded by generally NW-trending dikes. These regional scale dikes extend over 1000 km in Burkina Faso, mali and Niger. We present chemical and Sr-Nd isotope compositions, as well as 40 Ar/39 Ar ages of these dikes with the following strikes N98°-N112°, and N114°-N124° in NE Burkina Faso. Field relationships show that the dikes are posterior to all other rock types dated between 2.26 Ga and 2.0 Ga. Chemical data indicate that the dikes are continental flood basalts and composed of low-Ti(TiO2 ≤ 2 wt.%) sub-alkaline basalts and andesites. They exhibit a minor negative Europium anomaly(0.86-0.99) and slightly fractionated REE patterns((La/Yb)_N= 2.5-9.1; Yb_N = 9.5-19.9). The ratios of Th/Ta(1.3-11.4) and Ce/Pb(5.2-58.5)suggest a varying crustal assimilation of the dike magmas during ascent in the continental crust for all studied samples. Calculated P-T conditions indicate that the magma reached temperatures of 1285 ℃(calculated from olivine compositions) and pressures of 6.9 kbar(calculated for pyroxene minerals).Calculated initial 87 Sr/86 Sr(0.70040-0.70260) and ε_(Nd)(t)=+2.1 to-3.5 at 1575 ma,also point to a crustal contamination with the most primitive samples showing T_(Dm) values of 1946 ma and 2154 ma. The low values of La/Ba(low Th/Nb(uggest a lithospheric mantle or subduction-modified mantle as possible source for the dikes. Sr-Nd data, mg# and NbTa-Zr-Y-Th-Tb-Yb compositions further suggest that the most primitive samples were emplaced in a none orogenic setting and their magmas were subjected to variable crustal contaminations. Literature and the present whole rock 40 Ar/39 Ar age determinations show that the dikes were emplaced during a widespread mesoproterozoic magmatism between 1.6 Ga and 1.2 Ga, and were affected by a thermal event causing the argon systematics resetting, best constrained by the date of sample KK1(123

Exacerbated VEGF up-regulation accompanies diabetes-aggravated hemorrhage in mice after experimental cerebral ischemia and delayed reperfusion

Dear Editor,Since the late 1980s, trauma in China has been identified as a major public health challenge, with trafic-related fatalities accounting for 80% of accidental deaths[1]. In 2019, Shenzhen had a total population of approximately 20 million, and from 2010 to 2017, both emergency medical services and the total number of trauma patients increased, with trauma accounting for 47.0% and 38.4% of all patients in 2010 and 2017, respectively[2,3]. This report describes the efforts to implement programs and establish a trauma system in Shenzhen, China.

Prevacuolar compartments （PVCs） and endosomal compartments are membrane-bound organelles mediating protein traffic to vacuoles in the secretory and endocytic pathways of plant cells. Over the years, great progress has been made towards our understanding in these two compartments in plant cells. In this review, we will summarize our contributions toward the identification and characterization of plant prevacuolar and endosomal compartments. Our studies will serve as important steps in future molecular characterization of PVC biogenesis and PVC-mediated protein traffickinq in plant cells.

Primary and metastatic liver cancer is an increasingly common and difficult to control disease *** offers a non-invasive treatment alternative for these patients who often have few options and a poor ***,the anatomy and aggressiveness of liver cancer poses significant challenges such as accurate localization at simulation and treatment,management of motion and appropriate selection of dose *** article aims to review the options available and provide information for the practical implementation and/or improvement of liver cancer radiation programs within the context of stereotactic body radiotherapy and image-guided radiotherapy *** patient inclusion and exclusion criteria are presented given the significant toxicity found in certain sub-populations treated with ***,certain sub-populations,such as those with tumor thrombosis or those with larger lesions treated with transarterial chemoembolization,have been shown to have significant improvements in outcome with the addition of radiation and merit special *** a liver radiation programrequires three primary challenges to be addressed:(1) immobilization and motion management;(2) localization;and(3) dose regimen and constraint *** to deal with motion include simple internal target volume(ITV) expansions,non-gated ITV reduction strategies,breath hold methods,and surrogate marker methods to enable gating or *** of the tumor and organs-at-risk are addressed using contrast infusion techniques to take advantage of different normal liver and cancer vascular anatomy,imaging modalities,and margin ***,a dose response has been demonstrated and dose regimens appear to be converging.A more uniform approach to treatment in terms of technique,dose selection and patient selection will allow us to study liver radiation in larger and,hopefully,multicenter randomized studies.

Survival outcomes of liver transplantation for hepatocellular carcinoma in patients with normal, high and very high preoperative alpha-fetoprotein levels

AIm To investigate the impact of alpha-fetoprotein(AFP) on long-term recurrence rate and overall survival and we also aimed to define the level of AFP leading to a higher risk of disease recurrence and affecting patient *** Data of adult patients who received liver transplant(LT) for hepatocellular carcinoma(HCC) at our hospital from January 2000 to December 2013 were reviewed. Reviewed data included demographic characteristics, preoperative AFP level, operative details, follow-up details, and survival outcomes. Patients were mostly listed for LT based on milan or uCSF criteria. For the purpose of this study, normal AFP level was defined as AFP value m L, high AFP level was defined as AFP value ≥ 10 to m L, and very highAFP level was defined as AFP ≥ 400 ng/m L. The patients were divided into these 3 groups accordingly. Survival rates were plotted as Kaplan-meier curves and compared by log-rank analysis. Continuous variables were expressed as median(interquartile range). Categorical variables were compared by Spearman's test. Discriminative analysis was used to define the lowest value of AFP that could affect the overall survival in study population. Statistical significance was defined by a P value of ult patients underwent LT for HCC in the study period. Eight-four of them received deceaseddonor LT and 166 had living-donor LT. The patients were divided into 3 groups: Group A, AFP m L(n = 83); Group B, AFP ≥ 10 to m L(n = 131); Group C, AFP ≥ 400 ng/m L(n = 36). The commonest etiology was hepatitis-B-related cirrhosis. The model for End-stage Liver Disease scores in these groups were similar(median, 13 vs 13 vs 12; P = 0.745). The time to operation in Group A was longer(median, 94 vs 31 vs 35 d; P = 0.001). Structural, electronic, and optical properties of ZnO_(1-x)Se_x alloys using first-principles calculations

The structural, electronic, and optical properties of binary ZnO, ZnSe compounds, and their ternary ZnOl_xSex alloys are com.uted using the accurate full potential linearized augmented plane wave plus local orbital （FP-LAPW ＋ lo） method in the rocksalt （B 1） and zincblende （B3） crystallographic phases. The electronic band structures, fundamental energy band gaps, and densities of states for ZnO1_xSex are evaluated in the range 0 〈 x 〈 1 using Wu-Cohen （WC） generalized gradient approximation （GGA） for the exchange-correlation potential. Our calculated results of lattice parameters and bulk modulus reveal a nonlinear variation for pseudo-binary and their ternary alloys in both phases and show a considerable deviation from Vegard＇s law. It is observed that the predicted lattice parameter and bulk modulus are in good agreement with the available experimental and theoretical data. We establish that the composition dependence of band gap is semi-metallic in B1 phase, while a direct band gap is observed in B3 phase. The calculated density of states is described by taking into account the contribution of Zn 3d, O 2p, and Se 4s, and the optical properties are studied in terms of dielectric functions, refractive index, reflectivity, and energy loss function for the B3 phase and are compared with the available experimental data.
