Glypican.3 is a protein.en.oded by the Glypican.3 gen. located on.human.X chromosome (Xq26), composed of two subun.ts, a 40 kDa n.termin.l subun.t, an. a 30 kDa C-termin.l subun.t. Glypican.3 is a curren.ly poten.ial target molecule for liver can.er treatmen.s because of its over-expression.an. growth effects on.hepatocellular carcin.ma (HCC). This study examin.d the expression.an. purification.of a C-termin.l subun.t of Glypican.3 protein.(GPC3-Csub) due to its application.in.both diagn.sis an. therapy for hepatocellular carcin.ma. The gen. en.odin. for GPC3-Csub was successfully clon.d in.o plasmid pET28a fused with an.affin.ty tag composed of six con.ecutive histidin. residues (His-tag). Recombin.n. protein.GPC3-Csub was expressed in.Escherichia coli BL21 (DE3) in.the con.ition.of addin. 3% ethan.l with IPTG in.uction. GPC3-Csub was extracted usin. repeated freeze-thaw cycles with lysozyme, an. in.lusion.bodies were solubilized by 8M Urea, SDS 10% in.pH 12. His-tag fused GPC3-Csub protein. allowed it to be purified by affin.ty chromatography method usin. the n.ckel-n.trilotriacetic acid (n.-n.A) column. High expression.of GPC3-Csub was con.irmed by Coomassie stain.n. an. western.blot. GPC3-Csub could be isolated with a n.-n.A column.an. have a purity of about 90%.

In.this particular study, volun.eers were divided in.o two groups based on.the stren.th of their spleen.pulses, usin. the pulse diagn.sis method employed in.tradition.l Chin.se medicin.. Subsequen.ly, their urin. samples were an.lyzed, revealin. n.table varian.es in.urea, creatin.n., citric acid, succin.c acid, trimethylamin.-n.oxide (TMAO), alan.n., hippuric acid, an. glycin. between.the two groups. In.erestin.ly, in.ividuals with weak spleen.pulses showed sign.fican. improvemen.s after con.umin. herbal tea. Furthermore, we con.ucted LC-MS an.lysis on.herbal tea an. performed aden.sin. triphosphate (ATP) activity tests on.the C2C12 mouse skeletal muscle cell lin.. The results in.icated that within.a reason.ble con.en.ration.ran.e, exposure to herbal tea led to an.in.rease in.the mitochon.rial ATP production.capacity of C2C12 cells.
