Seasonal variations of nonmotor symptoms in patients with Parkinson's disease in Southeast China

Background:Studies suggest seasonal fluctuations of symptoms in Parkinson’s disease(PD)patients in Western countries.However,the association between seasonal change and variation in nonmotor symptoms(NMS)in Chinese PD patients is unclear.Here,we studied whether there is a change rule with annual cycle with severity of NMS for patients with PD in Southeast China.Methods:We studied 1005 PD patients between April 2008 and October 2020.Patients were classified into four seasons according to the 24 Chinese solar terms,based on assessment date.We compared comprehensive NMS scales and polysomnography parameters among groups and conducted further analysis of disease severity.Results:Among the 1005 patients studied,the mean age was 64.2±9.7 years and 569(56.6%)of them were men.Relative to the summer group,patients assessed during winter had higher Scales for Outcomes in Parkinson’s disease-Autonomic Dysfunction(SCOPA-AUT)scores(P=0.045).The sleep efficiency factor scores of Pittsburgh Sleep Quality Index in patients were higher during spring than summer(P=0.009).Among patients who completed polysomnography during the same period(n=135),compared with summer follow-ups,we observed a higher percentage of NREMS1 in winter and spring follow-ups(P=0.042,P=0.011),a higher NREMS1 time in spring follow-ups(P=0.0024),a lower NREMS2 time in winter follow-ups(P=0.007),and a higher percentage of phasic rapid eye movement(REM)-sleep without atonia in autumn and winter follow-ups(P=0.026 and P=0.020,respectively).In a subset of patients with PD and REM sleep behavior disorder(RBD;n=182),those visited during winter had higher scores for RBD questionnaire-Hong Kong and its factor 1(dream-related sub-score)than those visited during summer(P=0.034,P=0.020).We observed similar findings for SCOPA-AUT and sleep efficiency factor scores in early stage patients in subgroup analysis.Conclusions:PD patients assessed for follow-up during summer showed less severe symptoms of autonomic dysfunction and RBD symptoms than those assessed in winter,and l

Following nerve injury,axonal disconnection in neurons usually results in persistent functional deficits,such as paralysis.However,axons in the adult mammalian central nervous system (CNS) have very limited regenerative ability.Understanding the molecular mechanism of controlling axon regeneration can provide idea for the design of effective therapeutic interventions for CNS injury,such as spinal cord injuries.Efficient axonal regeneration is achieved via gene expression in the neuronal soma,axonal transport of raw materials along the shaft,and membrane and cytoskeleton assembly at the nerve growth cone.Each process is delicately regulated by spatial-temporal controlled signaling pathways that target distinct effectors.Gene expression in the neuronal soma,especially of transcription factors,is often activated immediately following nerve injury.Injury signals at distal axons are interpreted and transmitted back to the soma,initiating a stream of gene expression events which positively regulate subsequent axonal regeneration.Over the past few decades,extensive studies have identified many regeneration-associated genes,including CREB,nuclear factor of activated T-cells,protein 53,Sprr1a,c-Jun,Smad1,activating transcription factor 3,signal transducer and activator of transcription 3,SRF,Sox11,and Kruppel-like factors.However,we know far less about how the coordinated expression of these regeneration-associated genes is regulated during axonal regeneration.Indeed,it is possible that they are regulated by a single common upstream regulator.If so,identification of this upstream regulator will provide us with an invaluable target for the development of more effective treatments for traumatic nerve injuries.Adult dorsal root ganglion (DRG) neurons represent a favorable medium in which to study the molecular mechanisms controlling intrinsic neuronal axon growth ability.Axotomy of the peripheral branch of a DRG neuron,known as a “conditioning lesion”,has been well-documented to greatly accelerate axonal growth both in vivo

This study aimed to reveal the effect of salt reduction on the nutritional,functional and sensory aspects of doubanjiang through preparing three sets of doubanjiang fermentation(Sample H,Sample M and Sample L)with different salt concentrations.The results showed that fermentation with lower salt concentration led to significantly higher amino acids concentrations in doubanjiang,while the organic acids concentrations were also slightly increased.For biologically active compounds,the concentrations of total flavonoids,total phenols and five monomer isoflavones in Sample L were all significantly higher than those in Sample M and Sample H.Moreover,better anti-oxidant ability was observed in doubanjiang samples fermented with lower salt concentration.Bacillus and Millerozyma genus were found to be closely related to the formation of amino acids and biologically active compounds in doubanjiang,while organic acids were highly correlated with Cronobacter,Erwinia,Trabulsiella,Enterobacter and Millerozyma genus.Through sensory evaluation and electric tongue,unsatisfactory sensory characteristics,such as lighter color,strong acid taste and off-flavor,were found in lower salt fermented doubanjiang samples.This indicated that lower salt concentration favored the nutrition and function of doubanjiang while negatively influenced doubanjiang flavor.This study deepened our understanding of the roles of salt concentration on doubanjiang fermentation and provided guidance for the further development of low-salt doubanjiang.

To the Editor:Parkinson’s disease(PD)is one of the most common neurodegenerative disorders and the most common movement disorder worldwide.PD pathogenesis has been demonstrated to be associated with neuroinflammation and intestinal inflammation.Increased C-reactive protein(CRP)levels were related to freezing of gait and rapid PD progression,particularly in elderly patients.Recent research indicates that immune cells in peripheral blood might also contribute to PD pathogenesis,providing further insight into the relationship between inflammation and PD.

Facing the current energy crisis and climate change,the discovery of robust yet inexpensive photocatalysts that harvest solar energy for overall water splitting is highly desirable.Recently,organic materials have attracted much attention in photocatalysis,attributed to their facile molecular-level functionalization,adjustable optoelectronic properties,favorable chemical robustness,and lesser environmental hazards as compared with their inorganic counterparts.This minireview highlights the recent advances of organic photocatalysts in the field of solar water splitting.The challenges,opportunities,and future perspectives within the field are discussed to inspire future research studies into organic-based photocatalytic water splitting.

Autophagy in Atherosclerosis： A Phenomenon Found in Human Carotid Atherosclerotic Plaques

Study Objectives: Mounting evidence indicated the correlation between sleep and cerebral small vessel disease(CSVD).However, little is known about the exact causality between poor sleep and white matter injury, a typical signature of CSVD, as well as the underlying mechanisms.Methods: Spontaneously hypertensive rats(SHR) and control Wistar Kyoto rats were subjected to sleep fragmentation(SF) for 16 weeks.The effects of chronic sleep disruption on the deep white matter and cognitive performance were observed.Results: SHR were validated as a rat model for CSVD.Fragmented sleep induced strain-dependent white matter abnormalities, characterized by reduced myelin integrity, impaired oligodendrocytes precursor cells(OPC) maturation and pro-inflammatory microglial polarization.Partially reversible phenotypes of OPC and microglia were observed in parallel following sleep recovery.Conclusions: Long-term SF-induced pathological effects on the deep white matter in a rat model of CSVD.The pro-inflammatory microglial activation and the block of OPC maturation may be involved in the mechanisms linking sleep to white matter injury.

Background：Autophagy has been found to be involved in animal and cell models ofatherosclerosis,but to date,it lacks general observation in human atherosclerotic plaques.Here,we investigated autophagy in smooth muscle cells （SMCs）,endothelial cells （ECs）,and macrophages in human atherosclerotic plaques via transmission electron microscopy （TEM）,western blotting,and immunohistochemistry analysis.Methods：The histopathologic morphology of these plaques was observed via hematoxylin and eosin staining.The ultrastructural morphology of the SMCs,ECs,and macrophages in these plaques was observed via TEM.The localization ofmicrotubule-associated protein 1 light chain 3 （MAP 1-LC3）,a relatively special maker ofautophagy,in plaques was observed by double fluorescent immunochemistty and western blotting.Results：All of these human atherosclerotic plaques were considered advanced and unstable in histologically observation.By double fluorescent immunochemistry,the expression of LC3-Ⅱ increased in the SMCs of the fibrous cap,the macrophages,and the microvascular ECs of the plaque shoulders.The protein level of LC3-Ⅱ by western blotting significantly increased in plaques compared with normal controls.In addition,TEM observation of plaques revealed certain features of autophagy in SMCs,ECs,and macrophages including the formation of myelin figures,vacuolization,and the accumulation of inclusions in the cytosol.These results indicate that autophagy is activated in SMCs,ECs,and macrophages in human advanced atherosclerotic plaques.Conclusions：Our study is to demonstrate the existence of autophagy in human atherosclerotic plaques by different methods,which may contribute to the development of pharmacological approaches to stabilize vulnerable and rupture-prone lesions.
