R2R3-MYB转录因子家族在植物次生代谢物合成、胁迫应答和生长发育等生命过程起着重要的调控作用。本研究基于生物信息学鉴定了绿豆(Vigna radiata L.)全基因组水平的R2R3-MYB转录因子,并对其理化性质、系统进化、染色体定位、启动子顺式作用元件及基因结构做了预测分析;此外,转录组数据和实时荧光定量PCR(QuantitativeReal-timePCR,RT-PCR)分析该转录因子在不同组织、逆境胁迫下的表达模式;并基于相关分析及蛋白互作网络,筛选到可能参与调控绿豆类黄酮生物合成的R2R3-MYB成员。结果表明,共鉴定到168个R2R3-MYB成员,其中145个分布于11条染色体, 23个成员染色体信息未知;大多数R2R3-MYB含有3个外显子,编码99~1645个氨基酸,均为亲水性蛋白;系统进化将绿豆R2R3-MYB基因家族分为30个亚组(V1~V30),不同亚组成员的基因结构存在差异;共线性分析表明,片段复制事件均进行了纯化选择;启动子顺式作用元件分析表明,绿豆R2R3-MYB基因启动子区含有大量激素响应、胁迫响应及少量的类黄酮合成响应等元件;基因表达分析表明,在叶片、叶柄、下胚轴和籽粒种皮中表达量较高的成员分别占15.5%、16.1%、16.1%和10.7%。RT-PCR分析发现,几乎所有的R2R3-MYB家族成员在低温胁迫下的相对表达量显著下调,不同成员对逆境胁迫有不同的响应模式。蛋白互作与相关性分析可知,VrMYB6、VrMYB77、VrMYB93这3个基因可能参与了绿豆类黄酮生物合成的调控。

This paper proposes a multivariable fixed-time leaderfollower formation control method for a group of nonholonomic mobile robots, which has the ability to estimate multiple uncertainties. Firstly, based on the state space model of the leader-follower formation, a multivariable fixed-time formation kinematics controller is designed. Secondly, to overcome uncertainties existing in the nonholonomic mobile robot system, such as load change,friction, external disturbance, a multivariable fixed-time torque controller based on the fixed-time disturbance observer at the dynamic level is designed. The designed torque controller is cascaded with the formation controller and finally realizes accurate estimation of the uncertain part of the system, the follower tracking of reference velocity and the desired formation of the leader and the follower in a fixed-time. The fixed-time upper bound is completely determined by the controller parameters, which is independent of the initial state of the system. The multivariable fixed-time control theory and the Lyapunov method are adopted to ensure the system ***, the effectiveness of the proposed algorithm is verified by the experimental simulation.

In the intricate process of natural product biosynthesis, a metabolon can enhance metabolic flux by associating sequential enzymes. A fungal metabolon, comprising of flavin-dependent monooxygenase Spe F and P450 monooxygenase Spe G, is identified in the biosynthesis of spiro polycyclic alkaloids(+)-notoamide B and its diastereomer(+)-versicolamide B. Using notoamide E as a substance, Spe F/Spe G metabolon can control the stereoselectivity of its 2,3-epoxidation, followed by hydrogen atom ion at C-17 to generate reactive epoxide tau-m A with dienyl iminium unit. Subsequently,(+)-notoamide B and(+)-versicolamide B are produced via tandem nonenzymatic inverse-electron-demand Diels-Alder reaction and semipinacol rearrangement. This provides the first example of metabolon in the biosynthesis of spiro-prenylated indole alkaloids.

Combination of fruquintinib with venetoclax for the treatment of colorectal cancer

Background:As a novel blocker of vascular endothelial growth factor receptor (VEGFR), fruquintinib has been approved for treating colorectal cancer (CRC). However, its dosage and therapeutic efficacy are limited by its widespread adverse reactions. Venetoclax, recognized as the initial inhibitor of B-cell lymphoma protein 2 (BCL2), has shown potential in boosting the effectiveness of immunotherapy against CRC. This study investigated the efficacy and mechanisms of fruquintinib combined with venetoclax in treating CRC.Methods and Materials:We developed a colon cancer mouse model with the CT26 colon cell line to demonstrate fruquintinib and venetoclax's efficacy against tumors. Then we employed various techniques to evaluate different aspects of the experimental outcomes. Immunohistochemistry was used to detect cell proliferation and angiogenesis in tumor tissues. Western blot analysis was utilized to examine the occurrence of cell apoptosis, and flow cytometry to quantitate immune cells within the tumor tissues. Moreover, immunofluorescence was employed to measure cytokine levels.Results:The strongest inhibition on tumor growth was achieved by the combination of fruquintinib with venetoclax, as opposed to individual drug use. Venetoclax was found to amplify the impact of fruquintinib, leading to decreased cancer cell proliferation, increased cancer cell apoptosis, lowered angiogenesis, better vascular structure normalization, and improved immune cell infiltration.Conclusion:Our findings indicate that the addition of venetoclax enhances the impact of fruquintinib on vascular normalization and modulation of the tumor immune microenvironment. Our study presents the justification for utilizing the fruquintinib and venetoclax combination in treating CRC. Venetoclax holds promise in being assimilated into anticancer medications for CRC.

Noise-induced hearing loss is the primary non-genetic factor contributing to auditory ***,there are currently no effective pharmacological interventions for patients with noise-induced hearing ***,we present evidence suggesting that the lysine-specific demethylase 1 inhibitor–tranylcypromine is an otoprotective agent that could be used to treat noise-induced hearing loss,and elucidate its underlying regulatory *** established a mouse model of permanent threshold shift hearing loss by exposing the mice to white broadband noise at a sound pressure level of 120 d B for 4 *** found that tranylcypromine treatment led to the upregulation of Sestrin2(SESN2)and activation of the autophagy markers light chain 3B and lysosome-associated membrane glycoprotein 1 in the cochleae of mice treated with *** noise exposure group treated with tranylcypromine showed significantly lower average auditory brainstem response hearing thresholds at click,4,8,and 16 k Hz frequencies compared with the noise exposure group treated with *** findings indicate that tranylcypromine treatment resulted in increased SESN2,light chain 3B,and lysosome-associated membrane glycoprotein 1 expression after noise exposure,leading to a reduction in levels of 4-hydroxynonenal and cleaved caspase-3,thereby reducing noise-induced hair cell ***,immunoblot analysis demonstrated that treatment with tranylcypromine upregulated SESN2 expression via the autophagy *** treatment also reduced the production of NOD-like receptor family pyrin domaincontaining 3(NLRP3)*** conclusion,our results showed that tranylcypromine treatment ameliorated cochlear inflammation by promoting the expression of SESN2,which induced autophagy,thereby restricting NLRP3-related inflammasome signaling,alleviating cochlear hair cell loss,and protecting hearing *** findings suggest that inhibiting lysine-specific demethylase 1 is a

Bioenergy development improves energy security and mitigates climate *** the stable supply of agricultural resources provides a solid basis for efficient bioenergy production,the climate-induced impacts such as changes in lempeniture and precipitation that potentially alter the farming decisions and cropping patterns should be taken into *** study designs two-stage stochastic programming with recourse model to investigate the net bioenergy production and emission reduction,as well as the bioenergy producers'strategies under various market operations and climate *** results show that the net production of biopower and biofuel are relatively stable because the supply of bioenergy feedstocks could remain at a constant level,but the farming decisions,cropping pattern,and resource utilization are likely to be altered by climate-induced *** results also indicate that for efficient bioenergy production,approximately 41.5 billion TWD(9.65 billion CNY)and 33.6 billion TWD(7.81 billion CNY)of bond principal should be invested in crop-based and residual-based technology,*** the face of strict environmental regulations and emission management,biopower technology becomes more competitive,and under such a circumstance,more than 98.3%of the low-cost bond should be invested in biopower development to achieve development efficiency.

Effects of seven different companion plants on cucumber productivity, soil chemical characteristics and Pseudomonas community

Companion cropping can influence cucumber productivity by altering soil chemical characteristics and microbial communities. However, how these alterations affect the growth of cucumber is still unknown. In this study, seven different plant species were selected as companion plants for testing their effects on cucumber productivity. The effects of different companion plants on changes in soil chemical properties such as electrical conductivity （EC） and contents of essential nutrients as well as the structure and abundance of the soil Pseudomonas community were evaluated. The results showed a higher cucumber yield in the wheat/cucumber companion system than that in the cucumber monocultured and other companion cropping systems. The lowest phosphorus （P） and potassium （K） contents in the soil were found in the cucumber monocultured system, and the highest NO3＋-N and NH4＊-N contents were observed in the rye/cucumber companion system. PCR-denaturing gradient gel electrophoresis （DGGE） and real-time PCR analysis showed that the trifolium/cucumber companion system increased the diversity of the soil Pseudomonas community, while the chrysanthemum/cucumber companion system increased its abundance. Interestingly, plant-soil feedback trials revealed that inoculating the soil of the wheat/cucumber companion system increased the growth of cucumber seedlings. In conclusion, the effects of different companion plants on cucumber productivity, soil chemical characteristics and the soil Pseudomonas community were different, and wheat was a more suitable companion plant for increasing cucumber productivity. In addition, the altered microbial community caused by companion cropping with wheat contributed to increased cucumber productivity.

The melting mechanisms of Pt-based multimetallic nanoparticles(NPs)are important to help determine their optimal melting *** understand the melting and coalescence behaviors of heterogeneous NPs(Pd-Pt NPs)with various sizes and compositions,molecular dynamics(MD)simulation was *** MD results for larger Pd-Pt NPs with an effective diameter of4.6-7.8 nm show that PtPd alloy can form at Pd/Pt interface before Pd NP melted completely,while for Pt-core/Pdshell NP and Pd-core/Pt-shell NP,PtPd alloy formed only after Pd portion melted *** smaller Pd-Pt NPs with an effective diameter of 2.5-4.0 nm,PdPt alloy is not formed until both Pd and Pt NPs melted ***,the coalescence process of Pd-Pt NPs depends on the melting temperature of Pt NP when Pt composition is higher than 20 at%.Furthermore,the melting mechanisms of Pd/Pt/Ir trimetallic NPs are investigated.A two-step melting process occurs in Pd-Pt-Ir NPs and Ir-core/Ptshell/Pd-shell NP,and the melting sequence of Pd-core/Ptshell/Ir-shell NP and Pt-core/Pd-shell/Ir-shell NP is different from Pd/Pt bimetallic NPs.
