Inflammatory and Immunomodulatory Effects of Tripterygium wilfordii Multiglycoside in Mouse Models of Psoriasis Keratinocytes

Objective:To determine the role of Tripterygium wilfordii multiglycoside(TGW) in the treatment of psoriatic dermatitis from a cellular immunological ***:Mouse models of psoriatic dermatitis were established by imiquimod(IMQ).Twelve male BALB/c mice were assigned to MQ or IMQ+TGW groups according to a random number *** changes in vivo were assessed by hematoxylin and eosin *** of immune cells and cytokines in mice,as well as PAM212 cell proliferation in vitro were assessed by flow ***-inflammatory cytokine expression was determined using reverse transcription quantitative polymerase chain ***:TGW significantly ameliorated the severity of IMQ-induced psoriasis-like mouse skin lesions and restrained the activation of CD45^(+)cells,neutrophils and T lymphocytes(all P<0.01).Moreover,TGW significantly attenuated keratinocytes(KCs) proliferation and downregulated the mRNA levels of inflammatory cytokines including interleukin(IL)-17A,IL-23,tumor necrosis factor α,and chemokine(C-X-C motif) ligand 1(P<0.01 or P<0.05).Furthermore,it reduced the number of γδ T17 cells in skin lesion of mice and draining lymph nodes(P<0.01).Conclusions:TGW improved psoriasis-like inflammation by inhibiting KCs proliferation,as well as the associated immune cells and cytokine *** inhibited IL-17 secretion from γδ T cells,which improved the immune-inflammatory microenvironment of psoriasis.

Background:Chimeric antigen receptor T(CAR-T)cell therapy has achieved marked therapeutic success in ameliorating hematological ***,there is an extant void in the clinical guidelines concerning the most effective chemotherapy regimen prior to chimeric antigen receptor T(CAR-T)cell therapy,as well as the optimal timing for CAR-T cell infusion *** and Methods:We employed cell-derived tumor xenograft(CDX)murine models to delineate the optimal pre-conditioning chemotherapy regimen and timing for CAR-T cell ***,transcriptome sequencing was implemented to identify the therapeutic targets and elucidate the underlying mechanisms governing the treatment ***:Our preclinical in vivo evaluation determined that a combination of cyclophosphamide and fludarabine,followed by the infusion of CD19 CAR-T cells five days subsequent to the chemotherapy,exerts the most efficacious therapeutic effect in B-cell hematological ***,RNA-seq data indicated that the therapeutic efficacy predominantly perturbs tumor cell metabolism,primarily through the inhibition of key mitochondrial targets,such as C-Jun Kinase enzyme(C-JUN).Conclusion:In summary,the present study offers critical clinical guidance and serves as an authoritative reference for the deployment of CD19 CAR-T cell therapy in the treatment of B-cell hematological malignancies.

High density dislocations enhance creep ageing response and mechanical properties in 2195 alloy sheet

The creep strain of conventionally treated 2195 alloy is very low,increasing the difficulty of manufacturing Al-Cu-Li alloy sheet parts by creep age ***,finding a solution to improve the creep formability of Al-Cu-Li alloy is vital.A thorough comparison of the effects of cryo-deformation and ambient temperature large pre-deformation(LPD)on the creep ageing response in the 2195 alloy sheet at 160℃with different stresses has been *** evolution of dislocations and precipitates during creep ageing of LPD alloys are revealed by X-ray diffraction and transmission electron ***-quality 2195 alloy sheet largely pre-deformed by 80%without edge-cracking is obtained by cryo-rolling at liquid nitrogen temperature,while severe edge-cracking occurs during room temperature *** creep formability and strength of the 2195 alloy are both enhanced by introducing pre-existing dislocations with a density over 1.4×10^(15)m^(−2).At 160℃and 150 MPa,creep strain and creep-aged strength generally increases by 4−6 times and 30−50 MPa in the LPD sample,respectively,compared to conventional T3 alloy *** elongation of creep-aged LPD sample is low but remains relevant for *** high-density dislocations,though existing in the form of dislocation tangles,promote the formation of refined T1 precipitates with a uniform dispersion.

To assess whether a development strategy will be profitable enough,production forecasting is a crucial and difficult step in the *** development history of other reservoirs in the same class tends to be studied to make predictions ***,the permeability field,well patterns,and development regime must all be similar for two reservoirs to be considered in the same *** results in very few available experiences from other reservoirs even though there is a lot of historical information on numerous reservoirs because it is difficult to find such similar *** paper proposes a learn-to-learn method,which can better utilize a vast amount of historical data from various ***,the proposed method first learns how to learn samples before directly learning rules in ***,by utilizing gradients from networks with independent parameters and copied structure in each class of reservoirs,the proposed network obtains the optimal shared initial parameters which are regarded as transferable information across different *** on that,the network is able to predict future production indices for the target reservoir by only training with very limited samples collected from reservoirs in the same *** cases further demonstrate its superiority in accuracy to other widely-used network methods.

Degradation of warm permafrost and talik formation on the Qinghai-Tibet Plateau in 2006-2021

Permafrost is degrading globally,particularly those with low thermal stability on the Qinghai-Tibet Plateau,owing to climate ***,the inadequacy of direct research on permafrost degradation based on in-situ monitoring limits the prediction of permafrost degradation and engineering *** study explored the processes and modes of permafrost degradation into talik by analyzing ground temperature data from five points in the hinterland of the Qinghai-Tibet Plateau from 2006 to *** results showed that the degradation of the warm permafrost layer with a geothermal gradient of zero occurred simultaneously in the top and bottom *** rate of permafrost degradation from the top down and bottom up increase during the degradation process,but the increase of the former is more drastic after the formation of thawed ***,the construction of the Qinghai-Tibet Railway changed the degradation modes of the permafrost in adjacent natural sites through horizontal heat transfer,particularly after through talik formation under the *** findings suggest that taking countermeasures before or immediately after forming thawed interlayer is more *** evaluating the thermal impact of projects in warm permafrost regions,special attention should be given to the horizontal heat transfer process that may result from the formation of a through talik.

A novel route to enhance high-temperature mechanical property and thermal shock resistance of low-carbon Mgo-C bricks by introducing ZrSiO_(4)

Conventional MgO-C bricks(graphite content>14 wt.%)produce a great deal of greenhouse gas emission,while low-carbon MgO-C bricks have serious thermal shock resistance during high-temperature *** enhance the high-temperature mechanical property and thermal shock resistance of low-carbon MgO-C bricks,a novel route of introducing ZrSiO_(4) powder into low-carbon MgO-C bricks was reported in such refractories with 2 wt.% flaky *** results indicate that the low-carbon MgO-C brick with 0.5 wt.%ZrSiO_(4) addition has the maximum hot modulus of rupture at 1400℃ and the corresponding specimen fired in the carbon embedded atmosphere has the maximum residual strength ratio(98.6%)after three thermal shock *** is found that some needle-like AlON and plate-like Al_(2)O_(3)-ZrO_(2) composites were in situ formed in the matrices after the low-carbon MgO-C bricks were coked at 1400℃,which can enhance the high-temperature mechanical property and thermal shock resistance due to the effect of fiber toughening and particle ***,CO_(2) emission of the newly developed low-carbon MgO-C bricks is reduced by 58.3% per ton steel after using them as the working lining of a 90 t vacuum oxygen decarburization ladle.

Because of the low price and abundant reserves of sodium compared with lithium,the research of sodium-ion batteries(SIBs)in the field of large-scale energy storage has returned to the research *** oxides distinguish themselves from the mains cathode materials of SIBs owing to their advantages such as high specific capacity,simple synthesis route,and environmental ***,the commercial development of the layered oxides is limited by sluggish kinetics,complex phase transition and poor air *** on the research ideas from macro-to micro-scale,this review systematically summarizes the current optimization strategies of sodium-ion layered oxide cathodes(SLOC)from different dimensions:microstructure design,local chemistry regulation and structural unit *** the dimension of microstructure design,the various structures such as the microspheres,nanoplates,nanowires and exposed active facets are prepared to improve the slow kinetics and electrochemical ***,from the view of local chemistry regulation by chemical element substitution,the intrinsic electron/ion properties of SLOC have been enhanced to strengthen the structural ***,the optimization idea of endeavors to regulate the physical and chemical properties of cathode materials essentially is put forward from the dimension of structural unit *** opinions and strategies proposed in this review will provide some inspirations for the design of new SLOC in the future.

Auxin regulates flower and fruit abscission,but how developmental signals mediate auxin transport in abscission remains ***,we reveal the role of the transcription factor BEL1-LIKE HOMEODOMAIN11(SlBEL11)in regulating auxin transport during abscission in tomato(Solanum lycopersicum).SlBEL11 is highly expressed in the fruit abscission zone,and its expression increases during fruit *** of SlBEL11 expression by RNA interference(RNAi)caused premature fruit drop at the breaker(Br)and 3d post-breaker(Br+3)stages of fruit *** and metabolome analysis of SlBEL11-RNAi lines revealed impaired flavonoid biosynthesis and decreased levels of most flavonoids,especially quercetin,which functions as an auxin transport *** suggested that SlBEL11 prevents premature fruit abscission by modulating auxin efflux from fruits,which is crucial for the formation of an auxin response ***,quercetin treatment suppressed premature fruit drop in SlBEL11-RNAi *** affinity purification sequencing(DAP-seq)analysis indicated that SlBEL11 induced expression of the transcription factor gene SlMYB111 by directly binding to its *** immunoprecipitation-quantitative polymerase chain reaction and electrophoretic mobility shift assay showed that *** MYELOBLASTOSIS VIRAL ONCOGENE HOMOLOG111(SlMYB111)induces the expression of the core flavonoid biosynthesis genes SlCHS1,SlCHI,SlF3H,and SlFLS by directly binding to their *** findings suggest that the SlBEL11-SlMYB111 module modulates flavonoid biosynthesis to fine-tune auxin efflux from fruits and thus maintain an auxin response gradient in the pedicel,thereby preventing premature fruit drop.

Mutations in mitochondrial DNA(mtDNA)are maternally inherited and have the potential to cause severe *** replacement therapies,including spindle,polar body,and pronuclear transfers,are promising strategies for preventing the hereditary transmission of mtDNA *** pronuclear transfer has been used to generate mitochondrial replacement mouse models and human embryos,its application in non-human primates has not been previously *** this study,we successfully generated four healthy cynomolgus monkeys(Macaca fascicularis)via female pronuclear *** individuals all survived for more than two years and exhibited minimal mtDNA carryover(3.8%–6.7%),as well as relatively stable mtDNA heteroplasmy dynamics during *** successful establishment of this nonhuman primate model highlights the considerable potential of pronuclear transfer in reducing the risk of inherited mtDNA diseases and provides a valuable preclinical research model for advancing mitochondrial replacement therapies in humans.

利用印迹-解析电喷雾/光电离质谱成像（IM-DESI/PI-MSI）技术对初烤后干烟叶不同部位化学成分的空间分布进行了研究，获得了烟叶中不同化学成分的纵/横向相对含量分布规律。采用印迹方法可将烟叶内部化合物转移至多孔聚四氟乙烯板的表面，实现对烟叶内部物质的检测和可视化分析，并且经解析电喷雾后，光电离质谱成像技术具有对植物样本的无极性歧视、软电离和高电离效率等优点，可同时检测和成像烟叶样本中丰富的化合物。基于高分辨质谱共鉴定出40种化学成分，包括植物碱、氨基酸、糖类、酸、酮和酚等。实验结果表明，烟草的代表性化学成分如植物碱、氨基酸和糖类等，从纵向分析主要分布在近叶尖处，从横向分析主要分布在烟叶左右边缘；并检测出Amadori化合物（1-脱氧-1-L-脯氨酸-D-果糖），发现其含量变化与游离氨基酸（脯氨酸）的含量变化趋势总体一致。此外，对烟草的气候斑点病区域进行了成像研究，发现化合物在气候斑区域呈特异性分布。本方法在烟叶生长应激研究方面具有优势。
