Omentin-1 prevents inflammation-induced osteoporosis by downregulating the pro-inflammatory cytokines

Osteoporosis is a frequent complication of chronic inflammatory diseases and increases in the pro-inflammatory cytokines make an important contribution to bone loss by promoting bone resorption and impairing bone formation. Omentin-1 is a newly identified adipocytokine that has anti-inflammatory effects, but little is known about the role of omentin-1 in inflammatory osteoporosis. Here we generated global omentin-1 knockout（omentin-1^-/-） mice and demonstrated that depletion of omentin-1 induces inflammatory bone loss-like phenotypes in mice, as defined by abnormally elevated pro-inflammatory cytokines, increased osteoclast formation and bone tissue destruction, as well as impaired osteogenic activities. Using an inflammatory cell model induced by tumor necrosis factor-α（TNF-α）, we determined that recombinant omentin-1 reduces the production of proinflammatory factors in the TNF-α-activated macrophages, and suppresses their anti-osteoblastic and pro-osteoclastic abilities. In the magnesium silicate-induced inflammatory osteoporosis mouse model, the systemic administration of adenoviral-delivered omentin-1 significantly protects from osteoporotic bone loss and inflammation. Our study suggests that omentin-1 can be used as a promising therapeutic agent for the prevention or treatment of inflammatory bone diseases by downregulating the proinflammatory cytokines.

In the independent electro-hydrogen system(IEHS)with hybrid energy storage(HESS),achieving optimal scheduling is ***,it presents a challenge due to the significant deviations in values ofmultiple optimization objective functions caused by their physical *** deviations seriously affect the scheduling process.A novel standardization fusion method has been established to address this issue by analyzing the variation process of each objective function’s *** optimal scheduling results of IEHS with HESS indicate that the economy and overall energy loss can be improved 2–3 times under different optimization *** proposed method better balances all optimization objective functions and reduces the impact of their *** the cost of BESS decreases by approximately 30%,its participation deepens by about 1 ***,if the price of the electrolyzer is less than 15￥/kWh or if the cost of the fuel cell drops below 4￥/kWh,their participation will increase *** study aims to provide a more reasonable approach to solving multi-objective optimization problems.

Osteoporosis is a debilitating bone disease affecting millions of people. Here, we used human urine-derived stem cells(USCs),which were noninvasively harvested from unlimited and easily available urine, as a "factory" to obtain extracellular vesicles(USCEVs) and demonstrated that the systemic injection of USC-EVs effectively alleviates bone loss and maintains bone strength in osteoporotic mice by enhancing osteoblastic bone formation and suppressing osteoclastic bone resorption. More importantly, the anti-osteoporotic properties of USC-EVs are not notably disrupted by the age, gender, or health condition(with or without osteoporosis) of the USC donor. Mechanistic studies determined that collagen triple-helix repeat containing 1(CTHRC1) and osteoprotegerin(OPG) proteins are enriched in USC-EVs and required for USC-EV-induced pro-osteogenic and anti-osteoclastic effects. Our results suggest that autologous USC-EVs represent a promising novel therapeutic agent for osteoporosis by promoting osteogenesis and inhibiting osteoclastogenesis by transferring CTHRC1 and OPG.

Background:Autophagy-associated long non-coding RNAs(aalncRNAs)take an important position in the tumorigeness of lung cancer,but current researches have not systematically investigated autophagy-associated lncRNAs in lung adenocarcinoma(LUAD).Methods:In this research,RNA-sequences of LUAD patients were downloaded from the TCGA and autophagy-associated genes were obtained from the GSEA *** Pearson's test was conducted to find the correlation between autophagy-associated lncRNAs and autophagy-associated *** with prognostic significance were identified by using Cox and LASSO regression analysis in R,*** score model was built to estimate prognosis-associated ***:A risk score model was established according to the expressions of 7 aalncRNAs(RP11-102K13.5,RP11-1029J19.4,LINC00942,KLHL7-AS1,AC092198.1,C20orf197,LINC01116),and low-risk group was found to have a better prognosis(P<0.001).Next,single gene expression survival analysis show that 4 out of these lncRNAs were significantly associated with the survival of *** addition,the AUC value of model reached 0.724,demonstrating the good predictive ability of the ***:These aalncRNAs in LUAD might possibly offered biological markers for the diagnosis and therapy of lung adenocarcinoma.

In the pathogenesis of major depressive disorder, chronic stress-related neuroinflammation hinders favorable prognosis and antidepressant response. Mitochondrial DNA may be an inflammatory trigger, after its release from stress-induced dysfunctional central nervous system mitochondria into peripheral circulation. This evidence supports the potential use of peripheral mitochondrial DNA as a neuroinflammatory biomarker for the diagnosis and treatment of major depressive disorder. Herein, we critically review the neuroinflammation theory in major depressive disorder, providing compelling evidence that mitochondrial DNA release acts as a critical biological substrate, and that it constitutes the neuroinflammatory disease pathway. After its release, mitochondrial DNA can be carried in the exosomes and transported to extracellular spaces in the central nervous system and peripheral circulation. Detectable exosomes render encaged mitochondrial DNA relatively stable. This mitochondrial DNA in peripheral circulation can thus be directly detected in clinical practice. These characteristics illustrate the potential for mitochondrial DNA to serve as an innovative clinical biomarker and molecular treatment target for major depressive disorder. This review also highlights the future potential value of clinical applications combining mitochondrial DNA with a panel of other biomarkers, to improve diagnostic precision in major depressive disorder.

Due to increasing morbidity worldwide,fractures are becoming an emerging public health *** study aimed to investigate the effect of metformin on the healing of osteoporotic as well as normal *** H vessels have recently been identified as a bone-specific vascular subtype that supports ***,we show that metformin accelerated fracture healing in both osteoporotic and normal ***,metformin promoted angiogenesis in vitro under hypoxia as well as type H vessel formation throughout fracture ***,metformin increased the expression of HIF-1α,an important positive regulator of type H vessel formation,by inhibiting the expression of YAP1/TAZ in calluses and hypoxia-cultured human microvascular endothelial cells(HMECs).The results of HIF-1αor YAP1/TAZ interference in hypoxia-cultured HMECs using si RNA further suggested that the enhancement of HIF-1αand its target genes by metformin is primarily through YAP1/TAZ ***,overexpression of YAP1/TAZ partially counteracted the effect of metformin in promoting type H vessel-induced angiogenesis-osteogenesis coupling during fracture *** summary,our findings suggest that metformin has the potential to be a therapeutic agent for fractures by promoting type H vessel formation through YAP1/TAZ inhibition.

Exploring efficient and nonprecious metal electrocatalysts of oxygen reduction reaction(ORR)and oxygen evolution reaction(OER)is crucial for developing rechargeable zinc-air batteries(ZABs).Herein,an alloying-degree control strategy was employed to fabricate nitrogen-doped carbon sphere(NCS)decorated with dual-phase Co/Co_(7)Fe_(3)heterojunctions(CoFe@NCS).The phase composition of materials has been adjusted by controlling the alloying *** optimal CoFe_(0.08)@NCS electrocatalyst displays a half-wave potential of 0.80 V for ORR and an overpotential of 283 mV at 10 mA·cm^(-2)for OER in an alkaline *** intriguing bifunctional electrocatalytic activity and durability is attributed to the hierarchically porous structure and interfacial electron coupling of highly-active Co_(7)Fe_(3)alloy and metallic Co *** the CoFe_(0.08)@NCS material is used as air-cathode catalyst of rechargeable liquid-state zinc-air battery(ZAB),the device shows a high peak power-density(157 mW·cm^(-2))and maintains a stable voltage gap over 150 h,outperforming those of the benchmark(Pt/C+RuO_(2))-based *** particular,the as-fabricated solid-state flexible ZAB delivers a reliable compatibility under different bending *** work provides a promising strategy to develop metal/alloy-based electrocatalysts for the application in renewable energy conversion technologies.

Analysis on Dynamic Heterosis for Allelopathy in Rice Under Different Environment Conditions

In this study, 5 parental rice varieties with different allelopathic potentials were employed in diallel cross [P(P+1)/2] to get a set of genetic materials including parental lines and two generations of F1s. The dynamic heterosis for allelopathy in rice under different environmental conditions, was analyzed by using additive-dominant developmental genetic model. The results indicated that heterosis in both F1 and F2 showed inhibitory effects on shoot and root length of receiver plant(Lactuca sativa L.). Heterosis over mid-parent based on population mean(HMP)in F2 was lower than that in differental environmental conditions, showing 1/2 HMP in F1 The heterosis in rice allelopathy was much higher under the field environmental conditions with lower temperature and weaker sunlight than that under favorable environment, implying that the allel-opthic potential could be increased by stress environment. This finding interpreted the genetic reason that plant could produce more allelochemicals under unfavorable environment.

Studies on the Developmental Genetics of Tiller Number in Three-line Indica Hybrid Rice

Following NCI design, the developmental genetic behavior of tiller number (TN) in three-line indica hybrid rice was studied using additive-dominance developmental genetic models and the corresponding statistical methods. The results showed that dominance effects were predominant for TN. The expression of those additive effects were affected by environment and genotype interaction, but the expression of dominance effects were not affected. Heterosis was the strongest in the middle developmental periods of TN. Additive effects and dominance effects were selectively expressed throughout in the entire tillering developmental stage. Analysis of genetic correlation between TN at different stages and the productive panicles indicated that a close correlation appeared earlier in the populations with higher heterosis than in those with less heterosis. Utilization of heterosis at the middle tillering stage might enhance the final biomass but reduce the percentage of productive panicles.
