Discontinuity development patterns and the challenges for 3D discrete fracture network modeling on complicated exposed rock surfaces

Natural slopes usually display complicated exposed rock surfaces that are characterized by complex and substantial terrain undulation and ubiquitous undesirable phenomena such as vegetation cover and *** study presents a systematic outcrop research of fracture pattern variations in a complicated rock slope,and the qualitative and quantitative study of the complex phenomena impact on threedimensional(3D)discrete fracture network(DFN)*** the studies of the outcrop fracture pattern have been so far focused on local variations,thus,we put forward a statistical analysis of global *** entire outcrop is partitioned into several subzones,and the subzone-scale variability of fracture geometric properties is analyzed(including the orientation,the density,and the trace length).The results reveal significant variations in fracture characteristics(such as the concentrative degree,the average orientation,the density,and the trace length)among different ***,the density of fracture sets,which is approximately parallel to the slope surface,exhibits a notably higher value compared to other fracture sets across all *** improve the accuracy of the DFN modeling,the effects of three common phenomena resulting from vegetation and rockfalls are qualitatively analyzed and the corresponding quantitative data processing solutions are ***,the 3D fracture geometric parameters are determined for different areas of the high-steep rock slope in terms of the subzone *** results show significant variations in the same set of 3D fracture parameters across different regions with density differing by up to tenfold and mean trace length exhibiting differences of 3e4 *** study results present precise geological structural information,improve modeling accuracy,and provide practical solutions for addressing complex outcrop issues.

The CRISPR/Cas9 technology revolutionizes targeted gene knockout in diverse organisms including ***,screening edited alleles,particularly those with multiplex editing,from herbicide-or antibiotic-resistant transgenic plants and segregating out the Cas9 transgene represent two laborious *** solutions to facilitate these processes rely on different selection ***,by taking advantage of the opposite functions of a D-amino acid oxidase(DAO)in detoxifying D-serine and in metabolizing non-toxic D-valine to a cytotoxic product,we develop a DAO-based selection system that simultaneously enables the enrichment of multigene edited alleles and elimination of Cas9-containing progeny in Arabidopsis *** five DAOs tested in Escherichia coli,the one encoded by Trigonopsis variabilis(TvDAO)could confer slightly stronger D-serine resistance than other *** expression of TvDAO in Arabidopsis allowed a clear distinction between transgenic and nontransgenic plants in both D-serine-conditioned positive selection and D-valine-conditioned negative *** a proof of concept,we combined CRISPR-induced single-strand annealing repair of a dead TvDAO with D-serine-based positive selection to help identify transgenic plants with multiplex editing,where D-serine-resistant plants exhibited considerably higher co-editing frequencies at three endogenous target genes than those selected by ***,D-valine-based negative selection successfully removed Cas9 and TvDAO transgenes from the survival offspring carrying inherited ***,this work provides a novel strategy to ease CRISPR mutant identification and Cas9 transgene elimination using a single selection marker,which promises more efficient and simplified multiplex CRISPR editing in plants.

In recent years,frequent fire disasters have led to enormous damage in *** firefighting rescues can minimize the losses caused by *** the rescue processes,the travel time of fire trucks can be severely affected by traffic conditions,changing the effective coverage of fire ***,it is still challenging to determine the effective coverage of fire stations considering dynamic traffic *** paper addresses this issue by combining the traveling time calculationmodelwith the effective coverage *** addition,it proposes a new index of total effective coverage area(TECA)based on the time-weighted average of the effective coverage area(ECA)to evaluate the urban fire *** also selects China as the case study to validate the feasibility of the models,a fire station(FS-JX)in ***-JX station and its surrounding 9,117 fire risk points are selected as the fire service supply and demand points,respectively.A total of 196 simulation scenarios throughout a consecutiveweek are ***,1,933,815 sets of valid sample data are *** results showed that the TECA of FS-JX is 3.27 km^(2),which is far below the standard requirement of 7.00 km^(2) due to the traffic *** visualization results showed that three rivers around FS-JX interrupt the continuity of its effective *** proposed method can provide data support to optimize the locations of fire stations by accurately and dynamically determining the effective coverage of fire stations.

Objective: To investigate the main components and potential mechanism of Shuxuening Injection(SXNI) in the treatment of myocardial ischemia-reperfusion injury(MIRI) through network pharmacology and in vivo research. Methods: The Traditional Chinese Medicine Systems Pharmacology(TCMSP) and Pharm Mapper databases were used to extract and evaluate the effective components of Ginkgo biloba leaves, the main component of SXNI. The Online Mendelian Inheritance in Man(OMIM) and Gene Cards databases were searched for disease targets and obtain the drug target and disease target intersections. The active ingredient-target network was built using Cytoscape 3.9.1 software. The STRING database, Metascape online platform, and R language were used to obtain the key targets and signaling pathways of the anti-MIRI effects of SXNI. In order to verify the therapeutic effect of different concentrations of SXNI on MIRI in rats, 60 rats were first divided into 5 groups according to random number table method: the sham operation group, the model group, SXNI low-dose(3.68 mg/kg), medium-dose(7.35 mg/kg), and high-dose(14.7 mg/kg) groups, with 12 rats in each group. Then, another 60 rats were randomly divided into 5 groups: the sham operation group, the model group, SXNI group(14.7 mg/kg), SXNI+LY294002 group,and LY294002 group, with 12 rats in each group. The drug was then administered intraperitoneally at body weight for 14 days. The main biological processes were validated using in vivo testing. Evans blue/triphenyltetrazolium chloride(TTC) double staining, hematoxylin-eosin(HE) staining, terminal deoxynucleotidyl transferase d UTP nick end labeling(TUNEL) assay, enzyme-linked immunosorbent assay(EliSA), and Western blot analysis were used to investigate the efficacy and mechanism of SXNI in MIRI rats. We have found recently that two-step intrinsic hydroxyl radical(·OH)-dependent chemiluminescence(CL)could be produced by carcinogenic tetrahaloquinone and H_(2)O_(2).However,the first-step CL was too fast to clearly detect the stepwise generation of·OH and CL,and to distinguish the exact dividing point between the first-step and second-step *** we found that,extremely clear two-step intrinsic CL could be produced by the relative slow reaction of tetrabromohydroquinone(TBHQ)with H_(2)O_(2),which was directly dependent on the two-step·OH ***,the second-step,but not the first-step CL production of TBHQ/H_(2)O_(2)(CRET donor)was markedly enhanced by fluorescein(a typical xanthene dye,CRET acceptor)through a unique chemiluminescence resonance energy transfer(CRET)*** novel CRET system of TBHQ/H_(2)O_(2)/fluorescein was successfully applied for the sensitive detection of TBHQ with the detection limit as low as 2.5μmol/*** findings will help to develop more sensitive and highly efficient CL or CRET systems and specific CL sensor to detect the carcinogenic haloquinones,which may have broad environmental applications.
