A preliminary study of imaging paclitaxel-induced tumor apoptosis with 99Tcm-His10-Annexin V

Backgroud in tumors the process of apoptosis occurs over an interval of time after chemotherapy, it is important to determine the best time for detecting apoptosis by in vivo imaging. In this study, we evaluated the dynamics and feasibility of imaging non-small cell lung cancer （NSCLC） apoptosis induced by paclitaxel treatment using a 99Tcm-labeled Annexin V recombinant with ten consecutive histidines （Hislo-Annexin V） in a mouse model. Methods 99Tcm-His10-Annexin V was prepared by one step direct labeling; radio-chemical purity （RCP） and radio-stability was tested. The binding of 99Tcm-His10-Annexin V to apoptotic cells was validated in vitro using camptothecin-induced Jurkat cells. In vivo bio-distribution was determined in mice by dissection. The human H460 NSCLC tumor cell line （H460） tumor-bearing mice were treated with intravenous paclitaxel 24, 48 and 72 hours later. 99Tcm-His10-Annexin V was injected intravenously, and planar images were acquired at 2, 4 and 6 hours post-injection on a dual-head gamma camera fitted with a pinhole collimator. Tumor-to-normal tissue ratios （T/NT） were calculated by ROI analysis and they reflected specific binding of 99Tcm-His10-Annexin V. Mice were sacrificed after imaging. Caspase-3, as the apoptosis detector, was determined by flow cytometry, and DNA fragmentation was analyzed by the terminal deoxynucleotidytransferase mediated dUTP nick-end labeling （TUNEL） assay. Nonspecific accumulation of protein was estimated using bovine serum albumin （BSA）. The imaging data were correlated with TUNEL-positive nuclei and caspase-3 activity. Results 99Tcm-His10-Annexin V had a RCP 〉98% and high stability 2 hours after radio-labeling, and it could bind to apoptotic cells with high affinity. Bio-distribution of 99Tcm-His10-Annexin V showed predominant uptake in kidney, relatively low uptake in myocardium, liver and gastrointestinal tract, and rapid clearance from blood and kidney was observed. A multiscale adaptive framework based on convolutional neural network: Application to fluid catalytic cracking product yield prediction

Since chemical processes are highly non-linear and multiscale, it is vital to deeply mine the multiscale coupling relationships embedded in the massive process data for the prediction and anomaly tracing of crucial process parameters and production indicators. While the integrated method of adaptive signal decomposition combined with time series models could effectively predict process variables, it does have limitations in capturing the high-frequency detail of the operation state when applied to complex chemical processes. In light of this, a novel Multiscale Multi-radius Multi-step Convolutional Neural Network(Msrt Net) is proposed for mining spatiotemporal multiscale information. First, the industrial data from the Fluid Catalytic Cracking(FCC) process decomposition using Complete Ensemble Empirical Mode Decomposition with Adaptive Noise(CEEMDAN) extract the multi-energy scale information of the feature subset. Then, convolution kernels with varying stride and padding structures are established to decouple the long-period operation process information encapsulated within the multi-energy scale data. Finally, a reconciliation network is trained to reconstruct the multiscale prediction results and obtain the final output. Msrt Net is initially assessed for its capability to untangle the spatiotemporal multiscale relationships among variables in the Tennessee Eastman Process(TEP). Subsequently, the performance of Msrt Net is evaluated in predicting product yield for a 2.80 × 10～6 t/a FCC unit, taking diesel and gasoline yield as examples. In conclusion, Msrt Net can decouple and effectively extract spatiotemporal multiscale information from chemical process data and achieve a approximately reduction of 30% in prediction error compared to other time-series models. Furthermore, its robustness and transferability underscore its promising potential for broader applications.

Enhanced Crystallization of Poly(butylene adipate-co-terephthalate)by a Self-assembly Nucleating Agent

Poly(butylene adipate-co-terephthalate)(PBAT)is a promising biodegradable flexible polymer but suffers from slow crystallization rate,making it less attractive for some applications like the injection-molded products in comparison with low-density polyethylene(LDPE).This work aimed to accelerate the crystallization of PBAT by adding a self-assembly nucleating agent octamethylenedicarboxylic dibenzoylhydrazide(OMBH).PBAT/OMBH composites with various OMBH contents(0 wt%,0.5 wt%,0.7 wt%,1 wt%,2 wt%,3 wt%and 5 wt%)were prepared through *** effect of OMBH on the crystallization behavior,morphologies and mechanical properties of PBAT was *** highest nucleation efficiency value of 59.6%was achieved for PBAT with 0.7 wt%OMBH,much higher than that of 22.7%for PBAT with 0.7 wt%*** force microscopy results showed that OMBH formed fine fibers and induced the formation of transcrystalline layers of *** transform infrared spectroscopy(FTIR)combined with two-dimensional correlation spectra suggested that the intermolecular dipole-dipole N—H…O=C interactions but not hydrogen bond between OMBH and PBAT promoted the crystallization of PBAT in the initial period of *** presence of OMBH did not change the crystal form of PBAT but had positive contribution in enhancing its crystallinity and mechanical *** work is essential for preparing PBAT with high crystallization rate,enhancing its potential applications in injection-molded products.

Dynamic performance and parameter optimization of a half-vehicle system coupled with an inerter-based X-structure nonlinear energy sink

Inspired by the demand of improving the riding comfort and meeting the lightweight design of the vehicle, an inerter-based X-structure nonlinear energy sink(IXNES) is proposed and applied in the half-vehicle system to enhance the dynamic performance. The X-structure is used as a mechanism to realize the nonlinear stiffness characteristic of the NES, which can realize the flexibility, adjustability, high efficiency, and easy operation of nonlinear stiffness, and is convenient to apply in the vehicle suspension, and the inerter is applied to replacing the mass of the NES based on the mass amplification characteristic. The dynamic model of the half-vehicle system coupled with the IX-NES is established with the Lagrange theory, and the harmonic balance method(HBM) and the pseudo-arc-length method(PALM) are used to obtain the dynamic response under road harmonic excitation. The corresponding dynamic performance under road harmonic and random excitation is evaluated by six performance indices, and compared with that of the original half-vehicle system to show the benefits of the IX-NES. Furthermore, the structural parameters of the IX-NES are optimized with the genetic algorithm. The results show that for road harmonic and random excitation, using the IX-NES can greatly reduce the resonance peaks and root mean square(RMS) values of the front and rear suspension deflections and the front and rear dynamic tire loads, while the resonance peaks and RMS values of the vehicle body vertical and pitching accelerations are slightly *** the structural parameters of the IX-NES are optimized, the vehicle body vertical and pitching accelerations of the half-vehicle system could reduce by 2.41% and 1.16%,respectively, and the other dynamic performance indices are within the reasonable ***, the IX-NES combines the advantages of the inerter, X-structure, and NES, which improves the dynamic performance of the half-vehicle system and provides an effective option for vibrati

Multi-Robot Collaborative Hunting in Cluttered Environments With Obstacle-Avoiding Voronoi Cells

This work proposes an online collaborative hunting strategy for multi-robot systems based on obstacle-avoiding Voronoi cells in a complex dynamic environment. This involves firstly designing the construction method using a support vector machine(SVM) based on the definition of buffered Voronoi cells(BVCs). Based on the safe collision-free region of the robots, the boundary weights between the robots and the obstacles are dynamically updated such that the robots are tangent to the buffered Voronoi safety areas without intersecting with the obstacles. Then, the robots are controlled to move within their own buffered Voronoi safety area to achieve collision-avoidance with other robots and obstacles. The next step involves proposing a hunting method that optimizes collaboration between the pursuers and evaders. Some hunting points are generated and distributed evenly around a circle. Next, the pursuers are assigned to match the optimal points based on the Hungarian ***, a hunting controller is designed to improve the containment capability and minimize containment time based on collision risk. Finally, simulation results have demonstrated that the proposed cooperative hunting method is more competitive in terms of time and travel distance.
