Rasch validation of the Chinese parent-child interaction scale (CPCIS)

Background Proper parent-child interaction is crucial for child development, but an assessment tool in Chinese is currently lacking. This study aimed to develop and validate a parent-reported parent-child interaction scale for Chinese preschool children. Methods The Chinese parent-child interaction scale (CPCIS) was designed by an expert panel based on the literature and clinical observations in the Chinese context. The initial CPCIS had 14 parent-child interactive activity items. Psychometric properties of the CPCIS were examined using the Rasch model and confirmatory factor analysis (CFA). Convergent validity was investigated by the associations between CPCIS and family income, maternal education level, and children's school readiness. Results The study recruited 567 Chinese parent-child pairs from diverse socioeconomic backgrounds, who completed the CPCIS. Six out of the 14 items in the initial CPCIS were dropped due to suboptimal fi t values. The refined 8-item CPCIS was shown to be valid and reliable by Rasch models and CFA. The person separation reliability and Cronbach's α of the CPCIS were 0.81 and 0.82, respectively. The CPCIS scores were positively associated with family's socioeconomic status (η2 = 0.05,P < 0.001), maternal education level (η2 = 0.08,P < 0.001), and children's school readiness (η2 = 0.01,P < 0.01). Conclusion CPCIS is an easily administered, valid, and reliable tool for the assessment of parent-child interactions in Chinese families.

Effects of Logging on the Genetic Diversity of Quercus tiaoloshanica chun et ko in a Tropical Montane Forest of Hainan Island, Southern China

Quercus tiaoloshanica chun et ko, which has a narrow range of distribution, is one of the important endemic species of the tropical montane rain forest on Hainan Island, southern China. Long-term logging and habitat destruction have resulted in population decline and distribution retreat of Q. tiaoloshanica. To determine the impact of logging on the genetic diversity of Q. tiaoloshanica, the authors investigated the genetic structures using amplified fragment length polymorphism markers in four regenerated stands after logging and in one unlogged stand. Compared with the unlogged stand, the effective number of alleles per locus dropped by 1.0% in selective logging stands and by 2.0% in clear logging stands, corresponding to reductions of 3.8% and 5.2%, respectively, in mean Nei＇s gene diversity and 2.9% and 3.5%, respectively, in mean Shannon diversity index. No substantial genetic erosion was detected in any of the regenerated stands owing to the high tree density and high heterogeneity of the Q. tiaoloshanica stands investigated. Meanwhile, there was no natural regeneration of the species observed in a Dacrydium pierrei Hickel plantation 700 m away from the regenerated stands, suggesting the limited ability of seed dispersal of Q. tiaoloshanica. Clear logging should be undertaken cautiously because the total number of plant species dropped by 15.2% in the clear-logged stands compared with the unlogged stand. To conserve the genetic diversity of this species, as well as the plant biodiversity of tropical forests, the habitats of Q. tiaoloshanica should be protected against exploitation in terms of agricultural or other forms of land use, and some mature trees should be preserved as seed sources to maintain an adequate regeneration base for this species in the management of logging.

Background: Obesity is a fundamental factor in metabolic disorders such as hyperlipidemia, insulin resistance, fatty liver, and atherosclerosis. However, effective preventive measures are still lacking. This study aimed to investigate different surgical protocols for removing partial adipose tissue before the onset of obesity and determine whether, and by which protocol, preliminary adipose removal could exert potent preventive effects against diet-induced metabolic ***: Male low-density lipoprotein receptor (LDL-R) knockout (ko) mice were randomly divided into four groups and subjected to epididymal fat removal (Epi-FR) surgery, subcutaneous fat removal (suQ-FR) surgery, both subcutaneous and epididymal fat removal (Epi + suQ-FR) surgery, or sham-operation. After 1 week of recovery, all mice were given a high-fat diet (HFD) for 10 weeks to induce metabolic ***: In the Epi-FR group and the sham-operated group, the mean numbers of the residual subcutaneous fat were 28.59 mg/g and 18.56 mg/g, respectively. The expression of relative genes such asPparg, Cebpa, Dgat2, Fabp4 and Cd36 in the residual subcutaneous fat increased 2.62, 3.90, 3.11, 2.06, 1.78 times in the Epi-FR group compared with that in the sham-operated group. Whereas in the other fat-removal groups, the residual fat depots had no significant change in either size or gene expression, as compared with those of the sham-operated group. Plasma lipid and glucose levels and insulin sensitivity, as detected by the glucose tolerance test, were not significantly alleviated in the three fat removal groups. Liver mass or lipid content was not attenuated in any of the three fat removal groups. BACKGROUND: Although perioperative portal vein (PV) stent implantation is an effective treatment for steno-occlusive disease in adult living donor liver transplantation (LDLT) re-cipients, we experienced high incidence of biliary anastomotic strictures (BAS) after PV stenting. In this study, we sought to clarify the relation between BAS and PV stenting and to suggest the possible mechanism of BAS and measures to reduce its in-cidence. METHODS: We retrospectively analyzed 44 LDLT recipients who underwent PV stent implantation across the line of PV anastomosis regardless of the location of steno-occlusion (stent group) and their matched controls (non-stented LDLT recipients, n=131). RESULTS: The incidence of BAS was higher in patients in the stent group than that in the control group (43.2% vs 17.6%, P=0.001). Cumulative 6-month and 1-, 2- and 5-year BAS rates were 31.8%, 34.1%, 41.4% and 43.2%, respectively, in the stent group and 13.0%, 13.8%, 16.1% and 17.8%, respectively, in the control group (P=0.001). Multivariate analysis revealed that PV stenting was an independent risk factor for BAS. CONCLUSIONS: Although PV stent implantation is a reliable treatment modality for steno-occlusive PV in adult LDLT recipients, innovative methods to prevent the PV stent from crossing the line of PV anastomosis may be necessary to reduce the incidence of postoperative BAS.

N, N-dimethyltryptamine, a natural psychedelic, rapidly improves spatial memory and hippocampal longterm potentiation: a potential rapid-acting therapy for dementia-associated wandering

There is no fast-acting treatment strategies against Alzheimer′s disease(AD), in particular dementia-related wandering. N, N-dimethyltryptamine(DMT) is a natural psychedelic that may have rapid-onset nootropic effects. In this study, 5 × FAD transgenic mice which recapitulated amyloid neuropathological features of AD received one single injection of 6 or 12 mg · kg-1DMT and tested at 0.5, 1, and 2 h thereafter in Y-maze for spatial memory. 5×FAD transgenic mice exhibited pronounced decreases in time spent, number entered, and distance travelled in the novel arm of Y-maze. DMT at 12 mg · kg-1partially or completely reversed the three behavioral indices at multiple time points, up to 2 h post injection. The rapid-onset behavioral improvement was consistent with pharmacokinetic analysis of DMT, showing approximately 30 min to reach the maximum concentration in the brain tissue. The transgenic mice also displayed dramatically impaired hippocampal long-term potentiation(LTP), an electrophysiological feature of memory formation and consolidation. DMT potently enhanced LTP and restored intracellular calcium activity, expression and phosphorylation of calcium/calmodulin-dependent protein kinase Ⅱ(CaMKⅡ) and AMPA-type glutamate receptor1(GluR1), the two key calcium-activated mediators involved in LTP induction. Adenosine triphosphate(ATP)is purinergic signalling molecules that are involved in LTP induction and maintenance. DMT rapidly increased mitochondrial ATP dynamics in in vivo and in vitro *** results suggest that DMT rapidly improve spatial memory and hippocampal LTP by restoring the CaMKⅡ-GluR1 signaling pathway and mitochondrial ATP production. It may be served as a fast-acting nootropic agent for the treatment of AD in particular wandering.

Aggravated inflammation and increased expression of cysteinyl leuko-triene receptors in the brain after focal cerebral ischemia in AQP4-deficient mice

Objective Aquaporin-4 （AQP4）, the main water channel protein in the brain, plays a critical role in water homeostasis and brain edema. Here, we investigated its role in the inflammatory responses after focal cerebral ischemia. Methods In AQP4-knockout （ko） and wild-type mice, focal cerebral ischemia was induced by 30 rain of middle cerebral arterial occlusion （MCAO）. Ischemic neuronal injury and cellular inflammatory responses, as well as the expression and localization of cysteinyl leukotriene CysLT2 and CysLT~ receptors, were determined at 24 and 72 h after MCAO. Results AQP4-ko mice showed more neuronal loss, more severe microglial activation and neutrophil infiltration, but less astrocyte proliferation in the brain after MCAO than wild-type mice. In addition, the protein levels of both CysLT1 and CysLT2 receptors were up-regulated in the ischemic brain, and the up-regulation was more pronounced in AQP4-ko mice. The CysLT1 and CysLT2 receptors were primarily localized in neurons, microglia and neutrophils; those localized in microglia and neutrophils were enhanced in AQP4-ko mice. Conclusion AQP4 may play an inhibitory role in postischemic inflammation.

目的探究文冠果油神经酸乙酯对双环己酮草酰二腙(cuprizone,CPZ)诱导的脱髓鞘小鼠脂肪酸、血脂的影响。方法SPF级雌性昆明小鼠50只,除空白组(ko)外,通过在饲料中加入0.25%CPZ饲喂雌性昆明小鼠造模并分为模型组(CPZ)、神经酸乙酯低剂量组(NA-L,0.001912 mL/g)、神经酸乙酯中剂量组(NA-M,0.003824 mL/g)、神经酸乙酯高剂量组(NA-H,0.007648 mL/g),持续5周至实验结束。于造模2周后开始给药至第5周结束后取材,并通过气相色谱仪、血脂仪分别检测文冠果油神经酸乙酯对CPZ诱导的脱髓鞘小鼠脑组织、肝脏、心脏、血清、粪便里的脂肪酸含量及血液中血脂水平的影响。结果与ko组相比,CPZ组脱髓鞘小鼠脑组织、肝脏、心脏中神经酸、芥酸含量显著下降(Pko、CPZ组小鼠血清、粪便中未检测到神经酸和芥酸,NA-L、NA-M、NA-H组小鼠血清、粪便中含有一定量的神经酸和芥酸;与ko组相比,CPZ组血脂升高;与CPZ组相比,NA-L、NA-M、NA-H组均可降低血液中的甘油三酯(triglyceride,TG)含量,升高高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)含量。结论在饲料中添加0.25%CPZ诱导小鼠脱髓鞘会导致小鼠血脂升高,降低小鼠体内神经酸、芥酸含量,脱髓鞘疾病与神经酸的积累有关,脱髓鞘疾病会导致神经酸的供应受损。在这种情况下,文冠果油神经酸乙酯提取物能够增加脱髓鞘小鼠体内神经酸及芥酸含量,提高脑组织不饱和脂肪酸含量,调节脱髓鞘小鼠血脂水平。此外,神经酸、芥酸参与脱髓鞘小鼠体内代谢,并在小鼠体内共存,文冠果油神经酸乙酯提取物可作为脱髓鞘小鼠补充神经酸的外源性物质。
