Mechanism of Key Ingredient of Astragalus membranaceus on Lung Adenocarcinoma via PI3K/AKT Signaling Clarifled by Utilizing Network Pharmacology Approach and Experimental Validation

Objective:To investigate the mechanism of the effect of Astragalus membranaceus(A.membranaceus)on lung adenocarcinoma at the molecular level to elucidate the specific targets according to the network pharmacology approach.Methods:The active components of A.membranaceus and their potential targets were col ected from the Traditional Chinese Medicine Systems Pharmacology Database.Lung adenocarcinoma-associated genes were acquired based on Gene Cards,Online Mendelian Inheritance in man(OMIM),Pharm GKB,and Therapeutic Targets databases.The PI3K/AKT signaling pathway-related genes were obtained using Reactome portal.Networks of"ingredient-target"and"ingredient-target-pathway-disease"were constructed using the Cytoscape3.6.0 software.The relationships among targets were analyzed according protein-protein interaction(PPI)network.Finally,molecular docking was applied to construct the binding conformation between active ingredients and core targets.Cel counting kit 8(CCK8)and Western blot assays were performed to determine the mechanism of the key ingredient of A.membranaceus.Results:A total of 20 active components and their 329 targets,and 7,501 lung adenocarcinoma-related genes and 130 PI3K/AKT signaling pathway-related genes were obtained.According to Venn diagram and PPI network analysis,2 mainly active ingredients,including kaempferol and quercetin,and 6 core targets,including TP53,maPK1,EGF,AKT1,ERBB2,and EGFR,were identified.The two important active ingredients of A.membranaceus,kaempferol and quercetin,exert the therapeutic effect in lung adenocarcinoma partly by acting on the 6 core targets(TP53,maPK1,EGF,AKT1,ERBB2,and EGFR)of PI3K/AKT signaling pathway.Expressions of potential targets in lung adenocarcinoma and normal samples were analyzed by using UALCAN portal and found that ERBB2 was overexpressed in lung adenocarcinoma tissues and upregulation of it correlated with clinicopathological characteristics.Final y,quercetin repressed viabilities of lung adenocarcinoma cells by targeting ERBB2 on PI3K/AKT signalin

Enhanced Phase Transition in Poly(ethylene glycol) Grafted Butene-1 Copolymers

In the present work,a series of poly(ethylene glycol) grafted polybutene-1 copolymers(PB-g-PEG) were designed and successfully synthesized with reaction intermediate method,of which the branch densities and length could be tuned independently.The branch density varied from 0.39 mol% to 1.23 mol% and the branch length was tuned by employing PEG number-averaged molecular weight(Mn) from 350 Da to 750 Da and 1000 Da.The non-isothermal crystallization and phase transition behaviors of these butene-1 copolymers with varying branches were systematically explored by differential scanning calorimeter.The results first showed that compared with the linear polybutene-1 homopolymer,presence of steric 4-[4-(trimethylsilylalkynyl)-phenyl]-1-butene(TMS) groups suppressed crystallization and phase transition of copolymers.Interestingly,the introduction of long poly(ethylene glycol)(PEG) branches with M_(n)=350-1000 Da not only elevated the peak temperature of the non-isothermal crystallization but also enhanced the Ⅱ-Ⅰ phase transition,with respect to copolymers with TMS groups.Furthermore,the quantitative correlations between phase transition kinetics with the branch parameters(density and length) were explored.It was found that the phase transition presents a non-monotonic trend with increasing branch density and branch length.In this case,the optimal branch density and length obtained were 0.77 mol% and 750 Da,respectively.Employing the small angle X-ray scattering and transmission electron microscopy,a direct correlation between the phase transition kinetics and the long periodicity of separated phase was obtained,which may be due to the increased internal stress and facilitated stress transport.

Whispering gallery mode(WGM)microcavities provide increasing opportunities for precision measurement due to their ultrahigh sensitivity,compact size,and fast response.However,the conventional WGM sensors rely on monitoring the changes of a single mode,and the abundant sensing information in WGM transmission spectra has not been fully utilized.Here,empowered by machine learning(ML),we propose and demonstrate an ergodic spectra sensing method in an optofluidic microcavity for high-precision pressure measurement.The developed ML method realizes the analysis of the full features of optical spectra.The prediction accuracy of 99.97%is obtained with the average error as low as 0.32 kPa in the pressure range of 100 kPa via the training and testing stages.We further achieve the real-time readout of arbitrary unknown pressure within the range of measurement,and a prediction accuracy of 99.51%is obtained.Moreover,we demonstrate that the ergodic spectra sensing accuracy is∼11.5%higher than that of simply extracting resonating modes’wavelength.With the high sensitivity and prediction accuracy,this work opens up a new avenue for integrated intelligent optical sensing.

Parametric study of Liutex-based force field models

The motion of fluid consists of different scales of coherent vortical structures.These vortical structures determine the characteristics of fluid motion and are key to understand fluid dynamics.In this paper,we study the fine control method of vortical structures based on the Liutex force field model.This is achieved by constructing a source term using Liutex and directly add it to the Navier-Stokes equations.To investigate the influence of the constructed Liutex force model on vortical structures,a flow past a cylinder at Reynolds number of 100 is numerically studied with different source term magnitude and region.The drag and lift forces on the cylinder,as well as the flow field near and behind the cylinder are compared and analyzed.Results show that Liutex force model can effectively strengthen or weaken the vortical structures based on different purpose.

BACKGROUND Hepatitis B cirrhosis(HBC)is a chronic disease characterized by irreversible diffuse liver damage and aggravated by intestinal microbial imbalance and metabolic dysfunction.Although the relationship between certain single probiotics and HBC has been explored,the impact of the complex ready-to-eat Lactobacillus paracasei N1115(LP N1115)supplement on patients with HBC has not been determined.AIM To compare the changes in the microbiota,inflammatory factor levels,and liver function before and after probiotic treatment in HBC patients.METHODS This study included 160 HBC patients diagnosed at the General Hospital of Ningxia Medical University between October 2018 and December 2020.Patients were randomly divided into an intervention group that received LP N1115 supplementation and routine treatment and a control group that received routine treatment only.Fecal samples were collected at the onset and conclusion of the 12-wk intervention period.The structure of the intestinal microbiota and the levels of serological indicators,such as liver function and inflammatory factors,were assessed.RESULTS Following LP N1115 intervention,the intestinal microbial diversity significantly increased in the intervention group(Pmatory factors(Pmatory factor levels.

Configurational information entropy(CIE)theory was employed to determine the neutron-skin thickness of neutron-rich calcium isotopes.The nuclear density distributions and fragment cross sections in 350 MeV/u ^(40-60)Ca+^(9)Be projectile fragmentation reactions were calculated using a modified statistical abrasion-ablation model.CIE quantities were determined from the nuclear density,isotopic,mass,and charge distributions.The linear correlations between the CIE determined using the isotopic,mass,and charge distributions and the neutron-skin thickness of the projectile nucleus show that CIE provides new methods to extract the neutron-skin thickness of neutron-rich nuclei.

Assessing the effectiveness of nanoscale zero-valent iron particles produced by green tea for Cr(VI)-contaminated groundwater remediation

Nanoscale zero-valent iron particles(NZVI) produced by using green tea(GT) extract as a reductant can remove Cr(Ⅵ) from water effectively,which can be utilized in groundwater remediation.In order to define the reaction mechanism and removal effect in the aquifer,in this study,GT-NZVI particles were prepared and measured by some characterization methods to define their surface performance,and then batch and one-dimensional experiments were carried out to reveal the reaction properties of GT-NZVI and Cr(Ⅵ) in groundwater.The results showed that the prepared GT-NZVI particles were regular spherical with a diameter of 10-20 nm,which could disperse in water stably.The main component of GT-NZVI wasα-Fe with superficial polyphenols as a stabilizer.GT-NZVI suspension had good ability to reduce the Cr(Ⅵ) to Cr(Ⅲ) in water.When the concentration of GT-NZVI was 1 g/L,the removal efficiency of Cr(Ⅵ)with an initial concentration of 100 mg/L reached 92.8% in 1 h reaction.In column tests,GT-NZVI passed through the natural sand column successfully with an average outflow percentage of 71.2%.The simulated in-situ reaction zone(IRZ) with GT-NZVI was used to remediate Cr(Ⅵ) contaminated groundwater.The oufflow concentration of Cr(Ⅵ) kept in 0.14-0.32 mg/L corresponding to the outflow rate below 0.32%within 15 days,and the removal efficiency of Cr(Ⅵ) by IRZ with GT-NZVI decreased with the increase of aquifer medium particle size,groundwater flow rate and ionic strength.Most of Cr(Ⅲ) as reduzate was adsorbed or immobilized on the surface or in the lattice of GT-NZVI,which indicated effective immobilization for chromium.

Exploring the Feasibility of machine Learning to Predict Risk Stratification Within 3 Months in Chest Pain Patients with Suspected NSTE-ACS

Objective We aimed to assess the feasibility and superiority of machine learning(ML)methods to predict the risk of major Adverse Cardiovascular Events(maCEs)in chest pain patients with NSTE-ACS.Methods Enrolled chest pain patients were from two centers,Beijing Anzhen Emergency Chest Pain Center Beijing Bo’ai Hospital,China Rehabilitation Research Center.Five classifiers were used to develop ML models.Accuracy,Precision,Recall,F-Measure and AUC were used to assess the model performance and prediction effect compared with HEART risk scoring system.Ultimately,ML model constructed by Naïve Bayes was employed to predict the occurrence of maCEs.Results According to learning metrics,ML models constructed by different classifiers were superior over HEART(History,ECG,Age,Risk factors,&Troponin)scoring system when predicting acute myocardial infarction(AMI)and all-cause death.However,according to ROC curves and AUC,ML model constructed by different classifiers performed better than HEART scoring system only in prediction for AMI.Among the five ML algorithms,Linear support vector machine(SVC),Naïve Bayes and Logistic regression classifiers stood out with all Accuracy,Precision,Recall and F-Measure from 0.8 to 1.0 for predicting any event,AMI,revascularization and all-cause death(vs.HEART≤0.78),with AUC from 0.88 to 0.98 for predicting any event,AMI and revascularization(vs.HEART≤0.85).ML model developed by Naïve Bayes predicted that suspected acute coronary syndrome(ACS),abnormal electrocardiogram(ECG),elevated hs-cTn I,sex and smoking were risk factors of maCEs.Conclusion Compared with HEART risk scoring system,the superiority of ML method was demonstrated when employing Linear SVC classifier,Naïve Bayes and Logistic.ML method could be a promising method to predict maCEs in chest pain patients with NSTE-ACS.

Three-microRNA signature identified by bioinformatics analysis predicts prognosis of gastric cancer patients

AIM To identify multiple micro RNAs(mi RNAs) for predicting the prognosis of gastric cancer(GC) patients by bioinformatics analysis.METHODS The original microarray dataset GSE93415,which included 20 GC and 20 tumor adjacent normal gastric mucosal tissues,was downloaded from the Gene Expression Omnibus database and used for screening differentially expressed mi RNAs(DEMs).The cutoff criteria were P 2.0.In addition,we acquired the mi RNA expression profiles and clinical information of 361 GC patients from The Cancer Genome Atlas database to assess the prognostic role of the DEMs.The target genes of mi RNAs were predicted using Target Scan,mi RDB,mi RWalk,and DIANA,and then the common target genes were selected for functional enrichment analysis.RESULTS A total of 110 DEMs including 19 up-regulated and 91 down-regulated mi RNAs were identified between 20 pairs of GC and tumor adjacent normal tissues,and the Kaplan-Meier survival analysis found that a threemi RNA signature(mi R-145-3 p,mi R-125 b-5 p,and mi R-99 a-5 p) had an obvious correlation with the survival of GC patients.Furthermore,univariate and multivariate Cox regression analyses indicated that the three-mi RNA signature could be a significant prognostic marker in GC patients.The common target genes of the three mi RNAs are added up to 108 and used for Gene Functional Enrichment analysis.Biological Process and Molecular Function analyses showed that the target genes are involved in cell recognition,gene silencing and nucleic acid binding,transcription factor activity,and transmembrane receptor activity.Cellular Component analysis revealed that the genes are portion of nucleus,chromatin silencing complex,and TORC1/2 complex.Biological Pathway analysis indicated that the genes participate in several cancer-related pathways,such as the focal adhesion,PI3 K,and m TOR signaling pathways.CONCLUSION This study justified that a three-mi RNA signature could play a role in predicting the survival of GC patients.

Liutex force field model applied to three-dimensional flows around a circular cylinder at Re=3900

Recently,the Liutex-based force field models are proposed and applied to some two-dimensional flows to explore the possibility of direct vortex control strategies.In this paper,we applied the Liutex-based centripetal force model to three-dimensional flow around a circular cylinder at Re=3900.The turbulent flows around the three-dimensional cylinder are modeled by the delayed detached-eddy simulations based on the two-equation k-ω shear stress transport(SST)model.The cylinder flow without adding the force field model is simulated in the first place.The statistical flow quantities are compared with experimental data to validate the accuracy of the current numerical models.Then the force field model is applied to the momentum equation in a specific control region near the cylinder to study the change of flow field and hydrodynamic performance.The effect of different control region locations with the same force field strength is discussed to find out the most influenced locations on the drag and lift.Based on the screened control region,the influence source term strength is further studied.Variations on the global statistical flow quantities,local instantaneous three-dimensional flow structures are analyzed and discussed.It is concluded that the Liutex-based force field model can effectively change the hydrodynamic behavior of flow past a cylinder and may provide a new direction for performance improvement and optimization.
