Expression of Embryonic Stem Cell Marker Oct-4 and Its Prognostic Significance in Rectal Adenocarcinoma

Objective： Recent evidence suggests that Oct-4 is highly expressed in several cancers, and its expression contributes to tumor growth. In this study, we investigated the level of Oct-4 expression in rectal adenocarcinoma, and evaluated the prognostic significance of Oct-4 expression in these cases. Methods： The immunohistochemical expression of Oct-4 was evaluated in 52 formalin-fixed paraffin-embedded postoperative rectal adenocarcinoma tissue samples. The impact of the immunoreactivity of Oct-4 in regard to clinical outcome was determined by Kaplan-Meier and log-rank. Results： The expression level of Oct-4 ranged from 0 to 18.5%. There was no significant association between Oct-4 expression and gender （P=0.772）, age （P=0.123）, clinical stage （P=0.391）, and histological grade （P=0.056）. The 3-year local recurrence-free rates with negative and positive expression of Oct-4 were 83.5% and 75.0%, respectively （P=0.583）. The 3-year metastasis-free rates with negative and positive expression of Oct-4 were 88.6% and 61.9%, respectively （P=0.035）. The 3-year overall survival rates with negative and positive expression of Oct-4 were 77.9% and 49.0%, respectively （P=0.037）. Conclusion： The results suggest that embryonic stem cell marker Oct-4 expression may have prognostic significance in patients with rectal adenocarcinoma. However, to confirm this more and larger studies are required.

Blocking NF-kB nuclear translocation leads to p53-related autophagy activation and cell apoptosis

AIM: To investigate the anti-tumor effects of nuclear factor-κB (NF-κB) inhibitor SN50 and related mechanisms of SGC7901 human gastric carcinoma cells. METHODS: MTT assay was used to determine the cytotoxic effects of SN50 in gastric cancer cell line SGC7901. Hoechst 33258 staining was used to detect apoptosis morphological changes after SN50 treatment. Activation of autophagy was monitored with monodansylcadaverine (MDC) staining after SN50 *** staining was used to detect the expression of light chain 3 (LC3). Mitochondrial membrane potential was measured using the fluorescent probe JC-1. Western blotting analysis were used to determine the expression of proteins involved in apoptosis and autophagy including p53, p53 upregulated modulator of apoptosis (PUMA), damage-regulated autophagy modulator (DRAM), LC3 and Beclin 1. We detected the effects of p53-mediated autophagy activation on the apoptosis of SGC7901 cells with the p53 inhibitor pifithrin-α. RESULTS: The viability of SGC7901 cells was inhibited after SN50 treatment. Inductions in the expression of apoptotic protein p53 and PUMA as well as autophagic protein DRAM, LC3 and Beclin 1 were detected with Western blotting analysis. SN50-treated cells exhibited punctuate microtubule-associated protein 1 LC3 in immunoreactivity and MDC-labeled vesicles increased after treatment of SN50 by MDC staining. Collapse of mitochondrial membrane potential Δψ were detected for 6 to 24 h after SN50 treatment. SN50-induced increases in PUMA, DRAM, LC3 and Beclin 1 and cell death were blocked by the p53 specific inhibitor pifithrin-α. CONCLUSION: The anti-tumor activity of NF-κB inhibitors is associated with p53-mediated activation of autophagy.

Effects of LY294002 on the invasiveness of human gastric cancer in vivo in nude mice

AIM: To investigate the effects of class phosphatidy-linositol 3-kinase (PI3K) inhibitor LY294002 on the invasiveness and related mechanisms of implanted tumors of SGC7901 human gastric carcinoma cells in nude mice. METHODS: Nude mice were randomly divided into model control groups and LY294002 treatment groups. On days 5, 10 and 15 after treatment, the inhibitory rate of tumor growth, pathological changes in tumor specimens, expression levels of matrix metalloproteinase (MMP)-2, MMP-9, CD34 [representing microvessel density (MVD)] and vascular endothelial growth factor (VEGF), as well as apoptosis indexes in tumor samples were ***: In this study, we showed that treatingthe tumors with LY294002 could significantly inhibit carcinoma growth by 11.3%, 29.4% and 36.7%, after 5, 10 and 15 d, respectively, compared to the control group. Hematoxylin & eosin staining indicated that the rate of inhibition increased progressively (23.51% ± 3.11%, 43.20% ± 3.27% and 63.28% ± 2.10% at 5, 10 and 15 d, respectively) along with apoptosis. The expression of MMP-2 was also downregulated (from 71.4% ± 1.6% to 47.9% ± 0.7%, 31.9% ± 0.9% and 7.9% ± 0.7%). The same effects were observed in MMP-9 protein expression (from 49.4% ± 1.5% to 36.9% ± 0.4%, 23.5% ± 0.9% and 7.7% ± 0.6%), the mean MVD (from 51.2% ± 3.1% to 41.9% ± 1.5%, 30.9% ± 1.7% and 14.9% ± 0.8%), and the expression of VEGF (from 47.2% ± 3.1% to 25.9% ± 0.5%, 18.6% ± 1.2% and 5.1% ± 0.9%) by immunohistochemical ***: The classPI3K inhibitor LY294002could inhibit the invasiveness of gastric cancer cells by downregulating the expression of MMP-2, MMP-9, and VEGF, and reducing MVD.

Simple instruments facilitating achievement of transanal total mesorectal excision in male patients

AIM To assess the efficacy of a modified approach with transanal total mesorectal excision(ta TME) using simple customized instruments in male patients with low rectal *** A total of 115 male patients with low rectal cancer from December 2006 to August 2015 were retrospectively studied. All patients had a bulky tumor(tumor diameter ≥ 40 mm). Forty-one patients(group A) underwent a classical approach of transabdominal total mesorectal excision(TME) and transanal intersphincteric resection(ISR), and the other 74 patients(group B) underwent a modified approach with transabdominal TME,transanal ISR, and ta TME. Some simple instruments including modified retractors and an anal dilator with a papilionaceous fixture were used to perform ta TME. The operative time, quality of mesorectal excision, circumferential resection margin, local recurrence, and postoperative survival were *** All 115 patients had successful sphincter preservation. The operative time in group B(240 min, range: 160-330 min) was significantly shorter than that in group A(280 min, range: 200-360 min; P = 0.000). Co m pa r e d w it h g r o up A, m o r e c o m p le t e d is t a l mesorectum and total mesorectum were achieved in group B(100% vs 75.6%, P = 0.000; 90.5% vs 70.7%, P = 0.008, respectively). After 46.1 ± 25.6 mo followup, group B had a lower local recurrence rate and higher disease-free survival rate compared with group A, but these differences were not statistically significant(5.4% vs 14.6%, P = 0.093; 79.5% vs 65.1%, P = 0.130). CONCLUSION Retrograde ta TME with simple customized instruments can achieve high-quality TME, and it might be an effective and economical alternative for male patients with bulky tumors.

Effects of small interfering RNA inhibit Class Ⅰ phosphoinositide 3-kinase on human gastric cancer cells

AIM: To investigate the effects of small interfering RNA (siRNA)-mediated inhibition of Class?I?phosphoinositide 3-kinase (Class?I?PI3K) signal transduction on the proliferation, apoptosis, and autophagy of gastric cancer SGC7901 and MGC803 cells.METHODS: We constructed the recombinant replication adenovirus PI3K(I)-RNA interference (RNAi)-green fluorescent protein (GFP) and control adenovirus NC-RNAi-GFP, and infected it into human gastric cancer cells. MTT assay was used to determine the growth rate of the gastric cancer cells. Activation of autophagy was monitored with monodansylcadaverine (MDC) staining after adenovirus PI3K(I)-RNAi-GFP and control adenovirus NC-RNAi-GFP treatment. Immunofluorescence staining was used to detect the expression of microtubule-associated protein 1 light chain 3 (LC3). Mitochondrial membrane potential was measured using the fluorescent probe JC-1. The expression of autophagy was monitored with MDC, LC3 staining, and transmission electron microscopy. Western blotting was used to detect p53, Beclin-1, Bcl-2, and LC3 protein expression in the culture supernatant.RESULTS: The viability of gastric cancer cells was inhibited after siRNA targeting to the Class?I?PI3K blocked Class?I?PI3K signal pathway. MTT assays revealed that, after SGC7901 cancer cells were treated with adenovirus PI3K(I)-RNAi-GFP, the rate of inhibition reached 27.48% ± 2.71% at 24 h, 41.92% ± 2.02% at 48 h, and 50.85% ± 0.91% at 72 h. After MGC803 cancer cells were treated with adenovirus PI3K(I)-RNAi-GFP, the rate of inhibition reached 24.39% ± 0.93% at 24 h, 47.00% ± 0.87% at 48 h, and 70.30% ± 0.86% at 72 h (P < 0.05 compared to control group). It was determined that when 50 MOI, the transfection efficiency was 95% ± 2.4%. Adenovirus PI3K(I)-RNAi-GFP (50 MOI) induced mitochondrial dysfunction and activated cell apoptosis in SGC7901 cells, and the results described here prove that RNAi of Class?I?PI3K induced apoptosis in SGC7901 cells. A multiscale adaptive framework based on convolutional neural network: Application to fluid catalytic cracking product yield prediction

Since chemical processes are highly non-linear and multiscale, it is vital to deeply mine the multiscale coupling relationships embedded in the massive process data for the prediction and anomaly tracing of crucial process parameters and production indicators. While the integrated method of adaptive signal decomposition combined with time series models could effectively predict process variables, it does have limitations in capturing the high-frequency detail of the operation state when applied to complex chemical processes. In light of this, a novel Multiscale Multi-radius Multi-step Convolutional Neural Network(Msrt Net) is proposed for mining spatiotemporal multiscale information. First, the industrial data from the Fluid Catalytic Cracking(FCC) process decomposition using Complete Ensemble Empirical Mode Decomposition with Adaptive Noise(CEEMDAN) extract the multi-energy scale information of the feature subset. Then, convolution kernels with varying stride and padding structures are established to decouple the long-period operation process information encapsulated within the multi-energy scale data. Finally, a reconciliation network is trained to reconstruct the multiscale prediction results and obtain the final output. Msrt Net is initially assessed for its capability to untangle the spatiotemporal multiscale relationships among variables in the Tennessee Eastman Process(TEP). Subsequently, the performance of Msrt Net is evaluated in predicting product yield for a 2.80 × 10～6 t/a FCC unit, taking diesel and gasoline yield as examples. In conclusion, Msrt Net can decouple and effectively extract spatiotemporal multiscale information from chemical process data and achieve a approximately reduction of 30% in prediction error compared to other time-series models. Furthermore, its robustness and transferability underscore its promising potential for broader applications.

Viscoacoustic generalized screen propagator in constant-Q model

When seismic waves propagate through the geological formation, there is a significant loss of energy and a decrease in imaging resolution, because of the viscoacoustic properties of subsurface medium. This profoundly impacts seismic wavefield propagation, imaging and interpretation. To accurately image the true structure of subsurface medium, the consensus among geophysicists is to no longer treat subsurface medium as ideal homogeneous medium, but rather to incorporate the viscoacoustic properties of subsurface medium. Based on the generalized screen propagator using conventional acoustic wave equation(acoustic GSP), our developed method introduces viscoacoustic compensation strategy, and derives a one-way wave generalized screen propagator based on time-fractional viscoacoustic wave equation(viscoacoustic GSP). In numerical experiments, we conducted tests on two-dimensional multi-layer model and the Marmousi model. When comparing with the acoustic GSP using the acoustic data, we found that the imaging results of the viscoacoustic GSP using the viscoacoustic data showed a significant attenuation compensation effect, and achieved imaging results for both algorithms were essentially consistent. However, the imaging results of acoustic GSP using viscoacoustic data showed significant attenuation effects, especially for deep subsurface imaging. This indicates that we have proposed an effective method to compensate the attenuated seismic wavefield. Our application on a set of real seismic data demonstrated that the imaging performance of our proposed method in local areas surpassed that of the conventional acoustic GSP. This suggests that our proposed method holds practical value and can more accurately image real subsurface structures while enhancing imaging resolution compared with the conventional acoustic GSP. Robotic versus laparoscopic surgery for sporadic benign insulinoma:Short-and long-term outcomes

To better understand the failure behaviours and strength of bolt-reinforced blocky rocks,large scale extensive laboratory experiments are carried out on blocky rock-like specimens with and without rockbolt *** results show that both shear failure and tensile failure along joint surfaces are observed but the shear failure is a main controlling factor for the peak strength of the rock mass with and without *** rockbolts are necked and shear deformation simultaneously happens in bolt reinforced rock *** the joint dip angle increases,the joint shear failure becomes more *** number of rockbolts has a significant impact on the peak strain and uniaxial compressive strength(UCS),but little influence on the deformation modulus of the rock *** the Winkler beam model to represent the rockbolt behaviours,an analytical model for the prediction of the strength of boltreinforced blocky rocks is *** agreement between the UCS values predicted by proposed model and obtained from experiments suggest an encouraging performance of the proposed *** addition,the performance of the proposed model is further assessed using published results in the literature,indicating the proposed model can be used effectively in the prediction of UCS of bolt-reinforced blocky rocks.

Surface modification of graphite anode with electroactive matters has been proven of a more practical strategy in enhancing the performance of Li-ion batteries than exploring alternative novel anode ***,rutile TiNbO_(4-x) nanoparticles with a tunnel structure are employed as multifunctional decoration substances in combination with a carbon coating layer to improve the rate and cycle properties of mesocarbon microbeads(MCMBs).As compared to pristine MCMB,the Li^(+)diffusion coefficients of the composite anodes are enhanced due to the synergistic effect of TiNbO_(4-x)@***,the overcharge and voltage polarization of the composite anodes at high rate are obviously minimized due to the current sharing effect of the high-potential TiNbO_(4-x).Moreover,the amorphous Li_(y)TiNbO_(4-x) converted from TiNbO_(4-x) in the initial lithiation process can deliver pseudocapacitive capacity to the composite anodes from the second *** of these functions of TiNbO_(4-x)@Ccoating layer have directly contributed to the improved rate and cycle performance of the MCMB/TiNbO_(4-x)@C composite *** one containing 12.0 wt%TiNbO_(4-x) exhibits a high reversible specific capacity of 118 m Ah·g^(-1)at 10C(1C=372 m A·g^(-1)),together with a high capacity retention of 90.9%after 300 cycles at 3C,which are all much superior to those of pristine MCMB.
