Exploring the impact of non-small cell lung cancer tumor microbiome on the efficacy of chemotherapy and immune checkpoint inhibitors

Background:This study aimed to investigate the potential of intratumoral microbiota,gut microbiome,peripheral blood T-cell subsets,and inflammatory markers as predictive biomarkers for antitumor efficacy in patients with non-small cell lung cancer.Methods:This study observed patients with metastatic non-driver mutation non-small cell lung cancer who were initially diagnosed at the First Affiliated Hospital of Dalian Medical University’s Department of Oncology from August 2021 to July 2022 and completed at least four cycles of chemotherapy combined with immune checkpoint inhibitor treatment.Lung biopsy tissues,fecal,and peripheral blood samples were collected from these patients.Based on the efficacy of the combined chemotherapy and immunotherapy,patients were divided into an effective group and an ineffective group.The tumor microbiota,gut microbiome,peripheral blood T-cell subsets,and inflammatory markers were compared between the two groups.The lung and fecal microbiota were analyzed using 16S rRNA high-throughput sequencing.Flow cytometry was used to detect T-cell subsets,and enzyme-linked immunosorbent assay was employed to measure inflammatory factors.Results:A total of 21 patients were observed.There were significant differences in the tumor microbiota between the responsive and non-responsive groups,particularly the proportion of the genera Sphingomonas and Pseudomonas.The gut microbiome composition changed after treatment,but there were no differences between the two groups before treatment.There were no significant differences in T-cell subsets and inflammatory markers between the responsive and non-responsive groups.Conclusion:The composition of intratumoral microbiota in non-small cell lung cancer patients may serve as an indicator of response to chemotherapy combined with immunotherapy.The predictive value of gut microbiota,T-cell subsets,and inflammatory markers appears limited.Future research should further validate the predictive role of changes in gut microbiota on treatment outcomes.

Finiteness Conditions and Relative Singularity Categories

We introduce the n-pure projective(resp.,injective)dimension of complexes in n-pure derived categories,and give some criteria for computing these dimensions in terms of the n-pure projective(resp.,injective)resolutions(resp.,coresolutions)and n-pure derived functors.As a consequence,we get some equivalent characterizations for the finiteness of n-pure global dimension of rings.Finally,we study Verdier quotient of bounded n-pure derived category modulo the bounded homotopy category of n-pure projective modules,which is called an n-pure singularity category since it can reflect the finiteness of n-pure global dimension of rings.

Controlled preparation and application of glutathione capped gold and platinum alloy nanoclusters with high peroxidase-like activity

In the present study,we have prepared glutathione capped gold and platinum alloy nanoclusters(AuPtNCs)in a controlled way by employing the hydrothermal method and optimized through adjusting the ratio of raw materials,reaction temperature,and time.Compared with the corresponding monometallic gold and platinum nanoclusters,the alloy nanoclusters’catalytic activity is improved dramatically in the oxidation of 3,3,5,5-tetramethylbenzidine(TMB)in the presence of H_(2)O_(2).And the maximum velocity is calculated to be 106×10^(-8)M·s^(-1) for TMB as substrate,being much better than that of other reported metallic nanoclusters and nanoparticles.Further study shows that the high catalytic activity mainly attributes to the synergistic effect of gold and platinum.Besides,they have been applied to determine H_(2)O_(2)in the presence of TMB,which shows high sensitivity with a limit of detection(LOD)at 100 n M.The proposed method has been used to determine H_(2)O_(2)in milk and contact lens solutions,which shows very good recovery and exhibits high practical application potential.Therefore,the present study provides a new type of alloy nanoclusters with high peroxidase-like activity,which will inspire more research interests on doping and alloying with Pt to improve the catalytic activity of metal nanoclusters.

Avramov–Martsinkovsky Type Exact Sequences with Tor Functors

For two classes of right R-modules W, X such that P W X, where P is the class of projective right R-modules, we show that there is an Avramov-Martsinkovsky type exact sequence with generalized Tate homology functor Tor^X,W, relative homology functors Tor^W and Tor^X. Many results in Iacob [Comm. Algebra, 35, 1589-1606 （2007）] and Liang [Algebr. Represent. Theory, 16, 1541-1560 （2013）] are generalized and improved.

Strength Activity Index of Air Quenched Basic Oxygen Furnace Steel Slag

Air quenched basic oxygen furnace steel slag （BOF-SS） is processed at very high cooling rate, which is expected to have different cementitious properties from conventional slowly cooled BOF-SS. For this purpose, the strength activity indexes of air quenched and slowly cooled BOF-SS are investigated. The results reveal that, under the specific surface area （S） of 490 m^2/kg, the compressive strength activity index reaches 1.24 after 28 days with replacement of 15% air quenched BOF-SS and reaches 1.05 after 28 days with replacement of 20% air quenched BOF-SS and 30%granulated blast furnace slag （GBFS）. The cementitious activity of air quenched BOF-SS is obviously higher than that of slowly cooled BOF-SS, mainly because it contains more C3 S and glassy phases.

Development trends of environmental protection technologies for Chinese steel industry

Chinese steel industry has made significant progress on reducing fresh water consumption and CO_2/SO_2/COD（ chemical oxygen demand） /dust emissions,and improving comprehensive utilization of solid waste. Some steel companies have become topped worldwide. However,due to the large output quantity of crude steel,the whole steel industry is still a huge source of pollutants. At present,environmental protection standards are encountering challenges of lack of technical support,taxation policy and other issues. Steel industry is currently facing enormous environmental pressure. The development trends of environmental protection technologies were studied by summarizing different development stages. To realize the development targets,the industry needs to carry out its research with independent intellectual property rights,develop comprehensive management systems and establish ecological chain with other industries. This can not only raise the level of environmental protection in a broader range,but also improve the energy efficiency of iron and steel plants and increase added value of waste utilization. Finally,23 environmental protection technologies were proposed and 4 new environmental protection practices were studied.

AGK2 pre-treatment protects against thioacetamide-induced acute liver failure via regulating the MFN2-PERK axis and ferroptosis signaling pathway

Background:Acute liver failure(ALF)is an unpredictable and life-threatening critical illness.The pathological characteristic of ALF is massive necrosis of hepatocytes and lots of inflammatory cells infiltration which may lead to multiple organ failure.Methods:Animals were divided into 3 groups,normal,thioacetamide(TAA,ALF model)and TAA+AGK2.Cultured L02 cells were divided into 5 groups,normal,TAA,TAA+mitofusin 2(MFN2)-siRNA,TAA+AGK2,and TAA+AGK2+MFN2-siRNA groups.The liver histology was evaluated with hematoxylin and eosin staining,inositol-requiring enzyme 1(IRE1),activating transcription factor 6β(ATF6β),protein kinase R(PKR)-like endoplasmic reticulum kinase(PERK)and phosphorylated-PERK(p-PERK).C/EBP homologous protein(CHOP),reactive oxygen species(ROS),MFN2 and glutathione peroxidase 4(GPX4)were measured with Western blotting,and cell viability and liver chemistry were also measured.Mitochondriaassociated endoplasmic reticulum membranes(MAMs)were measured by immunofluorescence.Results:The liver tissue in the ALF group had massive inflammatory cell infiltration and hepatocytes necrosis,which were reduced by AGK2 pre-treatment.In comparison to the normal group,apoptosis rate and levels of IRE1,ATF6β,p-PERK,CHOP,ROS and Fe2+in the TAA-induced ALF model group were significantly increased,which were decreased by AGK2 pre-treatment.The levels of MFN2 and GPX4 were decreased in TAA-induced mice compared with the normal group,which were enhanced by AGK2 pretreatment.Compared with the TAA-induced L02 cell,apoptosis rate and levels of IRE1,ATF6β,p-PERK,CHOP,ROS and Fe2+were further increased and levels of MFN2 and GPX4 were decreased in the MFN2-siRNA group.AGK2 pre-treatment decreased the apoptosis rate and levels of IRE1,ATF6β,p-PERK,CHOP,ROS and Fe2+and enhanced the protein expression of MFN2 and GPX4 in MFN2-siRNA treated L02 cell.Immunofluorescence observation showed that level of MAMs was promoted in the AGK2 pre-treatment group when compared with the TAA-induced group in both mice and L02 cells.Conclusions:The d

Metallacyclopentadienes are important metallacycles and regarded as intermediates in many reactions,therefore,new methods to achieve them are anticipated.In this study,a formal[3+2]method,through the reactions of an osmapentalyne with benzyl carbanions,was developed.The reactions underwent a nucleophilic attack of carbanions to the Os≡C bond,followed by C—H activation to form the five-membered osmacyclopentadiene ring.Most of the reactions were carried out at room temperature,the substituents on the aromatic rings of benzyl carbanions are diverse,and the resulting products contain an Os—H bond,representing a novel type of 10C-carbolong complexes.This work provides a new convenient route to construct metallacyclopentadienes,which is expected to further promote the development of such a type of substances.

The role of innate immunity in diabetic nephropathy and their therapeutic consequences

Diabetic nephropathy (DN) is an enduring condition that leads to inflammation and affects a substantial number of individuals with diabetes worldwide. A gradual reduction in glomerular filtration and emergence of proteins in the urine are typical aspects of DN, ultimately resulting in renal failure. Mounting evidence suggests that immunological and inflammatory factors are crucial for the development of DN. Therefore, the activation of innate immunity by resident renal and immune cells is critical for initiating and perpetuating inflammation. Toll-like receptors (TLRs) are an important group of receptors that identify patterns and activate immune responses and inflammation. Meanwhile, inflammatory responses in the liver, pancreatic islets, and kidneys involve inflammasomes and chemokines that generate pro-inflammatory cytokines. Moreover, the activation of the complement cascade can be triggered by glycated proteins. This review highlights recent findings elucidating how the innate immune system contributes to tissue fibrosis and organ dysfunction, ultimately leading to renal failure. This review also discusses innovative approaches that can be utilized to modulate the innate immune responses in DN for therapeutic purposes.

Gorenstein Projective Dimensions of Complexes

We show that over a right coherent left perfect ring R, a complex C of left R-modules is Gorenstein projective if and only if C^m is Gorenstein projective in R-Mod for all m E Z. Basing on this we show that if R is a right coherent left perfect ring then Gpd（C） = sup{Gpd（C^m）｜m ∈ Z} where Gpd（-） denotes Gorenstein projective dimension.
