Mechanisms and Efficacy of Chinese Herbal Medicines in Benign Prostatic Hyperplasia

Benign prostatic hyperplasia(BPH) is one of the most common diseases in elderly men, the incidence of which gradually increases with age and leads to lower urinary tract symptoms(LUTS), which seriously affects the quality of life of patients. Chinese herbal medicines(CHMs) are widely used for the treatment of BPH in China and some other countries. To explore the molecular mechanisms of CHMs for BPH, we conducted a review based on peer-reviewed English-language publications in Pub Med and Web of Science databases from inception to December 31, 2023. This article primarily reviewed 32 papers on the use of CHMs and its active compounds in the treatment of BPH, covering animal and cell experiments, and identified relevant mechanisms of action. The results suggest that the mechanisms of action of CHMs in treating BPH may involve the regulation of sex hormones,downregulation of cell growth factors, anti-inflammatory and antioxidative effects, inhibition of cell proliferation, and promotion of apoptosis. CHMs also exhibit α-blocker-like effects, with the potential to relax urethral smooth muscle and alleviate LUTS. Additionally, we also reviewed 4 clinical trials and meta-analyses of CHMs for the treatment of BPH patients, which provided initial evidence of the safety and effectiveness of CHMs treatment. CHMs treatment for BPH shows advantages as a multi-component, multi-target, and multi-pathway therapy, which can mitigate the severity of the disease, improve LUTS, and may become a reliable treatment option in the future.

BACKGROUND Gastric cancer(GC)poses a substantial risk to human health due to its high prevalence and mortality ***,current therapeutic strategies remain ***-cell RNA sequencing(scRNA-seq)offers the potential to provide comprehensive insights into GC *** To explore the distribution and dynamic changes of cell populations in the GC tumor microenvironment using scRNA-seq *** Cancerous tissues and paracancerous tissues were obtained from patients diagnosed with GC at various stages(I,II,III,and IV).Single-cell suspensions were prepared and analyzed using scRNA-seq to examine transcriptome profiles and cell-cell ***,quantitative real-time polymerase chain reaction(qRT-PCR)and flow cytometry were applied for measuring the expression of cluster of differentiation(CD)2,CD3D,CD3E,cytokeratin 19,cytokeratin 8,and epithelial cell adhesion *** Transcriptome data from 73645 single cells across eight tissues of four patients were categorized into 25 distinct cell clusters,representing 10 different cell *** were observed in these cell type *** adjacent epithelial cells in stages II and III exhibited a degenerative ***,the quantity of CD4 T cells and CD8 T cells were evidently elevated in cancerous *** analysis displayed a remarkable increase in interaction between B cells and other mast cells in stages II,III,and IV of *** findings were further validated through qRT-PCR and flow cytometry,demonstrating elevated T cells and declined epithelial cells within the cancerous *** This study provides a comprehensive analysis of cell dynamics across GC stages,highlighting key interactions within the tumor *** findings offer valuable insights for developing novel therapeutic strategies.

Gentiopicroside Alleviates Atherosclerosis by Suppressing Reactive Oxygen Species-Dependent NLRP3 Inflammasome Activation in Vascular Endothelial Cells via SIRT1/Nrf2 Pathway

Objective:To evaluate the protective effects of gentiopicroside(GPS)against reactive oxygen species(ROS)-induced NOD-like receptor family,pyrin domain containing 3(NLRP3)inflammasome activation in endothelial cells,aiming to reduce ***:Eight-week-old male ApoE-deficient mice were randomly divided into 2 groups(n=10 per group):the vehicle group and the GPS treatment *** groups were fed a high-fat diet for 16 ***(40 mg/kg per day)was administered by oral gavage to the GPS group,while the vehicle group received an equivalent volume of the vehicle *** the end of the treatment,blood and aortic tissues were collected for assessments of atherosclerosis,lipid profiles,oxidative stress,and molecular expressions related to NLRP3 inflammasome activation,ROS production,and ***,in vitro experiments on human aortic endothelial cells treated with oxidized low-density lipoprotein(ox-LDL)were conducted to evaluate the effects of GPS on NLRP3 inflammasome activation,pyroptosis,apoptosis,and ROS production,specifically examining the role of the sirtuin 1(SIRT1)/nuclear factor erythroid 2-related factor 2(Nrf2)***1 and Nrf2 inhibitors were used to confirm the pathway's ***:GPS treatment significantly reduced atherosclerotic lesions in the en face aorta(P<0.01),as well as in the thoracic and abdominal aortic regions,and markedly decreased sinus lesions within the aortic root(P<0.05 or P<0.01).Additionally,GPS reduced oxidative stress markers and proinflammatory cytokines,including interleukin(IL)-1βand IL-18,in lesion areas(P<0.05,P<0.01).In vitro,GPS inhibited ox-LDL-induced NLRP3 activation,as evidenced by reduced NLRP3(P<0.01),apoptosis-associated speck-like protein containing a CARD,cleaved-caspase-1,and cleavedgasdermin D expressions(all P<0.01).GPS also decreased ROS production,apoptosis,and pyroptosis,with the beneficial effects being significantly reversed by SIRT1 or Nrf2 ***:GPS exe

Prognostic impact of hypernatremia for septic shock patients in the intensive care unit

BACKGROUND Hypernatremia represents a significant electrolyte imbalance associated with numerous adverse outcomes,particularly in cases of intensive care unit(ICU)-acquired hypernatremia(IAH).Nevertheless,its relevance in patients with septic shock remains *** To identify independent risk factors and their predictive efficacy for IAH to improve outcomes in patients with septic *** In the present retrospective single-center study,a cohort of 157 septic shock patients with concurrent hypernatremia in the ICU at The First Affiliated Hospital of Soochow University,between August 1,2018,and May 31,2023,were *** were categorized based on the timing of hypernatremia occurrence into the IAH group(n=62),the non-IAH group(n=41),and the normonatremia group(n=54).RESULTS In the present study,there was a significant association between the high serum sodium concentrations,excessive persistent inflammation,immunosuppression and catabolism syndrome and chronic critical illness,while rapid recovery had an apparent association with ***,multivariable analyses revealed the following independent risk factors for IAH:Total urinary output over the preceding three days[odds ratio(OR)=1.09;95%CI:1.02–1.17;P=0.014],enteral nutrition(EN)sodium content of 500 mg(OR=2.93;95%CI:1.13–7.60;P=0.027),and EN sodium content of 670 mg(OR=6.19;95%CI:1.75–21.98;P=0.005)were positively correlated with the development of ***,the area under the curve for total urinary output over the preceding three days was 0.800(95%CI:0.678–0.922,P=0.001).Furthermore,maximum serum sodium levels,the duration of hypernatremia,and varying sodium correction rates were significantly associated with 28-day in-hospital mortality in septic shock patients(P<0.05).CONCLUSION The present findings illustrate that elevated serum sodium level was significantly associated with a poor prognosis in septic shock patients in the *** is highly recommended that hypernatremia be

截至2023年,谷歌的量子霸权芯片Sycamore尚不能用于密码攻击。量子计算在理论上对公钥密码有致命性威胁,但对对称密码影响甚微,目前业内已经从缩减版密码算法开始积极探索对称密码的量子攻击。SPN结构是对称密码算法中的一种代表性结构,目前基于真实量子计算机的各类量子算法均未能对该结构未经缩减的全规模密码算法进行攻击。基于量子退火算法独有的隧穿效应,使得该算法有利于科学问题的指数级解空间搜索,可将其视为一类具有全局寻优能力的人工智能算法。受传统密码分析方法的影响,本文提出一种对称密码攻击架构:量子退火耦合传统数学方法密码攻击的新型计算架构——QuCMC(Quantum Annealing-Classical Mixed Cryptanalysis)。基于该架构,首先利用可分性刻画SPN结构对称密码算法的线性层和非线性层传播规律,将区分器搜索问题转化为MILP模型求解问题。进一步将MILP模型转化为D-Wave CQM模型,在求解该模型的过程中利用量子波动产生的量子隧穿效应跳出传统智能算法极易陷入的局部亚优解,获得更优的解,即攻击目标对称密码算法的积分区分器。本文使用D-Wave Advantage量子计算机攻击了PRESENT、GIFT-64、RECTANGLE三种SPN结构代表算法,均成功搜索到最长9轮的积分区分器。并且实验结果表明,量子退火算法在跳出局部亚优解能力与降低求解时间两个方面,均优于经典启发式全局寻优算法模拟退火。本研究首次成功利用真实量子计算机对多种全规模SPN结构对称密码算法完成了攻击,攻击效果与传统数学方法持平。

卡夫卡《致某科学院的报告》的核心是现代人生命颓废、逃避现实的问题。红彼得打着科学进步的旗号走上了一条与柏拉图洞穴人脱困截然相反的道路。它所谓的成功不过是犹大获得的三十枚银币,其报告则是一份向人类出卖猿类共同体利益的投名状,既展现了个体堕落的平庸之恶,更可能带来猿类族群的灭顶之灾。红彼得从自然生存到家奴状态的“发迹”历程,隐喻了现代西方知识分子的精神颓废——在权力的打击与诱惑面前的退却与匍匐,而个体生命逃避现实、苟且地配合官僚制度文化,就会创造出适合于大屠杀主张构思、发展、实现所需要的特定环境,卡夫卡以此预警了20世纪的奥斯维辛集中营与大屠杀。

如何提高高大模板支撑体系施工的安全性和合理性是制约施工效率的关键问题,本研究以枣庄科教创新示范园建设项目看台工程为例,提出了基于数字孪生的分析方法。首先根据项目特点总结了高大模板支撑体系施工难点。为了解决施工安全、施工效率等问题,建立了基于数字孪生的高大模板支撑体系施工分析方法。该方法结合数字孪生的五维要素提出了施工分析的孪生架构。在此基础上,面向施工全过程形成工艺参数分析体系。针对本项目的关键施工工序,给出了关键技术控制要点。通过建立三维立体模型并分析构件受力性能,有效保证了施工的安全性和合理性,高效指导了施工全过程。
