The biologic antitumor necrosis factor alpha(anti-TNFα) agents have revolutionised the treatment of inflammatorybowel disease(IBD). However,some patients experience primary nonresponse,loss of response,or intolerance. Therefore,introducing a newer class of therapy with a mechanism of action that acts on different inflammatory pathways involved in IBD pathogenesis is appealing. Vedolizumab is a fully humanised monoclonal antibody that selectively targets α4β7 integrin. Based on the results of the pivotal clinical GEMINI trials,vedolizumab was approved for the treatment of adult patients with moderately to severely active ulcerative colitis(UC) and Crohn's disease(CD) refractory or intolerant to either conventional therapy or TNFα inhibitors. This review describes the efficacy,safety,and tolerability of vedolizumab reported in both randomized,controlled,clinical trials and from real-world experience in patients with UC and CD in order to identify its place in treatment algorithms for IBD.

The development of simple and high-throughput approaches to yield solid-state nanopores on large surface membranes may facilitate the prevalence of nanopore analysis technology and in-vitro diagnosis using portable ***,solidstate nanopores are typically realized by complex and highend nanofabrication ***,we present a method to achieve nanopores on polymer membranes using,silver nanoparticles(AgNPs)as templates and intense pulsed light(IPL)as a heating *** density and size of nanopores are controllable by adjusting the spin coating rate,the concentration of nanoparticle suspension,and the size of nanoparticles(NPs).The temperature of the AgNPs can rapidly reach 1132 K under instant heating of photothermal effect through light irradiation in 2 ms,resulting in localized melting and decomposition of an underneath polycarbonate(PC)membrane to yield nanopores with sizes ranging from 10 to 270 *** removing the nanoparticle residues,the flexible membrane with nanopores can be integrated into a flow cell to achieve a nanopore sensor that has been used to measure the translocation behaviors of bovine serum albumin(BSA).The results have demonstrated the capability of the sensor in protein denaturation *** low-cost and highthroughput technique to fabricate solid-state nanopores on flexible polymeric membranes may facilitate the development of more nanopore-based flexible sensors that can be integrated with other flexible components for wearable diagnosis.

Granite-gneiss rock outcrop inselbergs are ancient stable ecosystems with old,climaticallybuffered infertile landscapes(OCBILs).Although inselbergs provide key ecosystem services,little is done for their conservation and,so far,a lot of their unknown evolutionary history has already been lost by human *** a fine-scale approach,here we tested if habitat and environmental filtering(the inselberg’s harshness)affect the evolutionary diversity of an Atlantic Forest inselberg in *** recorded all trees with a diameter at breast height≥5cm in 20 plots in four habitat types(total sampled area of 0.8 hectares),from highest to lowest:island,hillside,foothill,and semideciduous forest(matrix).We also collected soil samples for chemical,textural and physical soil *** fitted linear models to test the effects of soil and habitat on plotlevel metrics of phylogenetic diversity and structure,lineage diversity,phylogeneticβ-diversity,and evolutionary *** found that the upper inselberg habitats contain a distinct set of ancient,closely related,harsh-tolerant lineages,as well as a subset of lineages that persist under harsh conditions with a certain degree of water *** inferior inselberg habitats harbor higher lineage diversity than expected by *** strongly predicted evolutionary *** concluded that soil depth,slope,nutrients and texture(environmental filtering)and habitat types and topography(habitat filtering)shape the evolutionary history contained in fine-scale inselberg habitats,which should encourage the conservation of these ancient ecosystems.

Objective:To investigate whether acupotomy could inhibit subchondral bone remodeling in knee osteoarthritis(KOA)rabbits by regulating the activity of osteoblasts and ***:KOA rabbits were prepared by immobilization for 6 and 9 weeks by Videman *** groups of rabbits(control,6 weeks and 9 weeks model,6 weeks and 9 weeks acupotomy,6 weeks and 9 weeks electroacupuncture,and 6 weeks and 9 weeks drug groups)received acupotomy,electroacupuncture and risedronate sodium intervention,respectively,for 3 ***:Acupotomy can inhibit the activity of osteoclasts and osteoblasts in subchondral bone by reducing the proteins expression of cathepsin K(CK)and tartrate-resistant acid phosphatase(TRAP)and decreasing the proteins expression of osteocalcin(OCN)and alkaline phosphatase(ALP),to intercept the abnormal bone resorption and bone formation of subchondral bone in 6-week and 9-week immobilization-induced KOA ***:These findings indicated that acupotomy may be more advantageous than risedronate sodium intervention in modulating subchondral bone remodeling in KOA rabbits,especially in 9-week immobilization-induced KOA rabbits.

To prevent, detect, and protect against forest fires, forest personnel need to define rules for determining forest fire risk. In Portugal, all municipalities must annually produce forest fire risk （FFR） maps. To produce more reliable FFR maps more easily, we developed an open source model using the Modeler plugin of SEXTANTE in the program QGIS version 2.0 Dufour. The model provides all the maps involved in the FFR model （susceptibility map, hazard map, vulnerability map, economic value map, and potential loss map） and was produced according to Portuguese Forest Authority＇s （AFN, Autoridade Florestal Nacional） rules for determining the FFR. This model was tested for the Portuguese municipality Santa Maria da Feira, where 40 % of the total municipality area falls in the category ＂very high＂ or ＂high＂ fire risk. The ＂very high＂ fire risk area is mainly classified as broad-leaved forest and has the steepest slopes （〉15 %）. The distance of burned areas to roads was also analyzed; the proportion of burned areas increased with increasing distance to the main roads. In addition, 92.6 % of the ＂high＂ and ＂very high＂ risk zones were located in areas with lower elevation. These results confirmed that forest fire is strongly influenced not only by environmental factors but also by anthropogenic factors. The procedure implemented here was compared with our open source application already available in QGIS and also to the same procedure implemented in GIS pro- prietary software. Although the results were obviously the same, the model developed here presents several advan- tages over the other two approaches. Besides being faster, it is easy to change the model parameters according to user needs （i.e., to the rules of different countries）, and can be modified and adapted to other variables and other areas to create risk maps for different natural phenomena （e.g., floods, earthquakes, landslides）. Nucleic acid extraction(NAE)is an essential component of many assays used for diagnostic and research *** a pandemic,performing NAE is associated with significant challenges due to material shortages,high costs,limited laboratory facilities,and the time *** delays patient diagnosis and treatment,particularly in countries that do not produce NAE ***,we tested several procedures to speed up the extraction process using common laboratory reagents,finding that heat-based nucleic acid extraction of severe acute respiratory syndrome coronavirus 2(SARS-COV2)is a detection method that can be easily implemented in laboratories,even those without centrifugation facilities.

In this article we improve a goodness-of-fit test, of the Kolmogorov-Smirnov type, for equally distributed- but not stationary-strongly dependent data. The test is based on the asymptotic behavior of the empirical process, which is much more complex than in the classical case. Applications to simulated data and discussion of the obtained results are provided. This is, to the best of our knowledge, the first result providing a general goodness of fit test for non-weakly dependent data.

In this paper, we provide a method based on quantiles to estimate the parameters of a finite mixture of Fréchet distributions, for a large sample of strongly dependent data. This is a situation that appears when dealing with environmental data and there was a real need of such method. We validate our approach by means of estimation and goodness-of-fit testing over simulated data, showing an accurate performance.
