Silk as a natural biomaterial is considered as a promising bone substitute in tissue *** and fibroin are the main components of silk and display unique features for their programmable mechanical properties,biocompatibility,biodegradability and morphological *** has been reported that sericin recombinant growth factors(GFs)can support cell proliferation and induce stem cell differentiation through cross-talk of signaling pathways during tissue *** transgenic technology allows the productions of bioactive heterologous GFs as fusion proteins with sericin,which are then fabricated into solid matrix or hydrogel ***,using an injectable hydrogel derived from transgenic platelet-derived GF(PDGF)-BB silk sericin,we demonstrated that the PDGF-BB sericin hydrogel effectively augmented osteogenesis induced by bone morphogenetic protein(BMP9)-stimulated mesenchymal stem cells(MSCs)in vivo and in vitro,while inhibiting adipogenic *** gene expression and protein-protein interactions studies demonstrated that BMP9 and PDGF-BB synergistically induced osteogenic differentiation through the cross-talk between Smad and Stat3 pathways in ***,our results provide a novel strategy to encapsulate osteogenic factors and osteoblastic progenitors in transgenic sericin-based hydrogel for robust bone tissue engineering.

Experimental animal model is an essential tool in the study of lipid metabolism disorders and *** the first use of rabbits fed a high-fat diet by Russian researchers in the last century,a variety of animal models have been used to study ***,engineered mice arethe most common animal ***,either in the basic study to elucidate the mechanisms,orin the clinical bridging study to develop new diagnostic and therapeutic methods for atherosclerosis,there are still many challenges:Which animal model should be selected?And how to interpret the results obtained from the selected animal model?Notably,human atherosclerosis is a disease that develops over time through a complex interaction of genetic and environmental factors,finally leading to complications such as myocardial infarction and ***,many results of atherosclerosis studies cannot be applied to humans,due to the current experimental animal models cannot mimic the complex lesions and complications in *** this review,we would like to discuss the current status and future challenges of experimental animal models for the study of lipid metabolism disorders and atherosclerosis.

OBJECTIVE Alzheimer disease(AD),the most common cause of dementia among older people,could not be prevented,halted,or reversed up till now.A large body of pharmacological study has revealed that Liuwei Dihuang(LW) possesses potential therapeutic effects on ***-AFC is key fractions *** the present study,we investigated the effect of LW-AFC on AD mouse *** PrP-hAβPPswe/PS1ΔE9(APP/PS1) mice and senescence-accelerated mouse prone 8 strain(SAMP8),classic AD animal models,were *** the treatment of LW-AFC,mice were cognitively evaluated in behavioral *** loss,amyloid-β(Αβ)deposition,and Αβ level were analyzed using Nissl staining,immunofluorescence,and an AlphaLISA assay,*** bead analysis,a radioimmunoassay,immunochemiluminometry,and an ELISA were used to measure cytokine and hormone *** subsets were detected using fl ow *** LW-AFC ameliorated the cognitive impairment observed in APP/PS1 and SAMP8 mice,including the impairment of object recognition memory,spatial learning and memory,and active and passive *** addition,LW-AFC alleviated the neuron loss in the hippocampus,suppressed amyloid-β(Αβ) deposition in the brain,and reduced the concentration of Aβ1-42 in the *** and plasma of APP/PS1 ***-AFC treatment also significantly restored the imbalance of hypothalamic-pituitary-adrenal(HPA) and hypothalamic-pituitary-gonadal(HPG) axis,enhanced the proliferation of splenocytes,corrected the disorder of lymphocyte subsets,and regulated the abnormal production of cytokine in APP/PS1 and SAMP8 *** of LW-AFC on pharmacodynamics and neuroendocrine immunomodulation network in APP/PS1 and SAMP8 mice were better than *** and *** LW-AFC ameliorated the behavioral and pathological deterioration of AD mouse models via the restoration of the NIM network,which supports the use of LW-AFC as a potential agent for AD therapy.

OBJECTIVE To investigate the machenism that isorhapontigenin(ISO)induced autophagy and cell growth inhibition in human bladder cancer cells on the basis of the existing research results,for providing a new mechanistic insight into understanding the anti-cancer properties of *** Western blotting analysis was used to detect the expression of *** Jet transfection technology was used to establish transfected cell lines,and indirect immunofluorescence staining was applied to observe the punctuate dots of ***-independent growth assaywas used to detect cell colony ***-PCR was used to detect the m RNA levelof SESN2 after ISO *** chromatin immunoprecipitation(CHIP)was used to detect the binding of transcription factor c-jun and specificity SESN2 promoter *** Our studies revealed that ISO-mediated autophagy induction occurred in a Sestrin2(SESN2)-dependent and Beclin 1(BECN1)-independent ***,we identified that ISO treatment induced SESN2 expression via a c-jun N-terminal kinase(JNK)/c-jun-dependent mechanism,in which ISO triggered JNK1-dependent c-jun activation and facilitated the binding of c-jun to consensus AP-1 binding site in the SESN2 promoter region,thereby led to a significant transcriptional induction of ***,we found that SESN2 expression was dramatically down-regulated or even lost in human bladder cancer tissues as compared to their paired adjacent normal tissues,while ISO treatment was capable of elevating SESN2 expression effectively and inhibiting bladder cancer formation in BBN-induced mouse bladder tumors in *** ISO treatment is able to induce autophagy and inhibit bladder cancer growth through JNK1/c-jun dependent transcriptional induction of SESN2,which provides a novel mechanistic insight into understanding the inhibitory effect of ISO on bladder cancers and suggests that ISO might act as a promising preventive and/or therapeutic drug ag
