With our continuous endeavors in seeking potent anti-HIV-1 agents,we reported here the discovery,biological characterization,and druggability evaluation of a class of nonnucleoside reverse transcriptase *** fully explore the chemical space of the NNRTI-binding pocket,novel series of dihydrothiopyrano[3,2-d]pyrimidines were developed by employing the structure-based design *** of the derivatives were endowed with prominent antiviral activities against HIV-1 wild-type and resistant strains at nanomolar *** them,compound 23h featuring the aminopiperidine moiety was identified as the most potent inhibitor,with EC50values ranging from 3.43 to 21.4 nmol/***,for the challenging double-mutants F227L+V106A and K103N+Y181C,23h exhibited 2.3-to 14.5-fold more potent activity than the first-line drugs efavirenz and ***,the resistance profiles of 23h achieved remarkable improvement compared to efavirenz and *** binding target of 23h was further confirmed to be HIV-1 reverse *** modeling studies were also performed to elucidate the biological evaluation results and give guidance for the optimization ***,no apparent inhibition of the major CYP450 enzymes and hERG channel was observed for *** importantly,23h was characterized by good pharmacokinetic properties and excellent safety in ***,23h holds great promise as a potential candidate for its effective antiviral efficacy and favorable drug-like profiles.

HIV-1 reverse transcriptase(RT)has received great attention as an attractive therapeutic target for acquired immune deficiency syndrome(AIdS),but the inevitable drug resistance and side effects have always been major challenges faced by non-nucleoside reverse transcriptase inhibitors(NNRTIs).This work aimed to identify novel chemotypes of anti-HIV-1 agents with improved drugresistance profiles,reduced toxicity,and excellent druggability.A series of diarylpyrimidine(dAPY)derivatives were prepared via structural modifications of the leads K-5a2 and *** them,15a with dimethylphosphine oxide moiety showed the most prominent antiviral potency against all of the tested viral panel,being 1.6-fold(WT,EC_(50) Z 1.75 nmol/L),3.0-fold(L100I,EC_(50) Z 2.84 nmol/L),2.4-fold(K103N,EC_(50) Z 1.27 nmol/L),3.3-fold(Y181C,EC50 Z 5.38 nmol/L),2.9-fold(Y188L,EC_(50) Z 7.96 nmol/L),2.5-fold(E138K,EC_(50) Z 4.28 nmol/L),4.8-fold(F227L/V106A,EC_(50) Z 3.76 nmol/L)and 5.3-fold(RES056,EC_(50) Z 15.8 nmol/L)more effective than that of the marketed drug *** docking results illustrated the detailed interactions formed by compound 15a and WT,F227L/V106A,and RES056 ***,15a-HCl carried outstanding pharmacokinetic(t1/2 Z 1.32 h,F Z 40.8%)and safety profiles(Ld_(50)>2000 mg/kg),which demonstrated that 15a HCl is a potential anti-HIV-1 drug candidate.

Inspired by our previous studies to discover novel human immunodeficiency virus-1(HIV-1)nonnucleoside reverse transcriptase inhibitors(NNRTIs)by targeting the tolerant region II of the NNRTIs binding pocket(NNIBP),a series of novel benzo[4,5]thieno[2,3-d]pyrimidine derivatives were designed through structure-based drug design as novel potent HIV-1 *** results showed that compound16b was the most active inhibitor,exhibiting 50% effective concentration(EC50)values from 0.021μmol/L to 0.298μmol/L against wild-type(WT)and a panel of NNRTIs-resistant HIV-1 ***,16b was demonstrated with a significantly low 50% cytotoxicity concentration(CC_(50))value(>200μmol/L)and high selectivity index(SI)*** addition,16b yielded moderate reverse transcriptase(RT)enzyme inhibition with a 50% inhibition concentration(IC_(50))value of 0.183μmol/L,which demonstrated that it acted as HIV-1 *** binding mode of 16b with RT was also illustrated via molecular ***,this work provided a novel lead compound for developing potent HIV-1 NNRTIs.

The molecular engineering of fluorescent organic/polymeric materials, specifically those emitting in the deep red to near-infrared spectrum, is vital for advancements in optoelectronics and biomedicine. Perylene diimide(PdI), a well-known fluorescent scaffold, offers high thermal and photophysical stability but suffers from fluorescence quenching in solid or aggregate states due to intense π-π interactions. To mitigate this, simple and versatile methods for strong PdI aggregate emission without extensive synthetic demands are highly desirable but still lacking. Here, we report a straightforward strategy to enhance the solid-state emission of PdI by introducing certain degree of through-space charge transfer(TSCT) via controlled radical polymerization,which can efficiently distort the typical face-to-face PdI stacking, enabling greatly enhanced deep red emission. This is achieved by growing electron-donating star-shape styrenic(co)polymers from a multidirectional electron-accepting PdI initiator. The incorporation of polycyclic aromatic monomers further shifted the emission into the near-infrared region, albeit with a reduced intensity. Overall, the emission of the PdI-based TSCT polymers can be systematically manipulated by leveraging the balance between PdI stacking and the TSCT degree, as confirmed by both experimental study and theoretical calculations. Our approach circumvents complex synthetic procedures, offering highly emissive materials with large Stokes shifts and showing broad potential for optoelectronic technology.

Alzheimer dementia （Ad） and type 2 diabetes （T2d） are interrelated global public health problems， and the current epidemics of both Ad and T2d are insulin resistance diseases. Thus， Ad and T2d may share common risk factors such as an unhealthy diet， lifestyle， and obesity. Meat products is an important part of the diet of consumers worldwide. This systematic review and meta-analysis aims to assess and estimate the effect of meat products consumption on Ad and T2d in humans. Web of Science， MEdLINE， PubMed， Cochrane Library， and Embase were searched from January 2012 to April 2024. 29 articles reported 32 cohort studies with 1，785，769 subjects， with 3，546 Ad cases and 91，092 T2d cases that met the inclusion criteria and were included in our analysis. Consumption of various meat products increased the risk of T2d (HR = 1.19， 95% CI: 1.13 - 1.26， p = 0.000; I2= 88.5%)， consumption of smoked， grilled/roasted and fried meat products was more likely to induce T2d (HR = 1.24， 95% CI: 1.18 - 1.30， p = 0. 000; I2= 76.1%)， but was borderline significant for the risk of Ad (HR = 1.11， 95% CI: 0.98 - 1.25， p = 0.094; I2= 58.8%)， with consumption of mainly livestock and poultry products increasing the risk (HR = 1.21， 95% CI: 1.03 - 1.42， p = 0.017; I2= 66.8%). The association between meat products consumption and Ad risk was influenced by meat type and sample size， while the risk of T2d was influenced by meat type， follow-up and sex. A daily intake of 27 g， 123 g and 170 g of livestock products increased the risk of T2d by 10%， 51% and 70% respectively， whereas the risk of T2d was reduced when the intake of various meat products was less than 23 g/d.

The objective of this work was to carry out a morphological characterization of arbuscular mycorrhizal fungi in the rhizosphere of Xanthosoma sagittifolium L. Schott plants. The plant material used was the white and red cultivars of X. sagittifolium, belonging to age intervals of 3 - 6, 6 - 9, and 9 - 12 months. Three harvest sites were chosen in the Central Region of Cameroon. In each site, soil from the rhizosphere and plant roots was collected in a randomized manner. In the field, the agronomic parameters were evaluated. The physicochemical characteristics of the soils, the mycorrhization index, and the morphological characterization of the mycorrhizal types of each site were carried out. The results obtained show that the agronomic growth parameters varied significantly using the Student Newman and Keuls Test depending on the harvest sites. The soils’ pH in all sites was acidic and ranged between 4.6 and 5.8. The Nkometou site has a loamy texture while the Olembe and Soa sites have loam-clay-sandy and loam-clay textures respectively. The highest mycorrhization frequencies appeared at the Nkometou site, with 75 and 87.33% of the white and red cultivars plant roots at 6 - 9 and 3 - 6 months. The relative abundance of AMF arbuscular mycorrhizal fungal spores in the rhizosphere of X. sagittifolium plants varied with age and cultivar. There were 673 spores between 9 - 12 months in Nkometou in the red cultivar. Six AMF genera were identified in all the different soils collected: Acaulospora sp., Funneliformis sp., Gigaspora sp., Glomus sp., Scutellospora sp., and Septoglomus sp. The genus Glomus sp. was the most present at all age intervals in both cultivars.

Occurrence of Extreme Rainfall and Flood Risks in Yopougon, Abidjan, Southeast Côte d’Ivoire from 1971 to 2022

Yopougon, located in the western part of the Autonomous district of Abidjan, is the most heavily populated municipality in Côte d’Ivoire. However, this area is prone to floods and landslides during the rainy season. The study aims to assess recent flood risks in the municipality of Yopougon of the Autonomous district of Abidjan. To achieve this objective, the study analyzed two types of data: daily rainfall from 1971 to 2022 and parameters derived from a Numerical Field and Altitude Model (NFAM). The study examined six rainfall parameters using statistical analysis and combined land use maps obtained from the NFAM of Yopougon. The results indicated that, in 67% of cases, extreme rainfall occurred mainly between week 3 of May and week 1 of July. The peak of extreme rainfall was observed in week 2 of June with 15% of cases. These are critical periods of flood risks in the Autonomous district of Abidjan, especially in Yopougon. In addition, there was variability of rainfall parameters in the Autonomous district of Abidjan. This was characterized by a drop of annual and seasonal rainfall, and an increase of numbers of rainy days. Flood risks in Yopougon are, therefore, due to the regular occurrence of rainy events. Recent floods in Yopougon were caused by normal rains ranging from 55 millimeters (mm) to 153 mm with a return period of less than five years. Abnormal heavy rains of a case study on June 20-21, 2022 in Yopougon were detected by outputs global climate models. Areas of very high risk of flood covered 18% of Yopougon, while 31% were at high risk. Climate information from this study can assist authorities to take in advance adaptation and management measures.
