The paper presents a short overview of the contemporary approaches for calculating welding distortions. In order to meet the existing challenges, an advanced hybrid model for prediction of welding distortions in large structures is described. For the purpose of illustrating the capability of this model, a simulation case is put into discussion. The results are validated by comparison with experimental data, as well as with common simulation technique. Analysis of the calculation costs is also presented. The directions for development of calculation technique, based on the presented model, are also suggested.

BACKGROUND Endoscopic en bloc resection of larger polyps is relevant because risk of advanced neoplasia or malignancy correlates with tumor size. Recurrence rates after piecemeal endoscopic mucosal resection (EMR) are high and endoscopic submucosal dissection (ESD) is associated with higher complication rates in the western world. AIM To develop a modified endoscopic en bloc resection technique using an external additional working channel and novel agent for submucosal injection. METHODS EMR+ was considered as modified grasp and snare technique. For simultaneous use of a grasping and cutting device a novel additional working channel was used (AWC■, Ovesco Endoscopy, Tübingen, Germany). AWC■ is installed on the outer surface of the endoscope, covered with a plastic sleeve and designed for single use. For submucosal injection a new agent consisting of poloxamers was used (LiftUp■, Ovesco Endoscopy, Tübingen, Germany). The agent is liquid at room temperature and forms a stable and permanent gel cushion after injection. Safety of LiftUp? has been shown in a pre-clinical study in domestic pigs. LiftUp■ is commercially not yet available but approval is expected in early 2019. EMR+ was first developed ex vivo (explanted pig stomach) and subsequently evaluated in vivo (stomach, porcine model, 3 domestic pigs). Main outcome measurements were: Procedure time, macroscopic en bloc resection and adverse events. RESULTS Concept of EMR+ was first developed ex vivo (explanted pig stomach). Ex vivo, 22 resections were performed after technique was established. Median procedure time (measured from begin of injection to extraction of resection specimen) was 7 min (range 5-11, SD 1.68) and median size of resection specimens was 30 mm × 26 mm × 11 mm ex vivo. Subsequently 13 resections were performed in vivo (stomach, porcine model, 3 domestic pigs). In this contribution, we present iHEARu-PLAY, an online, multi-player platform for crowdsourced database collection and labelling, including the voice analysis application (VoiLA), a free web-based speech classification tool designed to educate iHEARu-PLAY users about state-of-the-art speech analysis paradigms. Via this associated speech analysis web interface, in addition, VoiLA encourages users to take an active role in improving the service by providing labelled speech data. The platform allows users to record and upload voice samples directly from their browser, which are then analysed in a state-of-the-art classification pipeline. A set of pre-trained models targeting a range of speaker states and traits such as gender, valence, arousal, dominance, and 24 different discrete emotions is employed. The analysis results are visualised in a way that they are easily interpretable by laymen, giving users unique insights into how their voice sounds. We assess the effectiveness of iHEARu-PLAY and its integrated VoiLA feature via a series of user evaluations which indicate that it is fun and easy to use, and that it provides accurate and informative results.

AIM:To search for further immunodominant peptides of the pyruvate dehydrogenase complex E2-component (PDC-E2) recognized by antimitochondrial antibodies (AMA) in primary biliary cirrhosis (PBC). METHODS:Sera from 95 patients with PBC were tested by enzyme-linked immunosorbent assay against 33 synthetic overlapping peptides (25 amino acids; aa) covering the entire length of the E2-subunit of PDC-E2. Furthermore,the inner lipoyl peptide 167-184 was used in an unlip oylated and a lipoylated form as well as coupled to ovalbumin. Sera from 11 AMA negative/ANA posit ive PBC patients,63 patients with other liver disorders and 22 healthy blood donors served as ***:Of the 95 PBC-sera,74% reacted with the peptide 475-499 and 58% with the pept ide 407-431 located within the catalytic domain of PDC-E2. Patients with other disorders or healthy controls were positive in only up to 18%. Antibodies to the unlipoylatedand lip oylated pept ide 167-184 within the inner lipoyl domain were found in only 5% and 11% of the PBC sera,respectively; using ovalbumin-coupled peptides,the incidence increased up to 57% (unlipoylated form). CONCLUSION:Peptides within the catalytic site of PDC-E2 rather than the previously reported lipoyl binding peptide 167-184 may represent major immunodomin ant epitopes recognized by AMA in PBC.

Long-term effects of environmentally relevant concentrations of silver nanoparticles on microbial biomass, enzyme activity,and functional genes involved in the nitrogen cycle of loamy soil

The increasing production and use of engineered silver nanoparticles（AgNP） in industry and private households are leading to increased concentrations of AgNP in the environment. An ecological risk assessment of AgNP is needed, but it requires understanding the long term effects of environmentally relevant concentrations of AgNP on the soil microbiome. Hence, the aim of this study was to reveal the long-term effects of AgNP on soil microorganisms. The study was conducted as a laboratory incubation experiment over a period of one year using a loamy soil and AgNP concentrations ranging from 0.01 to 1 mg AgNP/kg soil. The short term effects of AgNP were, in general, ***, after one year of exposure to 0.01 mg AgNP/kg, there were significant negative effects on soil microbial biomass（quantified by extractable DNA; p = 0.000） and bacterial ammonia oxidizers（quantified by amo A gene copy numbers; p = 0.009）. Furthermore, the tested AgNP concentrations significantly decreased the soil microbial biomass, the leucine aminopeptidase activity（quantified by substrate turnover; p = 0.014）, and the abundance of nitrogen fixing microorganisms（quantified by nif H gene copy numbers; p = 0.001）. The results of the positive control with Ag NO3 revealed predominantly stronger effects due to Ag＋ion release. Thus, the increasing toxicity of AgNP during the test period may reflect the long-term release of Ag^＋ions. Nevertheless, even very low concentrations of AgNP caused disadvantages for the microbial soil community, especially for nitrogen cycling, and our results confirmed the risks of releasing AgNP into the environment.

We used thermal images of bamboo culms of Guadua angustifolia Kunth to analyze the relationship between culm surface temperature and maturity, driven by the hypothesis that young culms may exhibit lower surface temperatures than old ones. The culm surface temperature shows small but constant differences between three age classes of 1, 2, and 3 years. Our findings indicate that surface temperature may be applied as an additional indi- cator to support the determination of maturity of guadua culms besides the visual assessment of the culms.
