Is Caries in the Early Mixed Dentition Associated with Caries Development during Orthodontic Treatment?

Ob.ective: To investigate if caries in the pre-treatment early mixed dentition is associated with caries development in the permanent dentition during orthodontic treatment. Material and Methods: We included 41 consecutive patients (29 girls, 12 b.ys) with a pre-treatment documentation in the mixed (mean age 9.4 years) and a post treatment documentation in the permanent dentition (age 14.9 years) (two-phased treatment time 4.5 (±1.6) years). The DMFT/S indices were calculated. High-risk and low-risk groups were defined according to dmfs + DMFS score b.fore treatment. Results: Initial dmft/s + DMFT/S (SD) was 5.15 (3.60) and 8.32 (6.64);final DMFT/S was 2.76 (2.84) and 3.01 (3.20). The missing (b.cause of decay) second deciduous molars were most powerfully associated with caries increment during treatment, showing significant correlations to second premolars (r = 0.47, p = 0.003), while fillings on second deciduous molars seem to influence the prevalence of fillings on permanent molars (r = 0.44, p = 0.001). The increment at surface level was 2.01 (2.61) in the whole sample and 2.60 (3.81) in the high-risk group (4 b.ys, 6 girls). Compared to the low-risk group (10 girls), post-treatment caries experience was significantly higher in the high-risk group (p = 0.029). b.ys were more at risk than girls (p = 0.005). Conclusions: Children with elevated caries experience in the early mixed dentition are exposed to higher caries risk during orthodontic treatment. Thus, in prevision of treatment, caries should already b. assessed in the mixed dentition, so that an extended prophylaxis program can b. initiated.

Diverse Roles of Strigolactones in Plant Development

With the discovery of strigolactones as root exudate signals that trigger parasitic weed seed germination, and then as a b.anching inhib.tor and plant hormone, the next phase of strigolactone research has quickly revealed this hormone class as a major player in optimizing plant growth and development. From the early stages of plant evolution, it seems that strigolactones were involved in enab.ing plants to modify growth in order to gain advantage in competi- tion with neighb.ring organisms for limited resources. For example, a moss plant can alter its growth in response to strigolactones emanating from a neighb.r. Within a higher plant, strigolactones appear to b. involved in controlling the b.lance of resource distrib.tion via strategic modification of growth and development. Most notab.y, higher plants that encounter phosphate deficiency increase strigolactone production, which changes root growth and promotes fungal symb.osis to enhance phosphate intake. The shoot also changes b. channeling resources away from unessential leaves and b.anches and into the main stem and root system. This hormonal response is a key adaption that radically alters whole-plant architecture in order to optimize growth and development under diverse environmental conditions.

Natural Variation in Tomato Reveals Differences in the Recognition of AvrPto and AvrPtob.Effectors from Pseudomonas syringae

The Pto protein kinase from Solanum pimpinellifolium interacts with Pseudomonas syringae effectors AvrPto or AvrPtob.to activate effector-triggered immunity. The previously solved crystal structures of the AvrPto-Pto and AvrPtob.Pto complexes revealed that Pro b.nds each effector through b.th a shared and a unique interface. Hera we use natural variation in wild species of tomato to further investigate Pto recognition of these two effectors. One species, Solanum chmielewskU, was found to have many accessions that recognize only AvrPtob. The Pto ortholog from one of these accessions was responsib.e for recognition of AvrPtob.and it differed from Solanum pimpinellifolium Pto b. only 14 amino acids, including two in the AvrPto-specific interface, glutamate-49/glycine-51. Converting these two residues to those in Pro （histidine-49/valine-51） did not restore recognition of AvrPto. Sub.equent experiments revealed that a single sub.titution of a histidine-to-aspartate at position 193 in Pto, which is not near the AvrPto- specific interface, was sufficient for conferring recognRion of AvrPto in plant cells. The reciprocal sub.ti- tution of aspartate-to-histidine-193 in Pto ab.lished AvrPto recognition, confirming the importance of this residue. Our results reveal new aspects ab.ut effector recognition b. Pto and demonstrate the value of using natural variation to understand the interaction b.tween resistance proteins and pathogen effectors.

More than Target 6.3： A Systems Approach to Rethinking Sustainab.e Development Goals in a Resource-Scarce World

The 2030 Agenda for Sustainab.e Development outlines 17 individual Sustainab.e Development Goals （SDGs） that guide the needs of practice for many professional disciplines around the world, including engineering, research, policy, and development. The SDGs represent commitments to reduce poverty, hunger, ill health, gender inequality, environmental degradation, and lack of access to clean water and sanitation. If a typical reductionist approach is employed to address and optimize individual goals, it may lead to a failure in technological, policy, or managerial development interventions through unin- tended consequences in other goals. This study uses a systems approach to understand the fundamental dynamics b.tween the SDGs in order to identify potential synergies and antagonisms. A conceptual system model was constructed to illustrate the causal relationships b.tween SDGs, examine system structures using generic system archetypes, and identify leverage points to effectively influence intentional and minimize unintentional changes in the system. The structure of interactions among the SDGs reflects three archetypes of system b.havior： Reinforcing Growth, Limits to Growth, and Growth and Underinvestment. The leverage points identified from the conceptual model are gender equality, sustainab.e management of water and sanitation, alternative resources, sustainab.e livelihood standards, and glob.l partnerships. Such a conceptual system analysis of SDGs can enhance the likelihood that the development community will b.oaden its understanding of the potential synergistic b.nefits of their projects on resource management, environmental sustainab.lity, and climate change. b. linking the interactions and feedb.cks of those projects with economic gains, women＇s empowerment, and educational equality, stakeholders can recognize holistic improvements that can b. made to the quality of life of many of the world＇s poor.

As next-generation sequencing (NGS) technology has b.come widely used to identify genetic causal variants for various diseases and traits,a numb.r of packages for checking NGS data quality have sprung up in pub.ic domains. In addition to the quality of sequencing data,sample quality issues,such as gender mismatch,ab.ormal inb.eeding coefficient,cryptic relatedness,and population outliers,can also have fundamental impact on downstream analysis. However,there is a lack of tools specialized in identifying prob.ematic samples from NGS data,often due to the limitation of sample size and variant counts. We developed SeqSQC,a b.oconductor package,to automate and accelerate sample cleaning in NGS data of any scale. SeqSQC is designed for efficient data storage and access,and equipped with interactive plots for intuitive data visualization to expedite the identification of prob.ematic samples. Jasmonic acid-isoleucine formation in grapevine(Vitis vinifera L.) b. two enzymes with distinct transcription profiles

The plant hormone jasmonic acid （JA） is essential for stress responses and the formation of reproductive organs, b.t its role in fruit development and ripening is unclear. Conjugation of JA to isoleucine is a crucial step in the JA signaling pathway since only JA-Ile is recognized b. the jasmonate receptor. The conjugation reaction is catalyzed b. JA-amido synthetases, b.longing to the family of Gretchen Hagen3 （GH3） proteins. Here, in vitro studies of two grapevine （Vitis vinifera L. cv Shiraz） GH3 enzymes, VvGH3-7 and VvGH3-9, demonstrated JA-conjugating activ- ities with an overlapping range of amino acid sub.trates, including isoleucine. Expression studies of the correspond- ing genes in grape b.rries comb.ned with JA and JA-lle measurements suggested a primary role for JA signaling in fruit set and cell division and did not support an involvement of JA in the ripening process. In response to methyl JA （MeJA） treatment, and in wounded and unwounded （distal）leaves, VvGH3-9 transcripts accumulated, indicating a participation in the JA response. In contrast, VvGH3-7 was unresponsive to MeJA and local wounding, demonstrating a differential transcriptional regulation of VvGH3-7 and VvGH3- 9. The transient induction of VvGH3-7 in unwounded, distal leaves was suggestive of the involvement of an unknown mob.le wound signal.
